    Intestinal cell migration damage induced by enteropathogenic Escherichia coli strains

    Epithelial cell migration is an essential response to enteric pathogens such as enteropathogenic Escherichia coli (EPEC). This study aimed to investigate the effects of EPEC infection on intestinal epithelial cell migration in vitro, as well as the involvement of type III secretion system (T3SS) and Rho GTPases. Crypt intestinal epithelial cells (IEC-6) were infected with EPEC strains (E2348/69, ΔescF, and the LDI001 strain isolated from a malnourished Brazilian child) and commensal E. coli HS. Wound migration and cell death assays were performed at different time-points. Transcription and expression of Rho GTPases were evaluated using real-time PCR and western blotting. Overall, EPEC E2348/69 reduced migration and increased apoptosis and necrosis levels compared to EPEC LDI001 and E. coli HS strains. Moreover, EPEC LDI001 impaired cell migration at a higher level than E. coli HS and increased necrosis after 24 hours compared to the control group. The different profiles of virulence genes between the two wild-type EPEC strains, characterized by the absence of espL and nleE genes in the LDI001, might explain the phenotypic results, playing significant roles on cell migration impairment and cell death-related events. Moreover, the type III secretion system is determinant for the inhibition of intestinal epithelial cell migration by EPEC 2348/69, as its deletion prevented the effect. Active Rac1 concentrations were increased in E2348/69 and LDI001-infected cells, while the T3SS-deficient strain did not demonstrate this activation. This study contributes with valuable insight to characterize the mechanisms involved in the impairment of intestinal cell migration induced by EPEC

    Matéria seca e altura das plantas de soja e arroz em função do grau de compactação e do teor de água de dois latossolos Height and dry matter of soybean and rice in function of degree of compaction and water content of two oxisols

    Este trabalho teve o objetivo de avaliar a altura de plantas e a produção de matéria seca da parte aérea e das raízes de soja e arroz em dois teores de água (água retida a 0,05 e a 0,01 MPa) e três de resistência do solo à penetração (entre 0,25 e 6,46 MPa), determinados com o penetrômetro de anel dinamométrico. Foram utilizadas amostras de Latossolo Vermelho, distrófico, textura média (LVd) e Latossolo Vermelho, eutroférrico, textura argilosa (LVef), coletadas na profundidade de 0,0-0,20 m e compactadas em camadas de 0,03 m, em vasos de 0,20 m de altura e 0,25 m de diâmetro. A altura das plantas foi reduzida para valores de resistência à penetração superiores a 3,0 MPa. A menor produção de matéria seca da parte aérea das plantas ocorreu na resistência à penetração de 3,76 e 3,37 MPa para soja; 3,93 e 3,37 MPa para arroz, no Latossolo Vermelho, textura média e Latossolo Vermelho argiloso, respectivamente, no teor de água retida na tensão de 0,05 MPa, e no maior teor de água somente ocorreu redução da produção de matéria seca das raízes.<br>This research was carried out to evaluate the height of plants and production of aerial and root dry matter of soybean and rice in two water contents (water content at tension 0.05 and 0.01 MPa) and three levels of resistance to penetration (between 0.25 and 6.46 MPa) determined with penetrometer dinamometric ring. Samples of Haplustox and Eutrustox, medium and clayey texture were collected in depth of 0.0-0.20 m and compact in layers of 0.03 m in pots of 0.20 m of height and 0.25 m of diameter. The height of plants was reduced in values of penetration resistance greater of 3.0 MPa. The smaller production of aerial part dry matter of the plants occurred in penetration resistance of 3.76 and 3.37 MPa for soybean; 3.93 and 3.37 MPa for rice, in Haplustox and Eutrustox, respectively, in the water content in tension of 0.05 MPa, and in the greater water content only occurred reduction of root dry matter

    o estudo investigou o tema da sexualidade junto a 14 jovens adultos com deficiência auditiva de 18 a 35 anos de idade, no Centro de Distúrbios da Audição Linguagem e Visão (CEDALVI) que faz parte do HRAC-USP/Bauru. Os objetivos foram: identificar quais as necessidades de informação no que diz respeito à sexualidade; intervir, através de um programa de orientação sexual, informando e discutindo sobre temas referentes à sexualidade e verificar a aquisição de informação obtida com o programa de orientação sexual. Para tal, foi realizado um programa de orientação sexual, com três encontros abordando os seguintes temas: Órgãos sexuais e relações de gênero, Relacionamento afetivo e Gravidez, doenças sexualmente transmissíveis e auto-estima. Utilizou-se para a coleta dos dados um questionário inicial, questionários com questões fechadas, falso e verdadeiro, tipo pré e pós-teste no início e ao final de cada encontro e uma entrevista de avaliação processual, ao final do programa. Observou-se que o termo sexualidade continua sendo reduzido ao ato sexual ou as formas de prevenção de doenças ou métodos contraceptivos por grande parte dos jovens e a fonte de informação mais citada foi a mídia. O tema de maior número de acertos no pós-teste foi Órgãos sexuais e relações de gênero e todos, em geral, avaliaram positivamente a participação no programa. Concluiu-se que é necessário que a sexualidade seja cada vez mais incluída em programas de reabilitação, garantindo o acesso a informação e a discussão do tema, tão importante na vida desses jovens.<br>the study investigated the theme of sexuality among 14 hearing impaired young adults, aged 18 to 35 years, at the Center for Hearing, Language and Visual Disturbances (CEDALVI), a service of the HRAC-USP/Bauru. The aims were: to identify what kinds of information on sexuality were needed; to intervene through a sexual orientation program, so as to inform and discuss themes related to sexuality and to assess the information obtained by means of the sexual education program. To this end, a sexual orientation program was set up with three sessions concerning the following themes: Sexual organs and gender relationships, Caring relationships and Pregnancy, sexually transmitted diseases and self-esteem. Data was collected through a first time questionnaire, with closed true and false questions, with pre and post-tests applied at the beginning and the end of each meeting, followed by interviews to evaluate the whole process at the end of the program. The results showed that most young adults still view the term sexuality as restricted to sexual intercourse or to prevention of sexually transmitted diseases or to contraceptive methods and the media was cited as the main source of information. In the post-test, the greatest number of correct responses occurred for the theme Sexual organs and gender relationships; in general, all participants offered positive reviews for participation in the program. The conclusion was that it is paramount to include sexual orientation in rehabilitation programs, guaranteeing access to information and promoting discussion of such subjects in these young people's lives
