    Diagnosing Mild TBIs with Fluid Biomarkers

    Upon head trauma, neurons within the brain can become stretched and injured. Continuous mild traumatic brain injuries (mTBIs) without proper healing can lead to chronic neurological symptoms. This is especially problematic in contact sports and military personnel. The purpose of this literature review is to take an in depth look at fluid biomarkers that could aid in the diagnosis of a concussion at a point of injury. Databases were used including PubMed, Clinical Key, Embase and Access Medicine. The studies chosen were published within the last 7 years, applicable to the topic and absent of pronounced bias. The 19 works include clinical trials, randomized control trials, and systematic reviews. There proved to be more research in blood biomarkers than salivary biomarkers, but the evidence is still lacking with both in relation to mTBIs. GFAP and UCHL1 show to be sufficient serum markers for individuals who have a significant concussion history or recurrent head trauma. Additionally, continued research on saliva miRNAs could prove them to be a diagnostic tool for quick, point of injury tests. The data shows that serum proteins respond in a much more delayed response, whereas saliva miRNA biomarkers respond more acutely. Additional studies will need to be done to properly examine both genders, other contact activities, and the polytrauma associated with military personnel

    Individual Differences in Well-Being in Older Breast Cancer Survivors

    Older women who survive breast cancer may differ significantly in their long-term well-being. Using a risk and protective factors model, we studied predictors of well-being in 127 women age 70 and above with a history of at least one year\u27s survival of breast cancer. Mean post-cancer survivorship was 5.1 years. Using life satisfaction, depression and general health perceptions as outcome variables, we assessed whether demographic variables, cancer-related variables, health status and psychosocial resources predicted variability in well-being using correlational and hierarchical regression analyses. Higher age predicted increased depression but was not associated with life satisfaction or general health perceptions. Cancer-related variables, including duration of survival, and type of cancer treatment, were not significantly associated with survivors\u27 well-being. Poorer health status was associated with poorer well-being in all three dependent variables. After controlling for demographics, cancer-related variables, and health status, higher levels of psychosocial resources including optimism, mastery, spirituality and social support predicted better outcome in all three dependent variables. While many older women survive breast cancer without severe sequelae, there is considerable variability in their well-being after survivorship. Successful intervention with older breast cancer survivors might include greater attention not only to cancer-specific concerns, but also attention to geriatric syndromes and functional impairment, and enhancement of protective psychosocial resources

    Photoacclimation by arctic cryoconite phototrophs

    © FEMS 2017. All rights reserved. Cryoconite is a matrix of sediment, biogenic polymer and a microbial community that resides on glacier surfaces. The phototrophic component of this community is well adapted to this extreme environment, including high light stress. Photoacclimation of the cryoconite phototrophic community on Longyearbreen, Svalbard, was investigated using in situ variable chlorophyll fluorescence. Rapid light curves (RLCs) and induction-recovery curves were used to analyse photosystem II quantum efficiency, relative electron transport rate and forms of downregulation including non-photochemical quenching (NPQ) and state transitions in cyanobacteria. Phototrophs used a combination of behavioural and physiological photochemical downregulation. Behavioural downregulation is hypothesised to incorporate chloroplast movement and cell or filament positioning within the sediment matrix in order to shade from high light, which resulted in a lack of saturation of RLCs and hence overestimation of productivity. Physiological downregulation likely consisted of biphasic NPQ, comprising a steadily induced light-dependent form and a light-independent NPQ that was not reversed with decreasing light intensity. State transitions by cyanobacteria were the most likely physiological downregulation employed by cyanobacteria within the mixed phototroph community. These findings demonstrate that cryoconite phototrophs combine multiple forms of physiological and behavioural downregulation to optimise light exposure and maximise photosynthetic productivity. This plasticity of photoacclimation enables them to survive productively in the high-light stress environment on the ice surface

    Differentiation of columnar epithelial cells involves a dramatic reorganization of the microtubules (MTs) and centrosomal components into an apico-basal array no longer anchored at the centrosome. Instead, the minus-ends of the MTs become anchored at apical non-centrosomal microtubule organizing centres (n-MTOCs). Formation of n-MTOCs is critical as they determine the spatial organization of MTs, which in turn influences cell shape and function. However, how they are formed is poorly understood. We have previously shown that the centrosomal anchoring protein ninein is released from the centrosome, moves in a microtubule-dependent manner and accumulates at n-MTOCs during epithelial differentiation. Here, we report using depletion and knockout (KO) approaches that ninein expression is essential for apico-basal array formation and epithelial elongation and that CLIP-170 is required for its redeployment to n-MTOCs. Functional inhibition also revealed that IQGAP1 and active Rac1 coordinate with CLIP-170 to facilitate microtubule plus-end cortical targeting and ninein redeployment. Intestinal tissue and in vitro organoids from the Clip1/Clip2 double KO mouse with deletions in the genes encoding CLIP-170 and CLIP-115, respectively, confirmed requirement of CLIP-170 for ninein recruitment to n-MTOCs, with possible compensation by other anchoring factors such as p150Glued and CAMSAP2 ensuring apico-basal microtubule formation despite loss of ninein at n-MTOCs

    Temporal patterns of wildlife roadkill in the UK

    Wildlife-vehicle collisions are one of the main causes of mortality for wild mammals and birds in the UK. Here, using a dataset of 54,000+ records collated by a citizen science roadkill recording scheme between 2014–2019, we analyse and present temporal patterns of wildlife roadkill of the 19 most commonly reported taxa in the UK (84% of all reported roadkill). Most taxa (13 out of 19) showed significant and consistent seasonal variations in road mortality and fitted one of two seasonal patterns; bimodal or unimodal: only three species (red fox Vulpes vulpes, European polecat Mustela putorius and Reeves’ muntjac deer Muntiacus reevesi) showed no significant seasonality. Species that increase movement in spring and autumn potentially have bimodal patterns in roadkill due to the increase in mate-searching and juvenile dispersal during these respective time periods (e.g. European badger Meles meles). Unimodal patterns likely represent increased mortality due to a single short pulse in activity associated with breeding (e.g. birds) or foraging (e.g. grey squirrels Sciurus carolinensis in autumn). Importantly, these patterns also indicate periods of increased risk for drivers, potentially posing a greater threat to human welfare. In addition to behaviour-driven annual patterns, abiotic factors (temperature and rainfall) explained some variance in roadkill. Notably, high rainfall was associated with decreased observations of two bird taxa (gulls and Eurasian magpies Pica pica) and European rabbit Oryctolagus cuniculus. By quantifying seasonal patterns in roadkill, we highlight a significant anthropogenic impact on wild species, which is important in relation to conservation, animal welfare, and human safety

    Roadkill scavenging behaviour in an urban environment

    Roads can have negative impacts on wildlife through indirect effects such as fragmentation of habitat, or through direct effects such as fatal collisions with vehicles. Wildlife deaths on British roads number in the millions per year, so the resulting carcasses represent a substantial carrion biomass available as food for scavengers. By removing roadkill in urban areas, scavengers perform a valuable ecosystem service, but the rapid removal of these carcasses by scavengers could bias estimates of the impacts of roads on wildlife. In order to evaluate the scale and context of urban roadkill scavenging, we examined: (i) which species scavenge on roadkill in urban areas, (ii) the likelihood of roadkill being removed by scavengers, and (iii) whether spatial and temporal factors (habitat type and time of day) influenced the rate of removal. Camera traps baited with chicken heads as simulated ‘roadkill corpses’ were deployed in six residential and six parkland sites in the city of Cardiff, UK. Seven species were observed removing the roadkill, with corvids being the most common scavengers, responsible for 42% of removals. Of the 120 corpses, 90 (76%) were removed within 12 h. Time of day had a significant effect on the rate of removal, with the number of carcass removals peaking in the first few hours of daylight. Of roadkill placed at 9 am, 62% of carcasses had been removed after only 2 h. Removal of corpses by scavengers could mean that the actual number of road deaths is six times more than that observed during surveys

    Adverse Childhood Experiences and Adult Cardiometabolic Risk Factors and Disease Outcomes: Cross-Sectional, Populationbased Study of Adults in Rural Uganda

    Background: Cardiovascular diseases (CVD) pose a major threat to public health in sub-Saharan African communities, where the burden of these classes of illnesses is expected to double by 2030. Growing research suggests that past developmental experiences and early life conditions may also elevate CVD risk throughout the life course. Greater childhood stress and adversity are consistently associated with a range of adult CVDs and associated risk factors, yet little research exists on the long-term effects of early life stress on adult physical health outcomes, especially CVD risk, in sub-Saharan African contexts. This study aims to evaluate the associations between adverse childhood experiences and adult cardiometabolic risk factors and health outcomes in a population-based study of adults living in Mbarara, a rural region of southwestern Uganda. Methods Data come from an ongoing, whole-population social network cohort study of adults living in the eight villages of Nyakabare Parish, Mbarara. A modified version of the Adverse Childhood Experiences-International Questionnaire (ACEs) assessed past exposure to physical, emotional, and sexual adversity. Participants also took part in a health fair where medical histories on cardiometabolic risk factors and cardiovascular diseases were gathered. Multiple logistic regression models estimated the associations between ACEs and cardiometabolic risk factors and health outcomes. Results Data were available on 545 adults. The average number of ACEs was 4.9 out of a possible 16. The cumulative number of ACEs were associated with having a history of heart attack and/or heart failure (adjusted odds ratio (AOR) = 1.11, 95% confidence interval (CI) = 0.999-1.234, P = 0.051), but the estimated association was not statistically significant. ACEs did not have statistically significant associations with any others measures of adult cardiometabolic risk and CVD. Conclusions: Adverse childhood experiences are not associated with a range of adult cardiometabolic risk factors and health outcomes in this sample of rural Ugandan adults. Further research in this sample is necessary to identify the pathways that may motivate these null relationship and possibly protect against adverse cardiometabolic and cardiovascular health outcomes

    Supermassive black holes at high redshifts

    MeV blazars are the most luminous persistent sources in the Universe and emit most of their energy in the MeV band. These objects display very large jet powers and accretion luminosities and are known to host black holes with a mass often exceeding 109M10^9 M_{\odot}. An MeV survey, performed by a new generation MeV telescope which will bridge the entire energy and sensitivity gap between the current generation of hard X-ray and gamma-ray instruments, will detect >>1000 MeV blazars up to a redshift of z=56z=5-6. Here we show that this would allow us: 1) to probe the formation and growth mechanisms of supermassive black holes at high redshifts, 2) to pinpoint the location of the emission region in powerful blazars, 3) to determine how accretion and black hole spin interplay to power the jet.Comment: 7 pages, 4 figure.     “Community members have more impact on their neighbors than celebrities”: Leveraging community partnerships to build COVID-19 vaccine confidence

    Vaccines are a strong public health tool to protect against severe disease, hospitalization, and death from COVID-19. Still, inequities in COVID-19 vaccination rates and health outcomes continue to exist among Black and Latino populations. Boston Medical Center (BMC) has played a significant role in vaccinating medically underserved populations, and organized a series of community-engaged conversations to better understand community concerns regarding the COVID-19 vaccine. We accessed and analyzed nine publicly available recordings of the community-engaged conversations which were held between Mar 2021-Sep 2021 (n=8-122 attendees). We employed a Consolidated Framework for Implementation Research-driven codebook to code our data and utilized a modified version of qualitative rapid analysis methods. Five main themes emerged: (1) Structural factors are important barriers to COVID-19 vaccination; (2) Mistrust exists due to the negative impact of systemic oppression and perceived motivation of the government; (3) There is a desire to learn more about biological and clinical characteristics of the COVID-19 vaccine as well as the practical implications of being vaccinated; (4) Community engagement is important for delivering COVID-19 information and education and; (5) Community leaders believe that the COVID-19 vaccine is a solution to address the pandemic. In highlighting the themes which resulted from these community-engaged conversations, this study illustrates a need for community-engaged COVID-19 vaccine messaging which reflects the nuances of the COVID-19 vaccine and pandemic without oversimplifying information and underscores important considerations for public health and healthcare leadership in the development of initiatives which work to advance health equity
