    An Ultrasoft X-ray Flare from 3XMM J152130.7+074916: a Tidal Disruption Event Candidate

    We report on the discovery of an ultrasoft X-ray transient source, 3XMM J152130.7+074916. It was serendipitously detected in an XMM-Newton observation on 2000 August 23, and its location is consistent with the center of the galaxy SDSS J152130.72+074916.5 (z=0.17901 and d_L=866 Mpc). The high-quality X-ray spectrum can be fitted with a thermal disk with an apparent inner disk temperature of 0.17 keV and a rest-frame 0.24-11.8 keV unabsorbed luminosity of ~5e43 erg/s, subject to a fast-moving warm absorber. Short-term variability was also clearly observed, with the spectrum being softer at lower flux. The source was covered but not detected in a Chandra observation on 2000 April 3, a Swift observation on 2005 September 10, and a second XMM-Newton observation on 2014 January 19, implying a large variability (>260) of the X-ray flux. The optical spectrum of the candidate host galaxy, taken ~11 yrs after the XMM-Newton detection, shows no sign of nuclear activity. This, combined with its transient and ultrasoft properties, leads us to explain the source as tidal disruption of a star by the supermassive black hole in the galactic center. We attribute the fast-moving warm absorber detected in the first XMM-Newton observation to the super-Eddington outflow associated with the event and the short-term variability to a disk instability that caused fast change of the inner disk radius at a constant mass accretion rate.Comment: 9 pages, 5 figures.     Bio-based pH-responsive superabsorbent polymers for self-healing cracks in concrete

    Cracks endanger the durability of concrete. Introducing a superabsorbent polymer (SAP) during concrete mixing can create a self-sealing and -healing construction. SAPs are able to take up aqueous solutions up to several hundred times their own weight. Bio-based SAPs starting from polysaccharides have gained increasing interest in recent years due to their biocompatibility, non-toxicity and low price. The use of pH-responsive SAPs can also be extremely useful as they should only swell more upon crack formation and less during mixing of the SAPs in the concrete. The present work describes the development and the characterization of SAPs based on methacrylated polysaccharides (alginate and chitosan) combined with pH-responsive monomers dimethylaminoethyl methacrylate (DMAEMA) and dimethylaminopropyl methacrylamide (DMAPMA). The materials exhibited a high moisture uptake capacity up to 120% of their original weight with a negligible hysteresis. The pHresponsive swelling behavior was studied in aqueous and cement filtrate solutions with a varying pH. Chitosan combined with DMAEMA or DMAPMA showed the targeted pHresponsive swelling. Chitosan combined with DMAPMA also showed a limited compression strength reduction and a promising self-sealing and -healing behavior and could thus be considered as a very interesting future solution to seal and heal cracks in concrete

    Persistent Coxiella burnetii Infection in Mice Overexpressing IL-10: An Efficient Model for Chronic Q Fever Pathogenesis

    Interleukin (IL)-10 increases host susceptibility to microorganisms and is involved in intracellular persistence of bacterial pathogens. IL-10 is associated with chronic Q fever, an infectious disease due to the intracellular bacterium Coxiella burnetii. Nevertheless, accurate animal models of chronic C. burnetii infection are lacking. Transgenic mice constitutively expressing IL-10 in macrophages were infected with C. burnetti by intraperitoneal and intratracheal routes and infection was analyzed through real-time PCR and antibody production. Transgenic mice exhibited sustained tissue infection and strong antibody response in contrast to wild-type mice; thus, bacterial persistence was IL-10-dependent as in chronic Q fever. The number of granulomas was low in spleen and liver of transgenic mice infected through the intraperitoneal route, as in patients with chronic Q fever. Macrophages from transgenic mice were unable to kill C. burnetii. C. burnetii–stimulated macrophages were characterized by non-microbicidal transcriptional program consisting of increased expression of arginase-1, mannose receptor, and Ym1/2, in contrast to wild-type macrophages in which expression of inducible NO synthase and inflammatory cytokines was increased. In vivo results emphasized macrophage data. In spleen and liver of transgenic mice infected with C. burnetii by the intraperitoneal route, the expression of arginase-1 was increased while microbicidal pathway consisting of IL-12p40, IL-23p19, and inducible NO synthase was depressed. The overexpression of IL-10 in macrophages prevents anti-infectious competence of host, including the ability to mount granulomatous response and microbicidal pathway in tissues. To our knowledge, this is the first efficient model for chronic Q fever pathogenesis

    Analysis of a key regulatory region upstream of the Myf5 gene reveals multiple phases of myogenesis, orchestrated at each site by a combination of elements dispersed throughout the locus

    Myf5 is the first myogenic regulatory factor to be expressed in the mouse embryo and it determines the entry of cells into the skeletal muscle programme. A region situated between -58 kb and -48 kb from the gene directs Myf5 transcription at sites where muscles will form. We now show that this region consists of a number of distinct regulatory elements that specifically target sites of myogenesis in the somite, limbs and hypoglossal cord, and also sites of Myf5 transcription in the central nervous system. Deletion of these sequences in the context of the locus shows that elements within the region are essential, and also reveals the combinatorial complexity of the transcriptional regulation of Myf5. Both within the -58 kb to -48 kb region and elsewhere in the locus, multiple sequences are present that direct transcription in subdomains of a single site during development, thus revealing distinct phases of myogenesis when subpopulations of progenitor cells enter the programme of skeletal muscle differentiation.This work in M.B.'s laboratory was supported by the Pasteur Institute and the CNRS and by grants from the ACI Integrative Biology Programme of the MJER, the AFM and the European Community (QLK3-CT-99/02).     A Study of the Diverse T Dwarf Population Revealed by WISE

    We report the discovery of 87 new T dwarfs uncovered with the Wide-field Infrared Survey Explorer (WISE) and three brown dwarfs with extremely red near-infrared colors that exhibit characteristics of both L and T dwarfs. Two of the new T dwarfs are likely binaries with L7+/-1 primaries and mid-type T secondaries. In addition, our follow-up program has confirmed 10 previously identified T dwarfs and four photometrically-selected L and T dwarf candidates in the literature. This sample, along with the previous WISE discoveries, triples the number of known brown dwarfs with spectral types later than T5. Using the WISE All-Sky Source Catalog we present updated color-color and color-type diagrams for all the WISE-discovered T and Y dwarfs. Near-infrared spectra of the new discoveries are presented, along with spectral classifications. To accommodate later T dwarfs we have modified the integrated flux method of determining spectral indices to instead use the median flux. Furthermore, a newly defined J-narrow index differentiates the early-type Y dwarfs from late-type T dwarfs based on the J-band continuum slope. The K/J indices for this expanded sample show that 32% of late-type T dwarfs have suppressed K-band flux and are blue relative to the spectral standards, while only 11% are redder than the standards. Comparison of the Y/J and K/J index to models suggests diverse atmospheric conditions and supports the possible re-emergence of clouds after the L/T transition. We also discuss peculiar brown dwarfs and candidates that were found not to be substellar, including two Young Stellar Objects and two Active Galactic Nuclei.     Double-outlet right ventricle and double septal defects in a Rhesus macaque (Macaca mulatta)

    Double-outlet right ventricle (DORV) is a rare form of congenital heart defect, in which both great arteries (aorta and pulmonary trunk) arise from the right ventricle. The defect affects 1-1.5% of patients with congenital heart disease with a frequency of 1 in 10,000 live births in human beings. A 6-year-old male India-origin Rhesus macaque (Macaca mulatta) with no history of illness or trauma presented for project pre-assignment physical examination from the outdoor breeding colony at the Tulane National Primate Research Center (Covington, Louisiana). Upon physical examination, the animal was found to have poor body condition with a body condition score of 1 out of 5. Thoracic auscultation revealed caudal displacement of the heart with a grade VI/VI pansystolic murmur bilaterally. Thoracic radiographs showed a globoid heart with significant enlargement of the left atrium, left auricle, and both ventricles. The aortic shadow was not prominent, but the pulmonary artery trunk was enlarged ( Grossly, the animal was severely emaciated and dehydrated with diffuse muscular atrophy. The skin and external mucosa were moderately cyanotic. The heart was markedly enlarged and globoid with severe dilation of the left atrium and auricle and marked enlargement of both ventricles.
