The total charge of pulsars is calculated after considering the electromagnetic field in and out the star under a specific condition.     Quercetin inhibits porcine intestinal inflammation in vitro

    Purpose: To investigate the effect of Quercetin (Que) on inflammatory reaction in intestinal porcine enterocyte cells, IPEC-J2, induced by lipopolysaccharide (LPS).Methods: IPEC-J2 cells were pretreated with Que and then incubated with LPS. Cell viability, cell morphology, nitric oxide (NO) content, gene expression levels of interleukin-6 (IL-6) and interleukin-8 (IL-8), were then evaluated.Results: LPS incubation at 10 ÎĽg/mL for 24 h showed no effect on the viability and cell morphology of IPEC-J2, while Que pre-incubation significantly enhanced the viability of IPEC-J2 and improved cell morphology (p < 0.05). Que decreased the expression levels of pro-inflammatory cytokines IL-6 mRNA and IL-8 mRNA raised by LPS (p < 0.05). Interestingly, both LPS and Que demonstrated no influence on the release of NO.Conclusion: Pre-treatment of Que shows a positive effect on intestinal porcine enterocyte cells and inhibited porcine intestinal inflammation in vitro induced by LPS, but the mechanism may be not associated with NO-related signaling pathway. Therefore, Que might have a potential effect as a veterinary drug or feed additive for the treatment of enteritis in pigs.Keywords: Quercetin, Inflammatory cytokines, Intestinal porcine enterocyte cells, Lipopolysaccharid

    Critical Role of FoxO1 in Granulosa Cell Apoptosis Caused by Oxidative Stress and Protective Effects of Grape Seed Procyanidin B2

    Reactive oxygen species (ROS) are closely related to the follicular granulosa cell apoptosis. Grape seed procyanidin B2 (GSPB2) has been reported to possess potent antioxidant activity. However, the GSPB2-mediated protective effects and the underlying molecular mechanisms in granulosa cell apoptosis process remain unknown. In this study, we showed for the first time that GSPB2 treatment decreased FoxO1 protein level, improved granulosa cell viability, upregulated LC3-II protein level, and reduced granulosa cell apoptosis rate. Under a condition of oxidative stress, GSPB2 reversed FoxO1 nuclear localization and increased its level in cytoplasm. In addition, FoxO1 knockdown inhibited the protective effects of GSPB2 induced. Our findings suggest that FoxO1 plays a pivotal role in regulating autophagy in granulosa cells, GSPB2 exerts a potent and beneficial role in reducing granulosa cell apoptosis and inducing autophagy process, and targeting FoxO1 could be significant in fighting against oxidative stress-reduced female reproductive system diseases

    HER2 Targeted Molecular MR Imaging Using a De Novo Designed Protein Contrast Agent

    The application of magnetic resonance imaging (MRI) to non-invasively assess disease biomarkers has been hampered by the lack of desired contrast agents with high relaxivity, targeting capability, and optimized pharmacokinetics. We have developed a novel MR imaging probe targeting to HER2, a biomarker for various cancer types and a drug target for anti-cancer therapies. This multimodal HER20targeted MR imaging probe integrates a de novo designed protein contrast agent with a high affinity HER2 affibody and a near IR fluorescent dye. Our probe can differentially monitor tumors with different expression levels of HER2 in both human cell lines and xenograft mice models. In addition to its 100-fold higher dose efficiency compared to clinically approved non-targeting contrast agent DTPA, our developed agent also exhibits advantages in crossing the endothelial boundary, tissue distribution, and tumor tissue retention over reported contrast agents as demonstrated by even distribution of the imaging probe across the entire tumor mass. This contrast agent will provide a powerful tool for quantitative assessment of molecular markers, and improved resolution for diagnosis, prognosis and drug discovery

    An anoikis-related gene signature predicts prognosis and reveals immune infiltration in hepatocellular carcinoma

    BackgroundHepatocellular carcinoma (HCC) is a global health burden with poor prognosis. Anoikis, a novel programmed cell death, has a close interaction with metastasis and progression of cancer. In this study, we aimed to construct a novel bioinformatics model for evaluating the prognosis of HCC based on anoikis-related gene signatures as well as exploring the potential mechanisms.Materials and methodsWe downloaded the RNA expression profiles and clinical data of liver hepatocellular carcinoma from TCGA database, ICGC database and GEO database. DEG analysis was performed using TCGA and verified in the GEO database. The anoikis-related risk score was developed via univariate Cox regression, LASSO Cox regression and multivariate Cox regression, which was then used to categorize patients into high- and low-risk groups. Then GO and KEGG enrichment analyses were performed to investigate the function between the two groups. CIBERSORT was used for determining the fractions of 22 immune cell types, while the ssGSEA analyses was used to estimate the differential immune cell infiltrations and related pathways. The “pRRophetic” R package was applied to predict the sensitivity of administering chemotherapeutic and targeted drugs.ResultsA total of 49 anoikis-related DEGs in HCC were detected and 3 genes (EZH2, KIF18A and NQO1) were selected out to build a prognostic model. Furthermore, GO and KEGG functional enrichment analyses indicated that the difference in overall survival between risk groups was closely related to cell cycle pathway. Notably, further analyses found the frequency of tumor mutations, immune infiltration level and expression of immune checkpoints were significantly different between the two risk groups, and the results of the immunotherapy cohort showed that patients in the high-risk group have a better immune response. Additionally, the high-risk group was found to have higher sensitivity to 5-fluorouracil, doxorubicin and gemcitabine.ConclusionThe novel signature of 3 anoikis-related genes (EZH2, KIF18A and NQO1) can predict the prognosis of patients with HCC, and provide a revealing insight into personalized treatments in HCC

    Common Features in Electronic Structure of the Fe-Based Layered Superconductors from Photoemission Spectroscopy

    High resolution photoemission measurements have been carried out on non-superconducting LaOFeAs parent compound and various superconducting R(O1-xFx)FeAs (R=La, Ce and Pr) compounds. We found that the parent LaOFeAs compound shows a metallic character. Through extensive measurements, we have identified several common features in the electronic structure of these Fe-based compounds: (1). 0.2 eV feature in the valence band; (2). A universal 13~16 meV feature; (3). A clear Fermi cutoff showing zero leading-edge shift in the superconducting state;(4). Lack of superconducting coherence peak(s); (5). Near EF spectral weight suppression with decreasing temperature. These universal features can provide important information about band structure, superconducting gap and pseudogap in these Fe-based materials.Comment: 5 pages,4 figure

    Multiple Nodeless Superconducting Gaps in (Ba0.6K0.4)Fe2As2 Superconductor from Angle-Resolved Photoemission Spectroscopy

    High resolution angle-resolved photoemission measurements have been carried out to study the superconducting gap in the (Ba0.6K0.4)Fe2As2 superconductor with Tc=35 K. Two hole-like Fermi surface sheets around the G(0,0) point exhibit different superconducting gaps. The inner Fermi surface sheet shows larger (10-12 meV) and slightly momentum-dependent gap while the outer one has smaller (7-8 meV) and nearly isotropic gap. The lack of gap node in both Fermi surface sheets favours s-wave superconducting gap symmetry. Superconducting gap opening is also observed at the M(pi,pi) point. The two Fermi surface spots near the M point are gapped below Tc but the gap persists above Tc. The rich and detailed superconducting gap information will provide key insights and constraints in understanding pairing mechanism in the iron-based superconductors.Comment: 11 pages, 4 figure
