    The conjunctival fungal microflora of horses in a North Queensland tropical environment and their in vitro susceptibilities to antifungal agents

    Fungi are ubiquitous in the environment and part of the commensal microflora on the conjunctiva of equine eyes. North Queensland, being tropical, presents an ideal environment for fungi growth. When the cornea is injured, fungi can invade the corneal stroma, resulting in keratomycosis. The objectives of this study were to determine the fungal species specific to the eyes of horses in the Townsville region; to investigate the potential risk factors associated with the presence of fungi; and to test their susceptibility to antifungals to create an empirical guide for treatment. The eyes of forty ophthalmologically normal horses from James Cook University were sampled throughout the summer months of December 2017, January 2018, and January and February 2020. Cultured fungi were identified morphologically, and their identity confirmed by comparing partial 18sRNA DNA sequences with the NCBI nucleotide database. Minimum inhibitory concentration testing of common antifungal medications was performed. Sixty-one out of eighty conjunctival samples grew fungi, and 21 different fungi genera were isolated. The most common genera were Aspergillus (18%, 26/141), Curvularia (14%,20/141), Rhodotorula (12%,17/141) and Penicillium (12%,17/141). No significant association was found between age or environmental factors and fungal culture status. Most fungi were highly susceptible to voriconazole and ketoconazole but resistant to fluconazole and amphotericin B. This adds to the body of evidence on which species of fungi are present as normal ocular microflora of horses living in tropical regions of Australia, and an avenue for treating them

    Utilisation and evaluation of cooperative case-based teaching for integration of microbiology and pharmacology in veterinary education

    Purpose: Integrating basic sciences with clinical disciplines while fostering clinical reasoning capabilities is difficult. We investigated the utilisation of diagnostic specimens and, a cooperative, case-based learning and teaching model to integrate principles of antimicrobial drug pharmacology and microbiology in the fifth year of a veterinary course. Methods: In small groups, students were assigned diagnostic specimens from which they isolated and identified clinically relevant microorganisms and then performed antimicrobial susceptibility tests based on a review of pharmacology, microbiology and pathophysiology. Results were recorded and analysed followed by a student-led integrative tutorial. Learning outcomes were assessed via individually written reports discussing the disease process, interpretation of diagnostic results and, recommendations and rationales for therapeutic interventions. Results: This approach yielded high quality student reports that conformed to antimicrobial prescription guidelines with consistently high summative assessment scores. Mean scores for the final report in this learning activity were: 82 ± 12%, 80 ± 12% and 80 ± 11% for 2015, 2016 and 2017 cohorts respectively; over the same time period, 98 ± 1% of students indicated that these learning activities facilitated the development of confidence, professional knowledge and skills. Discussion: This was a consistent approach for integrating principles of veterinary pharmacology and microbiology in clinical disciplines. These data illustrate the benefit of a systematic application of a cooperative, case-based learning and teaching model in integrating pre-clinical and clinical disciplines in a bachelor of veterinary science course

    Food rejection in young children:Validation of the Child Food Rejection Scale in English and cross-cultural examination in the UK and France.

    Although there are numerous studies examining food rejection (neophobia and pickiness), very few studies have directly compared these particular behaviors in children from different countries. Testing children from different countries is important for future research, as there are clear differences between European countries in early feeding practices and child fruit and vegetable consumption. In the present study we aimed to (i) validate the Child Food Rejection Scale in English and (ii) compare children’s food neophobia and pickiness in the UK and France. To that aim, the Child Food Rejection Scale was first translated into English and its reliability and validity were assessed. Second, food rejection scores in a UK sample (n=117) and in a French sample (n=256) were directly compared to examine cross-cultural differences. Results showed that the Child Food Rejection Scale can be successfully used outside France. Moreover results revealed that UK children are less neophobic and picky than French children. These cultural differences can be useful to inform targeted interventions to change food related behaviors in these different populations

    Antibiotic Resistant Bacterial Isolates from Captive Green Turtles and In Vitro

    This study aimed to test multidrug resistant isolates from hospitalised green turtles (Chelonia mydas) and their environment in North Queensland, Australia, for in vitro susceptibility to bacteriophages. Seventy-one Gram-negative bacteria were isolated from green turtle eye swabs and water samples. Broth microdilution tests were used to determine antibiotic susceptibility. All isolates were resistant to at least two antibiotics, with 24% being resistant to seven of the eight antibiotics. Highest resistance rates were detected to enrofloxacin (77%) and ampicillin (69.2%). More than 50% resistance was also found to amoxicillin/clavulanic acid (62.5%), ceftiofur (53.8%), and erythromycin (53.3%). All the enriched phage filtrate mixtures resulted in the lysis of one or more of the multidrug resistant bacteria, including Vibrio harveyi and V. parahaemolyticus. These results indicate that antibiotic resistance is common in Gram-negative bacteria isolated from hospitalised sea turtles and their marine environment in North Queensland, supporting global concern over the rapid evolution of multidrug resistant genes in the environment. Using virulent bacteriophages as antibiotic alternatives would not only be beneficial to turtle health but also prevent further addition of multidrug resistant genes to coastal waters

    Numerous studies have shown that the use of space resources to manufacture propellant and consumables can significantly reduce the launch mass of space exploration beyond earth orbit. Even the Moon, which has no atmosphere, is ricb in resources that can theoretically be harvested. A series of lunar missions over the last 20 years has shown an unexpected resource on the Moon. There is evidence that water ice and other volatiles useful for the production of propellants are located at the lunar poles, though most of it is located within permanently shadowed craters where accessing these resources is challenging

    Complementation of a pathogenic IFNGR2 misfolding mutation with modifiers of N-glycosylation

    Germline mutations may cause human disease by various mechanisms. Missense and other in-frame mutations may be deleterious because the mutant proteins are not correctly targeted, do not function correctly, or both. We studied a child with mycobacterial disease caused by homozygosity for a novel in-frame microinsertion in IFNGR2. In cells transfected with the mutant allele, most of the interferon γ receptor 2 (IFN-γR2) protein was retained within the cell, and that expressed on the cell surface had an abnormally high molecular weight (MW). The misfolding mutation was not gain-of-glycosylation, as it created no new N-glycosylation site. The mutant IFNGR2 allele was null, as the patient's cells did not respond to IFN-γ. Based on the well-established relationship between protein N-glycosylation and protein quality control processes, we tested 29 compounds affecting maturation by N-glycosylation in the secretory pathway. Remarkably, up to 13 of these compounds reduced the MW of surface-expressed mutant IFN-γR2 molecules and restored cellular responsiveness to IFN-γ. Modifiers of N-glycosylation may therefore complement human cells carrying in-frame and misfolding, but not necessarily gain-of-glycosylation, mutations in genes encoding proteins subject to trafficking via the secretory pathway. Some of these compounds are available for clinical use, paving the way for clinical trials of chemical complementation for various human genetic traits

    Near Real Time Prospecting for Lunar Volatiles: Demonstrating RESOLVE Science in the Field

    The Regolith and Environment Science and Oxygen & Lunar Volatile Extraction (RESOLVE) project aims to demonstrate the utility of "in situ resource utilization". In situ resource utilization (ISRU) is a way to rebalance the economics of spaceflight by reducing or eliminating materials that must be brought up from Earth and placed on the surface of the Moon for human use. RESOLVE is developing a rover-borne payload that (1) can locate near subsurface volatiles, (2) excavate and analyze samples of the volatile-bearing regolith, and (3) demonstrate the form, extractability and usefulness of the materials. Such investigations are important not only for ISRU but are also critically important for understanding the scientific nature of these intriguing lunar polar volatile deposits. Temperature models and orbital data suggest near surface volatile concentrations may exist at briefly lit lunar polar locations outside persistently shadowed regions. A lunar rover could be remotely operated at some of these locations for the 4-7 days of expected sunlight at relatively low cost. In July 2012 the RESOLVE project conducted a full-scale field demonstration. In particular, the ability to perform the real-time measurement analysis necessary to search for volatiles and the ability to combine the various measurement techniques to meet the mission measurement and science goals. With help from the Pacific International Space Center for Exploration Systems (PISCES), a lunar rover prototype (provided by the Canadian Space Agency) was equipped with prospecting instruments (neutron spectrometer and near-infrared spectrometer), subsurface access and sampling tools, including both an auger and coring drill (provided by CSA) and subsurface sample analysis instrumentation, including a sample oven system, the Oxygen and Volatile Extraction Node (OVEN), and Gas Chromatograph / Mass Spectrometer system, the Lunar Advanced Volatile Analysis (LAVA) system. Given the relatively short time period this lunar mission is being designed to, prospecting needs to occur in near real-time. The two prospecting instruments are the neutron and NIR spectrometers. In the field demo a small radioactive source was provided the neutron flux. The NIR spectrometer, which includes its own light source, looks at surface reflectance for signatures of bound H20/0H and general mineralogy. Once a "hot spot" was found by the prospecting instruments, the drill could either auger or core. The auger drill worked to a depth of 50 cm and is monitored with a drill camera and the NIR spectrometer. As cuttings are brought up the NIR spectra is monitored. If a particular location is considered of high -interest then the decision to core could be made. The coring drill (a push-tube) allowed a meter sample to be acquired processed by the OVEN/LAVA sys-tem. This presentation will provide details as how these instruments worked together and how and if the planned measurements and science was obtained
