    Variation and process of life history evolution in insular dwarfism as revealed by a natural experiment

    Islands are a classic focus for evolutionary studies. One topic of great interest has been the evolution of “dwarfs,” significantly smaller island mammals relative to their continental counterparts. Although a consensus has been achieved regarding the multivariate ecological causes behind changes in body size, the processes involved remain largely unexplored. Life history variables, including age at first reproduction, growth rate, and longevity, are likely to be key to understanding the process of insular dwarfism. The Japanese archipelago, with its numerous islands, offers a unique natural experiment for the evolution of different sizes within the same group of organisms; namely, deer. Thus, we investigated eight deer populations with a total number of 52 individuals exhibiting body size variation, both extant and fossil, to clarify the effect of insularity on life history traits. We applied several methods to both extant and extinct populations to resolve life history changes among these deer populations. Skeletochronology, using lines of arrested growth formed in long bones (femur and tibia), successfully reconstructed body growth curves and revealed a gradual change in growth trajectories reflecting the degree of insularity. Slower growth rates with prolonged growth periods in more isolated deer populations were revealed. An extensive examination of bone microstructure further corroborated this finding, with much slower growth and later somatic maturity evident in fossil insular deer isolated for more than 1.5 Myr.     Using Devices to Assess Physical Activity and Sedentary Behavior in a Large Cohort Study: The Women’s Health Study

    In recent years, it has become feasible to use devices for assessing physical activity and sedentary behavior among large numbers of participants in epidemiologic studies, allowing for more precise assessments of these behaviors and quantification of their associations with health outcomes. Between 2011–2015, the Women’s Health Study (WHS) used the Actigraph GT3X+ device to measure physical activity and sedentary behavior over seven days, during waking hours, among 17,708 women (Mage, 72 years) living throughout the United States. Devices were sent to and returned by participants via mail. We describe here the methods used to collect and process the accelerometer data for epidemiologic data analyses. We also provide metrics that describe the quality of the accelerometer data collected, as well as expanded findings regarding previously published associations of physical activity or sedentary behavior with all-cause mortality during an average follow-up of 2.3 years (207 deaths). The WHS is one of the earliest “next generation” epidemiologic studies of physical activity, utilizing wearable devices, in which long-term follow-up of participants for various health outcomes is anticipated.     Este artigo focaliza a reforma da formação inicial de professores da educação básica, em implementação pelo Estado brasileiro desde os anos de 1990. Os elementos centrais dessa reforma evidenciam que o processo de profissionalização toma por base conceitos e práticas que têm origem no campo do trabalho. A noção de competências ocupa lugar central e implementa uma nova lógica educativa, subordinando a esta o currículo e a organização das instituições de formação, objetivando construir um novo tipo de professor, com capacidades subjetivas consoantes àquelas demandadas pelo mercado e pelas novas formas de sociabilidade exclusiva que caracterizam as sociedades capitalistas contemporâneas. O dispositivo legal mais recente - a Portaria nº. 1.403, de 9 de junho de 2003, que pretendeu instituir o Sistema Nacional de Certificação e Formação Continuada de Professores, deu continuidade a esse processo de reforma referenciado em uma perspectiva técnico-instrumental, mas foi recentemente sustado pelo novo dirigente do Ministério de Educação, sob forte pressão do movimento dos educadores. As propostas pelas quais luta o movimento docente nas últimas décadas - formação de qualidade, incentivo às faculdades e centros de educação das universidades como espaços privilegiados de formação de professores, construção da profissionalização e da autonomia e do desenvolvimento intelectual do docente, precisam ser recuperadas neste momento, para que se transformem em efetivas políticas públicas

    A novel accelerometer-based method to describe day-to-day exposure to potentially osteogenic vertical impacts in older adults: findings from a multi-cohort study

    Summary: This observational study assessed vertical impacts experienced in older adults as part of their day-to-day physical activity using accelerometry and questionnaire data. Population-based older adults experienced very limited high-impact activity. The accelerometry method utilised appeared to be valid based on comparisons between different cohorts and with self-reported activity. Introduction: We aimed to validate a novel method for evaluating day-to-day higher impact weight-bearing physical activity (PA) in older adults, thought to be important in protecting against osteoporosis, by comparing results between four cohorts varying in age and activity levels, and with self-reported PA levels. Methods: Participants were from three population-based cohorts, MRC National Survey of Health and Development (NSHD), Hertfordshire Cohort Study (HCS) and Cohort for Skeletal Health in Bristol and Avon (COSHIBA), and the Master Athlete Cohort (MAC). Y-axis peaks (reflecting the vertical when an individual is upright) from a triaxial accelerometer (sampling frequency 50 Hz, range 0–16 g) worn at the waist for 7 days were classified as low (0.5–1.0 g), medium (1.0–1.5 g) or higher (≥1.5 g) impacts. Results: There were a median of 90, 41 and 39 higher impacts/week in NSHD (age 69.5), COSHIBA (age 76.8) and HCS (age 78.5) participants, respectively (total n = 1512). In contrast, MAC participants (age 68.5) had a median of 14,322 higher impacts/week. In the three population cohorts combined, based on comparison of beta coefficients, moderate-high-impact activities as assessed by PA questionnaire were suggestive of stronger association with higher impacts from accelerometers (0.25 [0.17, 0.34]), compared with medium (0.18 [0.09, 0.27]) and low impacts (0.13 [0.07,0.19]) (beta coefficient, with 95 % CI). Likewise in MAC, reported moderate-high-impact activities showed a stronger association with higher impacts (0.26 [0.14, 0.37]), compared with medium (0.14 [0.05, 0.22]) and low impacts (0.03 [−0.02, 0.08]). Conclusions: Our new accelerometer method appears to provide valid measures of higher vertical impacts in older adults. Results obtained from the three population-based cohorts indicate that older adults generally experience very limited higher impact weight-bearing PA

    Daily steps and all-cause mortality: a meta-analysis of 15 international cohorts

    Background Although 10000 steps per day is widely promoted to have health benefits, there is little evidence to support this recommendation. We aimed to determine the association between number of steps per day and stepping rate with all-cause mortality. Methods In this meta-analysis, we identified studies investigating the effect of daily step count on all-cause mortality in adults (aged ≥18 years), via a previously published systematic review and expert knowledge of the field. We asked participating study investigators to process their participant-level data following a standardised protocol. The primary outcome was all-cause mortality collected from death certificates and country registries. We analysed the dose– response association of steps per day and stepping rate with all-cause mortality. We did Cox proportional hazards regression analyses using study-specific quartiles of steps per day and calculated hazard ratios (HRs) with inversevariance weighted random effects models. Findings We identified 15 studies, of which seven were published and eight were unpublished, with study start dates between 1999 and 2018. The total sample included 47 471 adults, among whom there were 3013 deaths (10·1 per 1000 participant-years) over a median follow-up of 7·1 years ([IQR 4·3–9·9]; total sum of follow-up across studies was 297 837 person-years). Quartile median steps per day were 3553 for quartile 1, 5801 for quartile 2, 7842 for quartile 3, and 10 901 for quartile 4. Compared with the lowest quartile, the adjusted HR for all-cause mortality was 0·60 (95% CI 0·51–0·71) for quartile 2, 0·55 (0·49–0·62) for quartile 3, and 0·47 (0·39–0·57) for quartile 4. Restricted cubic splines showed progressively decreasing risk of mortality among adults aged 60 years and older with increasing number of steps per day until 6000–8000 steps per day and among adults younger than 60 years until 8000–10000 steps per day. Adjusting for number of steps per day, comparing quartile 1 with quartile 4, the association between higher stepping rates and mortality was attenuated but remained significant for a peak of 30 min (HR 0·67 [95% CI 0·56–0·83]) and a peak of 60 min (0·67 [0·50–0·90]), but not significant for time (min per day) spent walking at 40 steps per min or faster (1·12 [0·96–1·32]) and 100 steps per min or faster (0·86 [0·58–1·28]). Interpretation Taking more steps per day was associated with a progressively lower risk of all-cause mortality, up to a level that varied by age. The findings from this meta-analysis can be used to inform step guidelines for public health promotion of physical activity

    Identification and single-base gene-editing functional validation of a cis-EPO variant as a genetic predictor for EPO-increasing therapies

    Hypoxia-inducible factor prolyl hydroxylase inhibitors (HIF-PHIs) are currently under clinical development for treating anemia in chronic kidney disease (CKD), but it is important to monitor their cardiovascular safety. Genetic variants can be used as predictors to help inform the potential risk of adverse effects associated with drug treatments. We therefore aimed to use human genetics to help assess the risk of adverse cardiovascular events associated with therapeutically altered EPO levels to help inform clinical trials studying the safety of HIF-PHIs. By performing a genome-wide association meta-analysis of EPO (n = 6,127), we identified a cis-EPO variant (rs1617640) lying in the EPO promoter region. We validated this variant as most likely causal in controlling EPO levels by using genetic and functional approaches, including single-base gene editing. Using this variant as a partial predictor for therapeutic modulation of EPO and large genome-wide association data in Mendelian randomization tests, we found no evidence (at p < 0.05) that genetically predicted long-term rises in endogenous EPO, equivalent to a 2.2-unit increase, increased risk of coronary artery disease (CAD, OR [95% CI] = 1.01 [0.93, 1.07]), myocardial infarction (MI, OR [95% CI] = 0.99 [0.87, 1.15]), or stroke (OR [95% CI] = 0.97 [0.87, 1.07]). We could exclude increased odds of 1.15 for cardiovascular disease for a 2.2-unit EPO increase. A combination of genetic and functional studies provides a powerful approach to investigate the potential therapeutic profile of EPO-increasing therapies for treating anemia in CKD
