    Idling emission at intersection and exploring suitable mitigation measures

    Variety of road based transport modes catering to the transport demand ply in large number on the road system of urban India. As a result, the traffic and transportation problems are aggravating day by day. These problems manifest in the form of increased traffic congestion, increased air and noise pollution, accidents, delays etc. The consumption of fuel is on the increase due to enhanced trip lengths, shift of modal share towards personalized modes of travel and at signalized intersections due to idling of vehicles during stoppage phases. The results of a study conducted by CRRI in 2005 estimated that fuel worth of Indian Rupees. 1, 0000 Million is wasted every year in Delhi by vehicles idling at 600 signalized traffic signals and was around 15% of the total fuel consumed annually in Delhi. In addition, the time loss to the commuters is also associated with the delays. The running engines during idling also generate emissions, which are harmful both for human health and ecology. One of the ways to conserve fuels is to minimize its wastage. It is necessary to understand the amount of fuel loss and emissions generated at signalized intersections in the country. By applying various engineering / management measures the fuel wastage and associated emissions can be reduced. Presently there is no model that can give the amount of fuel loss and emissions in relation to the type of vehicle and delays. In this paper a review of the study for exploring suitable mitigation measures to improve the signalized intersections are presented

    PhyloToAST: Bioinformatics Tools for Species-Level Analysis and Visualization of Complex Microbial Datasets

    The 16S rRNA gene is widely used for taxonomic profiling of microbial ecosystems; and recent advances in sequencing chemistry have allowed extremely large numbers of sequences to be generated from minimal amounts of biological samples. Analysis speed and resolution of data to species-level taxa are two important factors in large-scale explorations of complex microbiomes using 16S sequencing. We present here new software, Phylogenetic Tools for Analysis of Species-level Taxa (PhyloToAST), that completely integrates with the QIIME pipeline to improve analysis speed, reduce primer bias (requiring two sequencing primers), enhance species-level analysis, and add new visualization tools. The code is free and open source, and can be accessed at http://phylotoast.org

    High-spin spectroscopy of I-122

    High-spin states in I-122 have been investigated using the Cd-116(B-11,5n)I-122 reaction at a beam energy of 65 MeV and gamma-ray coincidence events were recorded with the INGA spectrometer. The level scheme of I-122 has been extended up to spin I = 30. Experimental features, such as band-crossing frequencies, aligned angular momenta, signature splitting, and B(M1)/B(E2) ratios have been used for configuration assignments to low-energy band structures. Maximally aligned states involving all eight particles outside the Sn-114 core and states with one particle antialigned have been identified. Cranked Nilsson-Strutinsky calculations have been used to interpret high-spin structures

    Traditional birth attendants lack basic information on HIV and safe delivery practices in rural Mysore, India

    <p>Abstract</p> <p>Background</p> <p>There is little research on HIV awareness and practices of traditional birth attendants (TBA) in India. This study investigated knowledge and attitudes among rural TBA in Karnataka as part of a project examining how traditional birth attendants could be integrated into prevention-of-mother-to-child transmission of HIV (PMTCT) programs in India.</p> <p>Methods</p> <p>A cross-sectional survey was conducted between March 2008 and January 2009 among TBA in 144 villages in Mysore <it>Taluk</it>, Karnataka. Following informed consent, TBA underwent an interviewer-administered questionnaire in the local language of <it>Kannada </it>on practices and knowledge around birthing and HIV/PMTCT.</p> <p>Results</p> <p>Of the 417 TBA surveyed, the median age was 52 years and 96% were Hindus. A majority (324, 77.7%) had no formal schooling, 88 (21.1%) had up to 7 years and 5 (1%) had more than 7 yrs of education. Only 51 of the 417 TBA (12%) reported hearing about HIV/AIDS. Of those who had heard about HIV/AIDS, only 36 (72%) correctly reported that the virus could be spread from mother to child; 37 (74%) identified unprotected sex as a mode of transmission; and 26 (51%) correctly said healthy looking people could spread HIV. Just 22 (44%) knew that infected mothers could lower the risk of transmitting the virus to their infants. An overwhelming majority of TBA (401, 96.2%) did not provide antenatal care to their clients. Over half (254, 61%) said they would refer the woman to a hospital if she bled before delivery, and only 53 (13%) felt referral was necessary if excessive bleeding occurred after birth.</p> <p>Conclusions</p> <p>Traditional birth attendants will continue to play an important role in maternal child health in India for the foreseeable future. This study demonstrates that a majority of TBA lack basic information about HIV/AIDS and safe delivery practices. Given the ongoing shortage of skilled birth attendance in rural areas, more studies are needed to examine whether TBA should be trained and integrated into PMTCT and maternal child health programs in India.</p

    Magnetic field-induced non-trivial electronic topology in Fe3−xGeTe2

    The anomalous Hall, Nernst and thermal Hall coefficients of Fe3x_{3-x}GeTe2_2 display several features upon cooling, like a reversal in the Nernst signal below T=50T = 50 K pointing to a topological transition (TT) associated to the development of magnetic spin textures. Since the anomalous transport variables are related to the Berry curvature, a possible TT might imply deviations from the Wiedemann-Franz (WF) law. However, the anomalous Hall and thermal Hall coefficients of Fe3x_{3-x}GeTe2_2 are found, within our experimental accuracy, to satisfy the WF law for magnetic-fields μ0H\mu_0H applied along its inter-layer direction. Surprisingly, large anomalous transport coefficients are also observed for μ0H\mu_0H applied along the planar \emph{a}-axis as well as along the gradient of the chemical potential, a configuration that should not lead to their observation due to the absence of Lorentz force. However, as μ0H\mu_0H \| \emph{a}-axis is increased, magnetization and neutron scattering indicate just the progressive canting of the magnetic moments towards the planes followed by their saturation. These anomalous planar quantities are found to not scale with the component of the planar magnetization (MM_{\|}), showing instead a sharp decrease beyond μ0H=\sim \mu_0 H_{\|} = 4 T which is the field required to align the magnetic moments along μ0H\mu_0 H_{\|}. We argue that locally chiral spin structures, such as skyrmions, and possibly skyrmion tubes, lead to a field dependent spin-chirality and hence to a novel type of topological anomalous transport. Locally chiral spin-structures are captured by our Monte-Carlo simulations incorporating small Dzyaloshinskii-Moriya and biquadratic exchange interactions.Comment: 34 pages, 10 figures, submitted to Applied Physics Review

    Target Region Selection Is a Critical Determinant of Community Fingerprints Generated by 16S Pyrosequencing

    Pyrosequencing of 16S rRNA genes allows for in-depth characterization of complex microbial communities. Although it is known that primer selection can influence the profile of a community generated by sequencing, the extent and severity of this bias on deep-sequencing methodologies is not well elucidated. We tested the hypothesis that the hypervariable region targeted for sequencing and primer degeneracy play important roles in influencing the composition of 16S pyrotag communities. Subgingival plaque from deep sites of current smokers with chronic periodontitis was analyzed using Sanger sequencing and pyrosequencing using 4 primer pairs. Greater numbers of species were detected by pyrosequencing than by Sanger sequencing. Rare taxa constituted nearly 6% of each pyrotag community and less than 1% of the Sanger sequencing community. However, the different target regions selected for pyrosequencing did not demonstrate a significant difference in the number of rare and abundant taxa detected. The genera Prevotella, Fusobacterium, Streptococcus, Granulicatella, Bacteroides, Porphyromonas and Treponema were abundant when the V1–V3 region was targeted, while Streptococcus, Treponema, Prevotella, Eubacterium, Porphyromonas, Campylobacer and Enterococcus predominated in the community generated by V4–V6 primers, and the most numerous genera in the V7–V9 community were Veillonella, Streptococcus, Eubacterium, Enterococcus, Treponema, Catonella and Selenomonas. Targeting the V4–V6 region failed to detect the genus Fusobacterium, while the taxa Selenomonas, TM7 and Mycoplasma were not detected by the V7–V9 primer pairs. The communities generated by degenerate and non-degenerate primers did not demonstrate significant differences. Averaging the community fingerprints generated by V1–V3 and V7–V9 primers providesd results similar to Sanger sequencing, while allowing a significantly greater depth of coverage than is possible with Sanger sequencing. It is therefore important to use primers targeted to these two regions of the 16S rRNA gene in all deep-sequencing efforts to obtain representational characterization of complex microbial communities

    Yeast H2A.Z, FACT complex and RSC regulate transcription of tRNA gene through differential dynamics of flanking nucleosomes

    FACT complex is involved in elongation and ensures fidelity in the initiation step of transcription by RNA polymerase (pol) II. Histone variant H2A.Z is found in nucleosomes at the 5′-end of many genes. We report here H2A.Z-chaperone activity of the yeast FACT complex on the short, nucleosome-free, non-coding, pol III-transcribed yeast tRNA genes. On a prototype gene, yeast SUP4, chromatin remodeler RSC and FACT regulate its transcription through novel mechanisms, wherein the two gene-flanking nucleosomes containing H2A.Z, play different roles. Nhp6, which ensures transcription fidelity and helps load yFACT onto the gene flanking nucleosomes, has inhibitory role. RSC maintains a nucleosome abutting the gene terminator downstream, which results in reduced transcription rate in active state while H2A.Z probably helps RSC in keeping the gene nucleosome-free and serves as stress-sensor. All these factors maintain an epigenetic state which allows the gene to return quickly from repressed to active state and tones down the expression from the active SUP4 gene, required probably to maintain the balance in cellular tRNA pool

    Influenza A Virus Nucleoprotein Exploits Hsp40 to Inhibit PKR Activation

    BACKGROUND: Double-stranded RNA dependent protein kinase (PKR) is a key regulator of the anti-viral innate immune response in mammalian cells. PKR activity is regulated by a 58 kilo Dalton cellular inhibitor (P58(IPK)), which is present in inactive state as a complex with Hsp40 under normal conditions. In case of influenza A virus (IAV) infection, P58(IPK) is known to dissociate from Hsp40 and inhibit PKR activation. However the influenza virus component responsible for PKR inhibition through P58(IPK) activation was hitherto unknown. PRINCIPAL FINDINGS: Human heat shock 40 protein (Hsp40) was identified as an interacting partner of Influenza A virus nucleoprotein (IAV NP) using a yeast two-hybrid screen. This interaction was confirmed by co-immunoprecipitation studies from mammalian cells transfected with IAV NP expressing plasmid. Further, the IAV NP-Hsp40 interaction was validated in mammalian cells infected with various seasonal and pandemic strains of influenza viruses. Cellular localization studies showed that NP and Hsp40 co-localize primarily in the nucleus. During IAV infection in mammalian cells, expression of NP coincided with the dissociation of P58(IPK) from Hsp40 and decrease PKR phosphorylation. We observed that, plasmid based expression of NP in mammalian cells leads to decrease in PKR phosphorylation. Furthermore, inhibition of NP expression during influenza virus replication led to PKR activation and concomitant increase in eIF2α phosphorylation. Inhibition of NP expression also led to reduced IRF3 phosphorylation, enhanced IFN β production and concomitant reduction of virus replication. Taken together our data suggest that NP is the viral factor responsible for P58(IPK) activation and subsequent inhibition of PKR-mediated host response during IAV infection. SIGNIFICANCE: Our findings demonstrate a novel role of IAV NP in inhibiting PKR-mediated anti-viral host response and help us understand P58(IPK) mediated inhibition of PKR activity during IAV infection
