    Role of the aryl hydrocarbon receptor in Sugen 5416-induced experimental pulmonary hypertension

    Rationale: Rats dosed with the vascular endothelial growth factor (VEGF) inhibitor Sugen 5416 (Su), placed in hypoxia then restored to normoxia has become a widely used model of pulmonary arterial hypertension (PAH). The mechanism by which Su exaccerbates pulmonary hypertension is, however, unclear. Objectives: We investigated Su-activation of the aryl hydrocarbon receptor (AhR) in patient human pulmonary arterial smooth muscle cells (hPASMCs) and patient blood outgrowth endothelial cells (BOECs). We also examined the effect of AhR on aromatase and estrogen levels in the lung. Methods, Measurements and Main Results: Protein and mRNA analysis demonstrated that CYP1A1 was very highly induced in the lungs of Su/hypoxic (Su/Hx) rats. The AhR antagonist CH223191 (8mg/kg/day) reversed the development of PAH in this model in vivo and normalized lung CYP1A1 expression. Increased lung aromatase and estrogen levels in Su/Hx rats were also normalized by CH223191 as was AhR nuclear translocator (ARNT [HIF-1β]) which is shared by HIF-1α and AhR. Su reduced HIF1α expression in hPASMCs. Su induced proliferation in BOECs and increased apoptosis in human pulmonary microvascular endothelial cells (hPMECs) and also induced translocation of AhR to the nucleus in hPASMCs. Under normoxic conditions, hPASMCs do not proliferate to Su. However when grown in hypoxia (1%) Su induced hPASMC proliferation. Conclusion: In combination with hypoxia, Su is proliferative in patient hPASMCs and patient BOECs and Su/Hx-induced PAH in rats may be facilitated by AhR-induced CYP1A1, ARNT and aromatase. Inhibition of the AhR receptor may be a novel approach to the treatment of pulmonary hypertension

    Tujuan penelitian mengenai struktur morfologi dan anatomi perkembangan Cypraea moneta adalah mempelajari perkembangan morfologi dan anatomi C. moneta jantan dan betina. Spesimen diukur panjang cangkang dan berat, kemudian dikelompokkan menurut panjang cangkang. Cangkang spesimen dicerna dengan menggunakan HCl 5%, kemudian dibedah dengan menggunakan micro dissection kit di bawah mikroskop stereo yang telah terhubung dengan monitor PC dan kamera digital. Organ yang diamati meliputi mantel dan papila, tentakel, mata, kakiperut, dan organ reproduksi. Hasilnya memperlihatkan bahwa tidak ada perbedaan antara warna mantel, tipe papila, kakiperut, mata, tentakel jantan dan tentakel betina. Perkembangan morfologi ditandai dengan pertambahan ukuran cangkang, berat, jumlah geligi pada cangkang, dan bentuk cangkang. Jantan berkembang testis dan kelenjar testis, betina dengan ovarium dan lubang genital. Warna testis dan ovarium berubah sesuai dengan perkembangan kematangannya.

    Ancient language Font Recognition is one of the Challenging tasks in Optical Character Recognition and Document Analysis. Most of the existing methods are for font recognition make use of local typographical features and connected component analysis. In this paper, Ancient language font recognition is done based on global texture analysis. Ancient language characters are different from currentnbsp centuryrsquos Ancient language character. This paper concentrates on the century identification of ancient language characters and converting them into current centuryrsquos form using MATLAB. Recognition of ancient language hand written characters from inscriptions is difficult. In this paper, a method for recognizing Ancient language characters from stone inscriptions, called the contour-let transform, which has been recently introduced, is adopted. From the previous research works, itrsquos noticed that Wavelet transforms are not capable of reconstructing curved images are perfectly. The contour-let transform offers a solution to remedy to this insufficiency. Contour-let transform is a 3D approach technique where as wavelet transform is a 2D technique. The characters from the input image are recognized through the clustering mechanism. Further the noise is present in the image is removed by fuzzy median filters. Neural networks are been employed to train the image and compare the data with the current centuryrsquos character. hence a more accurate recognition of Ancient language characters from stone inscriptions is obtained

    Malignant transformation of a recurrent giant cell tumor of bone with lung metastasis: a case report

    Giant cell tumors (GCT) are benign tumors with potential for aggressive behaviour and capacity to metastasize. It is a locally destructive tumor that occurs predominantly in long bones of adolescents and young adults in the epiphysis. Although rarely lethal, benign bone tumors may be associated with a substantial disturbance of the local bony architecture that can be particularly troublesome in peri-articular locations. It is characterized by a proliferation of mononuclear stromal cells and the presence of many multi-nucleated giant cells with homogenous distribution. There are varying surgical techniques ranging from intra-lesional curettage to wide resection. As most giant cell tumors are benign and are located near a joint in young adults, several authors favour an intralesional approach that preserves anatomy of bone. Although GCT is classified as a benign lesion, few patients develop progressive lung metastases with poor outcomes. Malignant transformation without radiotherapy exposure, is an uncommon event, occurring in less than 1% of giant cell tumors of bone. Here we reported a case of recurrent GCT of tibia that at the time of final recurrence was found to have undergone malignant transformation over a period of 6 years following several limb salvaging procedures. Concurrent metastases were found in the lung, but these were non-transformed GCT following which the patient has undergone above knee amputation

    NeuroWrite: Predictive Handwritten Digit Classification using Deep Neural Networks

    The rapid evolution of deep neural networks has revolutionized the field of machine learning, enabling remarkable advancements in various domains. In this article, we introduce NeuroWrite, a unique method for predicting the categorization of handwritten digits using deep neural networks. Our model exhibits outstanding accuracy in identifying and categorising handwritten digits by utilising the strength of convolutional neural networks (CNNs) and recurrent neural networks (RNNs).In this article, we give a thorough examination of the data preparation methods, network design, and training methods used in NeuroWrite. By implementing state-of-the-art techniques, we showcase how NeuroWrite can achieve high classification accuracy and robust generalization on handwritten digit datasets, such as MNIST. Furthermore, we explore the model's potential for real-world applications, including digit recognition in digitized documents, signature verification, and automated postal code recognition. NeuroWrite is a useful tool for computer vision and pattern recognition because of its performance and adaptability.The architecture, training procedure, and evaluation metrics of NeuroWrite are covered in detail in this study, illustrating how it can improve a number of applications that call for handwritten digit classification.     A cross sectional study on the prescribing pattern, self medication and adverse reactions associated with topical corticosteroids

    Background: Glucocorticosteroids, one of the common drugs used by the dermatologists brought a revolutionary change in their practice. Unfortunately steroids are misused in dermatological panacea due to dramatic relief in inflammatory and pruritic skin conditions but may lead to deleterious effects if irrationally used. Hence this study was planned to know the prescribing pattern of corticosteroids.Methods: It was a cross sectional observational study. The prescriptions of patient attending the dermatology OPD were screened for the usage of the corticosteroids. The demographic data, chief complaints, diagnosis and the details of the drugs was collected.Results: Out of 384 prescriptions screened, 14.06% were prescribed corticosteroids. Average number of drugs per prescription was 2.28±0.83. Polypharmacy (≥4 drugs) observed in 9.26% of prescriptions. Corticosteroids prescribed by generic name were 24.07% and brand name was 75.93%. Corticosteroids alone prescribed in 38.89% and along with antihistaminics/antibiotics/emollients in 61.11%. Topical corticosteroids prescribed in 79.6% and 20.4% by systemic route. Moderately potent steroids prescribed in 74.42% followed by potent (13.95%) and very potent steroids (11.63%). Patients on corticosteroid self-medication were 28%, among them 4 developed adverse reactions with severe acneiform eruptions over the face. No fixed dose combination drugs were prescribed.Conclusions: In this study we observed the rationale and safe prescribing pattern. However, the corticosteroids prescribed by brand names (75.93%) were more than generic names (24.07%), information about the strength of the steroid not mentioned and usage of emollients was less. This indicates the need for continuous medical education for the clinicians
