Rotor design optimization using a free wake analysis
The aim of this effort was to develop a comprehensive performance optimization capability for tiltrotor and helicopter blades. The analysis incorporates the validated EHPIC (Evaluation of Hover Performance using Influence Coefficients) model of helicopter rotor aerodynamics within a general linear/quadratic programming algorithm that allows optimization using a variety of objective functions involving the performance. The resulting computer code, EHPIC/HERO (HElicopter Rotor Optimization), improves upon several features of the previous EHPIC performance model and allows optimization utilizing a wide spectrum of design variables, including twist, chord, anhedral, and sweep. The new analysis supports optimization of a variety of objective functions, including weighted measures of rotor thrust, power, and propulsive efficiency. The fundamental strength of the approach is that an efficient search for improved versions of the baseline design can be carried out while retaining the demonstrated accuracy inherent in the EHPIC free wake/vortex lattice performance analysis. Sample problems are described that demonstrate the success of this approach for several representative rotor configurations in hover and axial flight. Features that were introduced to convert earlier demonstration versions of this analysis into a generally applicable tool for researchers and designers is also discussed
Analysis of rotor vibratory loads using higher harmonic pitch control
Experimental studies of isolated rotors in forward flight have indicated that higher harmonic pitch control can reduce rotor noise. These tests also show that such pitch inputs can generate substantial vibratory loads. The modification is summarized of the RotorCRAFT (Computation of Rotor Aerodynamics in Forward flighT) analysis of isolated rotors to study the vibratory loading generated by high frequency pitch inputs. The original RotorCRAFT code was developed for use in the computation of such loading, and uses a highly refined rotor wake model to facilitate this task. The extended version of RotorCRAFT incorporates a variety of new features including: arbitrary periodic root pitch control; computation of blade stresses and hub loads; improved modeling of near wake unsteady effects; and preliminary implementation of a coupled prediction of rotor airloads and noise. Correlation studies are carried out with existing blade stress and vibratory hub load data to assess the performance of the extended code
Free wake analysis of hover performance using a new influence coefficient method
A new approach to the prediction of helicopter rotor performance using a free wake analysis was developed. This new method uses a relaxation process that does not suffer from the convergence problems associated with previous time marching simulations. This wake relaxation procedure was coupled to a vortex-lattice, lifting surface loads analysis to produce a novel, self contained performance prediction code: EHPIC (Evaluation of Helicopter Performance using Influence Coefficients). The major technical features of the EHPIC code are described and a substantial amount of background information on the capabilities and proper operation of the code is supplied. Sample problems were undertaken to demonstrate the robustness and flexibility of the basic approach. Also, a performance correlation study was carried out to establish the breadth of applicability of the code, with very favorable results
High-Temperature Smart Structures for Engine Noise Reduction and Performance Enhancement
One of key NASA goals is to develop and integrate noise reduction technology to enable unrestricted air transportation service to all communities. One of the technical priorities of this activity has been to account for and reduce noise via propulsion/airframe interactions, identifying advanced concepts to be integrated with the airframe to mitigate these noise-producing mechanisms. An adaptive geometry chevron using embedded smart structures technology offers the possibility of maximizing engine performance while retaining and possibly enhancing the favorable noise characteristics of current designs. New high-temperature shape memory alloy (HTSMA) materials technology enables the devices to operate in both low-temperature (fan) and high-temperature (core) exhaust flows. Chevron-equipped engines have demonstrated reduced noise in testing and operational use. It is desirable to have the noise benefits of chevrons in takeoff/landing conditions, but have them deployed into a minimum drag position for cruise flight. The central feature of the innovation was building on rapidly maturing HTSMA technology to implement a next-generation aircraft noise mitigation system centered on adaptive chevron flow control surfaces. In general, SMA-actuated devices have the potential to enhance the demonstrated noise reduction effectiveness of chevron systems while eliminating the associated performance penalty. The use of structurally integrated smart devices will minimize the mechanical and subsystem complexity of this implementation. The central innovations of the effort entail the modification of prior chevron designs to include a small cut that relaxes structural stiffness without compromising the desired flow characteristics over the surface; the reorientation of SMA actuation devices to apply forces to deflect the chevron tip, exploiting this relaxed stiffness; and the use of high-temperature SMA (HTSMA) materials to enable operation in the demanding core chevron environment. The overall conclusion of these design studies was that the cut chevron concept is a critical enabling step in bringing the variable geometry core chevron within reach. The presence of the cut may be aerodynamically undesirable in some respects, but it is present only when the chevron is not immersed in the core jet exhaust. When deployed, the gap closes as the chevron tip enters the high-speed, high-temperature core stream. Aeroacoustic testing and flow visualization support the contention that this cut is inconsequential to chevron performance
Computation of rotor aerodynamic loads in forward flight using a full-span free wake analysis
The development of an advanced computational analysis of unsteady aerodynamic loads on isolated helicopter rotors in forward flight is described. The primary technical focus of the development was the implementation of a freely distorting filamentary wake model composed of curved vortex elements laid out along contours of constant vortex sheet strength in the wake. This model captures the wake generated by the full span of each rotor blade and makes possible a unified treatment of the shed and trailed vorticity in the wake. This wake model was coupled to a modal analysis of the rotor blade dynamics and a vortex lattice treatment of the aerodynamic loads to produce a comprehensive model for rotor performance and air loads in forward flight dubbed RotorCRAFT (Computation of Rotor Aerodynamics in Forward Flight). The technical background on the major components of this analysis are discussed and the correlation of predictions of performance, trim, and unsteady air loads with experimental data from several representative rotor configurations is examined. The primary conclusions of this study are that the RotorCRAFT analysis correlates well with measured loads on a variety of configurations and that application of the full span free wake model is required to capture several important features of the vibratory loading on rotor blades in forward flight
Computational analysis of high resolution unsteady airloads for rotor aeroacoustics
The study of helicopter aerodynamic loading for acoustics applications requires the application of efficient yet accurate simulations of the velocity field induced by the rotor's vortex wake. This report summarizes work to date on the development of such an analysis, which builds on the Constant Vorticity Contour (CVC) free wake model, previously implemented for the study of vibratory loading in the RotorCRAFT computer code. The present effort has focused on implementation of an airload reconstruction approach that computes high resolution airload solutions of rotor/rotor-wake interactions required for acoustics computations. Supplementary efforts on the development of improved vortex core modeling, unsteady aerodynamic effects, higher spatial resolution of rotor loading, and fast vortex wake implementations have substantially enhanced the capabilities of the resulting software, denoted RotorCRAFT/AA (AeroAcoustics). Results of validation calculations using recently acquired model rotor data show that by employing airload reconstruction it is possible to apply the CVC wake analysis with temporal and spatial resolution suitable for acoustics applications while reducing the computation time required by one to two orders of magnitude relative to that required by direct calculations. Promising correlation with this body of airload and noise data has been obtained for a variety of rotor configurations and operating conditions
Development of a HTSMA-Actuated Surge Control Rod for High-Temperature Turbomachinery Applications
In recent years, a demand for compact, lightweight, solid-state actuation systems has emerged, driven in part by the needs of the aeronautics industry. However, most actuation systems used in turbomachinery require not only elevated temperature but high-force capability. As a result, shape memory alloy (SMA) based systems have worked their way to the forefront of a short list of viable options to meet such a technological challenge. Most of the effort centered on shape memory systems to date has involved binary NiTi alloys but the working temperatures required in many aeronautics applications dictate significantly higher transformation temperatures than the binary systems can provide. Hence, a high temperature shape memory alloy (HTSMA) based on NiTiPdPt, having a transformation temperature near 300 C, was developed. Various thermo-mechanical processing schemes were utilized to further improve the dimensional stability of the alloy and it was later extruded/drawn into wire form to be more compatible with envisioned applications. Mechanical testing on the finished wire form showed reasonable work output capability with excellent dimensional stability. Subsequently, the wire form of the alloy was incorporated into a benchtop system, which was shown to provide the necessary stroke requirements of approx.0.125 inches for the targeted surge-control application. Cycle times for the actuator were limited to ~4 seconds due to control and cooling constraints but this cycle time was determined to be adequate for the surge control application targeted as the primary requirement was initial actuation of a surge control rod, which could be completed in approximately one second
Current European Labyrinthula zosterae Are Not Virulent and Modulate Seagrass (Zostera marina) Defense Gene Expression
Pro- and eukaryotic microbes associated with multi-cellular organisms are receiving increasing attention as a driving factor in ecosystems. Endophytes in plants can change host performance by altering nutrient uptake, secondary metabolite production or defense mechanisms. Recent studies detected widespread prevalence of Labyrinthula zosterae in European Zostera marina meadows, a protist that allegedly caused a massive amphi-Atlantic seagrass die-off event in the 1930's, while showing only limited virulence today. As a limiting factor for pathogenicity, we investigated genotypeĆgenotype interactions of host and pathogen from different regions (10ā100 km-scale) through reciprocal infection. Although the endophyte rapidly infected Z. marina, we found little evidence that Z. marina was negatively impacted by L. zosterae. Instead Z. marina showed enhanced leaf growth and kept endophyte abundance low. Moreover, we found almost no interaction of protistĆeelgrass-origin on different parameters of L. zosterae virulence/Z. marina performance, and also no increase in mortality after experimental infection. In a target gene approach, we identified a significant down-regulation in the expression of 6/11 genes from the defense cascade of Z. marina after real-time quantitative PCR, revealing strong immune modulation of the host's defense by a potential parasite for the first time in a marine plant. Nevertheless, one gene involved in phenol synthesis was strongly up-regulated, indicating that Z. marina plants were probably able to control the level of infection. There was no change in expression in a general stress indicator gene (HSP70). Mean L. zosterae abundances decreased below 10% after 16 days of experimental runtime. We conclude that under non-stress conditions L. zosterae infection in the study region is not associated with substantial virulence
The steady-state transcriptome of the four major life-cycle stages of Trypanosoma cruzi
<p>Abstract</p> <p>Background</p> <p>Chronic chagasic cardiomyopathy is a debilitating and frequently fatal outcome of human infection with the protozoan parasite, <it>Trypanosoma cruzi</it>. Microarray analysis of gene expression during the <it>T. cruzi </it>life-cycle could be a valuable means of identifying drug and vaccine targets based on their appropriate expression patterns, but results from previous microarray studies in <it>T. cruzi </it>and related kinetoplastid parasites have suggested that the transcript abundances of most genes in these organisms do not vary significantly between life-cycle stages.</p> <p>Results</p> <p>In this study, we used whole genome, oligonucleotide microarrays to globally determine the extent to which <it>T. cruzi </it>regulates mRNA relative abundances over the course of its complete life-cycle. In contrast to previous microarray studies in kinetoplastids, we observed that relative transcript abundances for over 50% of the genes detected on the <it>T. cruzi </it>microarrays were significantly regulated during the <it>T. cruzi </it>life-cycle. The significant regulation of 25 of these genes was confirmed by quantitative reverse-transcriptase PCR (qRT-PCR). The <it>T. cruzi </it>transcriptome also mirrored published protein expression data for several functional groups. Among the differentially regulated genes were members of paralog clusters, nearly 10% of which showed divergent expression patterns between cluster members.</p> <p>Conclusion</p> <p>Taken together, these data support the conclusion that transcript abundance is an important level of gene expression regulation in <it>T. cruzi</it>. Thus, microarray analysis is a valuable screening tool for identifying stage-regulated <it>T. cruzi </it>genes and metabolic pathways.</p
Integrative Annotation of 21,037 Human Genes Validated by Full-Length cDNA Clones
The human genome sequence defines our inherent biological potential; the realization of the biology encoded therein requires knowledge of the function of each gene. Currently, our knowledge in this area is still limited. Several lines of investigation have been used to elucidate the structure and function of the genes in the human genome. Even so, gene prediction remains a difficult task, as the varieties of transcripts of a gene may vary to a great extent. We thus performed an exhaustive integrative characterization of 41,118 full-length cDNAs that capture the gene transcripts as complete functional cassettes, providing an unequivocal report of structural and functional diversity at the gene level. Our international collaboration has validated 21,037 human gene candidates by analysis of high-quality full-length cDNA clones through curation using unified criteria. This led to the identification of 5,155 new gene candidates. It also manifested the most reliable way to control the quality of the cDNA clones. We have developed a human gene database, called the H-Invitational Database (H-InvDB; http://www.h-invitational.jp/). It provides the following: integrative annotation of human genes, description of gene structures, details of novel alternative splicing isoforms, non-protein-coding RNAs, functional domains, subcellular localizations, metabolic pathways, predictions of protein three-dimensional structure, mapping of known single nucleotide polymorphisms (SNPs), identification of polymorphic microsatellite repeats within human genes, and comparative results with mouse full-length cDNAs. The H-InvDB analysis has shown that up to 4% of the human genome sequence (National Center for Biotechnology Information build 34 assembly) may contain misassembled or missing regions. We found that 6.5% of the human gene candidates (1,377 loci) did not have a good protein-coding open reading frame, of which 296 loci are strong candidates for non-protein-coding RNA genes. In addition, among 72,027 uniquely mapped SNPs and insertions/deletions localized within human genes, 13,215 nonsynonymous SNPs, 315 nonsense SNPs, and 452 indels occurred in coding regions. Together with 25 polymorphic microsatellite repeats present in coding regions, they may alter protein structure, causing phenotypic effects or resulting in disease.
