    Fadolmidine – Favourable adverse effects profile for spinal analgesia suggested by in vitro and in vivo models

    Fadolmidine is an α2-adrenoceptor full agonist developed for spinal analgesia with a local mode of action. The purpose of this study was to demonstrate the safety of fadolmidine on known α2-adrenoceptor-related effects: kidney function, urodynamics and cardiovascular variables. Furthermore, the binding affinity of fadolmidine for the 5-HT3 receptor prompted functional studies on 5-HT3. According to the binding affinity data, fadolmidine demonstrated partial agonism on the 5-HT3 receptor in transfected cells and in guinea pig ileum preparation. However, intravenous (IV) fadolmidine did not produce any 5-HT3-related hemodynamic effects in anaesthetised rats. In urodynamic studies, intrathecal (IT) fadolmidine interrupted volume-evoked voiding cycles and induced overflow incontinence at high concentrations in anaesthetised rats; however, at the analgesic dose range, the effects were mild. The effects of fadolmidine on kidney function were studied in conscious rats after IV and IT dosing. While IT fadolmidine increased dose-dependent urine output, sodium ion concentration, IV doses increased only sodium ion concentration The effects of IT fadolmidine on heart rate (HR), mean arterial pressure (MAP) and sedation were evaluated in the home cage and in the open field using a telemetry system. In resting conditions, fadolmidine decreased HR dose-dependently and increased initial MAP, whereas in actively moving rats, there were no effects at analgesic doses. The results suggest that at anticipated analgesic clinical doses, IT fadolmidine provides analgesia without significant adverse effects on sedation, MAP or HR and with only modest effects on kidney function and urodynamics.</p

    The involvement of autistic children in early childhood education

    Research on the involvement of autistic children in daily activities in inclusive early childhood education is scarce. In Finland, all children, including autistic children, under the age of seven (before basic education) are entitled to participate in early childhood education and care. Children also attend compulsory, free-of-charge pre-primary education during the year before their basic education begins. Furthermore, attending early childhood education and care is not dependent whether a child requires day care because of their parents’ work. Autistic children attend early childhood education in inclusive day care centres. Thus, in this study, we examined the involvement of autistic children by focusing on the objects of their attention during daily activities in inclusive day care centres in Finland. The data were collected between 2017 and 2020, during the research and development project Progressive Feedback, in which children were observed using systematic sampling. The study material consisted of observations of seven autistic children as part of a larger sample of children. The data were analysed using statistical methods. The results indicated that, during their deepest involvement, autistic children expressed positive emotions regarding participation and collaborated with and directed their focus towards other children. Moreover, autistic children demonstrated the most intensive involvement during adult-supported play.publishedVersionPeer reviewe

    Tracing Grog and Pots to Reveal Neolithic Corded Ware Culture Contacts in the Baltic Sea Region (SEM-EDS, PIXE)

    The Neolithic Corded Ware Culture (CWC) complex spread across the Baltic Sea region ca. 2900/2800-2300/2000 BCE. Whether this cultural adaptation was driven by migration or diffusion remains widely debated. To gather evidence for contact and movement in the CWC material culture, grog-tempered CWC pots from 24 archaeological sites in southern Baltoscandia (Estonia and the southern regions of Finland and Sweden) were sampled for geochemical and micro-structural analyses. Scanning electron microscopy with energy dispersive spectrometry (SEM-EDS) and particle-induced X-ray emission (PIXE) were used for geochemical discrimination of the ceramic fabrics to identify regional CWC pottery manufacturing traditions and ceramic exchange. Major and minor element concentrations in the ceramic body matrices of 163 individual vessels and grog temper (crushed pottery) present in the ceramic fabrics were measured by SEM-EDS. Furthermore, the high-sensitivity PIXE technique was applied for group confirmation. The combined pot and grog matrix data reveal eight geochemical clusters. At least five geochemical groups appeared to be associated with specific find locations and regional manufacturing traditions. The results indicated complex inter-site and cross-Baltic Sea pottery exchange patterns, which became more defined through the grog data, i.e., the previous generations of pots. The CWC pottery exhibited high technological standards at these latitudes, which, together with the identified exchange patterns and the existing evidence of mobility based on human remains elsewhere in the CWC complex, is indicative of the relocation of skilled potters, possibly through exogamy.     InDEx – Industrial Data Excellence

    InDEx, the Industrial Data Excellence program, was created to investigate what industrial data can be collected, shared, and utilized for new intelligent services in high-performing, reliable and secure ways, and how to accomplish that in practice in the Finnish manufacturing industry.InDEx produced several insights into data in an industrial environment, collecting data, sharing data in the value chain and in the factory environment, and utilizing and manipulating data with artificial intelligence. Data has an important role in the future in an industrial context, but data sources and utilization mechanisms are more diverse than in cases related to consumer data. Experiences in the InDEx cases showed that there is great potential in data utili zation.Currently, successful business cases built on data sharing are either company-internal or utilize an existing value chain. The data market has not yet matured, and third-party offerings based on public and private data sources are rare. In this program, we tried out a framework that aimed to securely and in a controlled manner share data between organizations. We also worked to improve the contractual framework needed to support new business based on shared data, and we conducted a study of applicable business models. Based on this, we searched for new data-based opportunities within the project consortium. The vision of data as a tradeable good or of sharing with external partners is still to come true, but we believe that we have taken steps in the right direction.The program started in fall 2019 and ended in April 2022. The program faced restrictions caused by COVID-19, which had an effect on the intensity of the work during 2020 and 2021, and the program was extended by one year. Because of meeting restrictions, InDEx collaboration was realized through online meetings. We learned to work and collaborate using digital tools and environments. Despite the mentioned hindrances, and thanks to Business Finland’s flexibility, the extension time made it possible for most of the planned goals to be achieved.This report gives insights in the outcomes of the companies’ work within the InDEx program. DIMECC InDEx is the first finalized program by the members of the Finnish Advanced Manufacturing Network (FAMN, www.famn.fi).</p
