    High bit-rate digital communication through metal channels

    The need to transmit digital information across metallic barriers arises frequently in industrial control applications. In some applications, the barrier can be penetrated with wiring, while in others this may not be possible. For example, metal bulkheads, pressure vessels, or pipelines may require a level of mechanical integrity that prohibits mechanical penetration. This study investigates the use of ultrasonic signaling for data transmission across metallic barriers, discusses the associated challenges, and analyzes several alternative communication system implementations.Several recent e orts have been made to develop through-metal ultrasonic communication systems, with approaches ranging widely in bitrate, complexity, and power requirements. The transceiver designs presented here are intended to cover a range of target applications. In systems having low data rate requirements, simple transceivers with low hardware/software complexity can be used. At high data rates, however, severe echoing in the ultrasonic channel leads to intersymbol interference. Reliable high speed communication therefore requires the use of channel equalizers, and results in a transceiver with higher hardware/software complexity.In this thesis, issues related to the design of reliable through-metal ultrasonic communication systems are discussed. These include (1) the development of mathematical models used to characterize the channel, (2) application of equalization techniques needed to achieve high-speed communication, and (3) analysis of hardware/software complexity for alternative transceiver designs.Several groups have developed through-metal ultrasonic communication systems in the recent past, though none has produced a mathematical model that accurately describes the phenomena found within the channel. The channel model developed in this thesis can be used at several stages of the transceiver design process, from transducer selection through channel equalizer design and ultimately system performance simulation.Using this channel model, we go on to develop and test several ultrasonic throughmetal transceiver designs. Ultrasonic through-metal communication systems are finding use in a wide variety of applications. Some require high throughput, while others require low power consumption. The motivation for developing several designs { ranging from low complexity, low power to high complexity, high throughput { is so that the best design can be matched to each application.After these transceiver designs are developed, we present an analysis of their computational requirements so that the most appropriate transceiver can be chosen for a given application.Ph.D., Electrical Engineering -- Drexel University, 201

    Cytokine Regulation in Human CD4 T Cells by the Aryl Hydrocarbon Receptor and Gq-Coupled Receptors

    Th17 cells contribute to host defense on mucosal surfaces but also provoke autoimmune diseases when directed against self-antigens. Identifying therapeutic targets that regulate Th17 cell differentiation and/or cytokine production has considerable value. Here, we study the aryl hydrocarbon receptor (AhR)-dependent transcriptome in human CD4 T cells treated with Th17-inducing cytokines. We show that the AhR reciprocally regulates IL-17 and IL-22 production in human CD4 T cells. Global gene expression analysis revealed that AhR ligation decreased IL21 expression, correlating with delayed upregulation of RORC during culture with Th17-inducing cytokines. Several of the AhR-dependent genes have known roles in cellular assembly, organization, development, growth and proliferation. We further show that expression of GPR15, GPR55 and GPR68 positively correlates with IL-22 production in the presence of the AhR agonist FICZ. Activation of GPR68 with the lorazepam derivative ogerin resulted in suppression of IL-22 and IL-10 secretion by T cells, with no effect on IL-17. Under neutral Th0 conditions, ogerin and the Gq/11 receptor inhibitor YM254890 blunted IL-22 induction by FICZ. These data reveal the AhR-dependent transcriptome in human CD4 T cells and suggest the mechanism through which the AhR regulates T cell function may be partially dependent on Gq-coupled receptors including GPR68

    RGS16, a novel p53 and pRb cross-talk candidate inhibits migration and invasion of pancreatic cancer cells

    Data collected since the discovery of p53 and pRb/RB1 suggests these tumor suppressors cooperate to inhibit tumor progression. Patients who have mutations in both p53 and RB1 genes have increased tumor reoccurrence and decreased survival compared to patients with only one tumor suppressor gene inactivated. It remains unclear how p53 and pRb cooperate toward inhibiting tumorigenesis. Using RNA expression profiling we identified 179 p53 and pRb cross-talk candidates in normal lung fibroblasts (WI38) cells exogenously coexpressing p53 and pRb. Regulator of G protein signaling 16 (RGS16) was among the p53 and pRb cross-talk candidates and has been implicated in inhibiting activation of several oncogenic pathways associated with proliferation, migration, and invasion of cancer cells. RGS16 has been found to be downregulated in pancreatic cancer patients with metastases compared to patients without metastasis. Expression of RGS16 mRNA was decreased in the pancreatic cancer cell lines tested compared to control. Expression of RGS16 inhibited migration of the BxPC-3 and AsPC-1 but not PANC-1 cells and inhibited invasion of BxPC-3 and AsPC-1 cells with no impact on cell viability.     Global analysis of gene expression changes during retinoic acid-induced growth arrest and differentiation of melanoma: comparison to differentially expressed genes in melanocytes vs melanoma

    <p>Abstract</p> <p>Background</p> <p>The incidence of malignant melanoma has significantly increased over the last decade. Some of these malignancies are susceptible to the growth inhibitory and pro-differentiating effects of all-<it>trans</it>-retinoic acid (RA). The molecular changes responsible for the biological activity of RA in melanoma are not well understood.</p> <p>Results</p> <p>In an analysis of sequential global gene expression changes during a 4–48 h RA treatment of B16 mouse melanoma cells, we found that RA increased the expression of 757 genes and decreased the expression of 737 genes. We also compared the gene expression profile (no RA treatment) between non-malignant melan-a mouse melanocytes and B16 melanoma cells. Using the same statistical test, we found 1495 genes whose expression was significantly higher in melan-a than in B16 cells and 2054 genes whose expression was significantly lower in melan-a than in B16 cells. By intersecting these two gene sets, we discovered a common set of 233 genes whose RNA levels were significantly different between B16 and melan-a cells and whose expression was altered by RA treatment. Within this set, RA treatment altered the expression of 203 (87%) genes toward the melan-a expression level. In addition, hierarchical clustering showed that after 48 h of RA treatment expression of the 203 genes was more closely related to the melan-a gene set than any other RA treatment time point. Functional analysis of the 203 gene set indicated that RA decreased expression of mRNAs that encode proteins involved in cell division/cell cycle, DNA replication, recombination and repair, and transcription regulation. Conversely, it stimulated genes involved in cell-cell signaling, cell adhesion and cell differentiation/embryonic development. Pathway analysis of the 203 gene set revealed four major hubs of connectivity: CDC2, CHEK1, CDC45L and MCM6.</p> <p>Conclusion</p> <p>Our analysis of common genes in the 48 h RA-treatment of B16 melanoma cells and untreated B16 vs. melan-a data set show that RA "normalized" the expression of genes involved in energy metabolism, DNA replication, DNA repair and differentiation. These results are compatible with the known growth inhibitory and pro-differentiating effects of RA. Pathway analysis suggests that CDC2, CHEK1, CDC45L and MCM6 are key players in mediating the biological activity of RA in B16 melanoma cells.</p

    Chemical and physical modifications of polyethylene containing nanostructures

    The polyethylene is an interesting polymer with a good mechanical strength, ductility, biocompatibility and chemical inertia. It founds several applications in many fields, such as chemistry, engineering, bio-medicine and micro-electronics. The polyethylene chemical and physical properties can be modified embedding different nanostructures in its bulk. Carbon nanotubes, metallic oxides, coloured pigments and other species can be inserted at different concentrations in the polymer material during the preparation phase of thin films and sheets. Mechanical, optical, thermal and chemical properties can be modified significantly depending on the filler concentration. The absorption coefficient at different radiations can be controlled by the amount of doping structure. In the visible region, for example, the high transparency of pure polyethylene can be strongly reduced by low concentrations of carbon nanotubes. The colour, the mechanical resistance, the wet ability of the polyethylene and other parameters can be changed by the doping species in order to prepare special devices useful for many applications

    Identification of the PS1 Thr147Ile Variant in a Family with Very Early Onset Dementia and Expressive Aphasia

    Background: Early onset dementias have variable clinical presentations and are often difficult to diagnose. We established a family pedigree that demonstrated consistent recurrence of very early onset dementia in successive generations. Objective and Method: In order to refine the diagnosis in this family, we sequenced the exomes of two affected family members and relied on discrete filtering to identify disease genes and the corresponding causal variants. Results: Among the 720 nonsynonymous single nucleotide polymorphisms (SNPs) shared by two affected members, we found a C to T transition that gives rise to a Thr147Ile missense substitution in the presenilin 1 (PS1) protein. The presence of this same mutation in a French early-onset Alzheimer’s disease family, other affected members of the family, and the predicted high pathogenicity of the substitution strongly suggest that it is the causal variant. In addition to exceptionally young age of onset, we also observed significant limb spasticity and early loss of speech, concurrent with progression of dementia in affected family members. These findings extend the clinical presentation associated with the Thr147Ile variant. Lastly, one member with the Thr147Ile variant was treated with the PKC epsilon activator, bryostatin, in a compassionate use trial after successful FDA review. Initial improvements with this treatment were unexpectedly clear, including return of some speech, increased attentional focus, ability to swallow, and some apparent decrease in limb spasticity. Conclusions: Our findings confirm the role of the PS1 Thr147Ile substitution in Alzheimer’s disease and expand the clinical phenotype to include expressive aphasia and very early onset of dementia

    Life cycle inventory data for the Italian agri-food sector: background, sources and methodological aspects

    Purpose: For the development of any life cycle assessment study, the practitioner frequently integrates primary data collected on-field, with background data taken from various life cycle inventory databases which are part of most commercial LCA software packages. However, such data is often not generally applicable to all product systems since, especially concerning the agri-food sector, available datasets may not be fully representative of the site specificity of the food product under examination. In this context, the present work investigates the background, sources and methodological aspects that characterise the most known commercial databases containing agri-food data, with a focus on four agri-food supply chains (olive oil, wine, wheat products and citrus fruit), which represent an important asset for the Italian food sector. Methods: Specifically, the paper entails a review of currently available LCI databases and their datasets with a twofold scope: firstly, to understand how agri-food data is modelled in these databases for a coherent and consistent representation of regional scenarios and to verify whether they are also suitable for the Italian context and, secondly, to identify and analyse useful and relevant methodological approaches implemented in the existing LCI databases when regional data are modelled. Results: Based on the aforementioned review, it is possible to highlight some problems which may arise when developing an LCI pertaining to the four Italian agri-food supply chains, namely: 1. The need for specific inventory datasets to tackle the specificities of agri-food product systems. 2. The lack of datasets, within the existing DBs, related to the Italian context and to the abovementioned supply chains. In fact, at present, in the currently available LCI DBs, there are very few (or in some cases none) datasets related to Italian wine, olive oil, wheat-based products and citrus fruit. The few available datasets often contain some data related to the Italian context but also approximate data with that of product systems representing other countries. Furthermore, the present study allowed to identify and discuss the main aspects to be used as starting elements for modelling regional data to be included in a future Italian LCI database of the abovementioned four supply chains. Conclusions: The results of the present study represent a starting point for the collection of data and its organisation, in order to develop an Italian LCI agri-food database with datasets which are representative of the regional specificities of four agri-food supply chains which play an important role in the Italian economy

    Genomic Analysis of Demographic History and Ecological Niche Modeling in the Endangered Sumatran Rhinoceros Dicerorhinus sumatrensis

    The vertebrate extinction rate over the past century is approximately 22–100 times greater than background extinction rates [1], and large mammals are particularly at risk [2, 3]. Quaternary megafaunal extinctions have been attributed to climate change [4], overexploitation [5], or a combination of the two [6]. Rhinoceroses (Family: Rhinocerotidae) have a rich fossil history replete with iconic examples of climate-induced extinctions [7], but current pressures threaten to eliminate this group entirely. The Sumatran rhinoceros(Dicerorhinus sumatrensis) is among the most imperiled mammals on earth. The 2011 population was estimated at ≤216 wild individuals [8], and currently the species is extirpated, or nearly so, throughout the majority of its former range [8–12]. Understanding demographic history is important in placing current population status into a broader ecological and evolutionary context. Analysis of the Sumatran rhinoceros genome reveals extreme changes in effective population size throughout the Pleistocene. Population expansion during the early to middle Pleistocene was followed by decline. Ecological niche modeling indicated that changing climate most likely played a role in the decline of the Sumatran rhinoceros, as less suitable habitat on an emergent Sundaland corridor isolated Sumatran rhinoceros populations. By the end of the Pleistocene, the Sundaland corridor was submerged, and populations were fragmented and consequently reduced to low Holocene levels from which they would never recover. Past events denuded the Sumatran rhinoceros of genetic diversity through population decline, fragmentation, or some combination of the two and most likely made the species even more susceptible to later exploitation and habitat loss

    Synthesis, characterization, and crystal structure of Ni(II) and Cu(II) complexes with N-furoyl-N',N'-diethylthiourea: antifungal activity

    The complexes cis-bis(N-furoyl-N′,N′-diethylthioureato-k 2O,S)nickel(II) and cis-bis(N-furoyl-N′,N′-diethylthioureato-k 2O,S)copper(II) were prepared by the reaction of metal acetate with the corresponding acylthiourea derivative. The complexes were characterized by IR, 1H-NMR, 13C-NMR, and single-crystal X-ray diffraction. Both complexes show two furoylthiourea ligands bonded to metal to form a four-coordinate complex with square-planar geometry. The antifungal activity of the prepared complexes was studied against the phytopathogenic fungi Botrytis cinerea and Colletotrichum gloeosporioides, responsible for important plant diseases.CAPES (018/05)CNPq (134576/2007-1)ICTP/CLA
