Fictive interaction in blended networks in the daily show with Jon Stewart: conceptualizing politican humor discourse not only for entertaining purpouses
Cryo-EM reveals the conformation of a substrate analogue in the human 20S proteasome core
The proteasome is a highly regulated protease complex fundamental for cell homeostasis and controlled cell cycle progression. It functions by removing a wide range of specifically tagged proteins, including key cellular regulators. Here we present the structure of the human 20S proteasome core bound to a substrate analogue inhibitor molecule, determined by electron cryo-microscopy (cryo-EM) and single-particle analysis at a resolution of around 3.5 Å. Our map allows the building of protein coordinates as well as defining the location and conformation of the inhibitor at the different active sites. These results open new prospects to tackle the proteasome functional mechanisms. Moreover, they also further demonstrate that cryo-EM is emerging as a realistic approach for general structural studies of protein–ligand interactions
Characterization of fully recombinant human 20S and 20S-PA200 proteasome complexes
Proteasomes are essential in all eukaryotic cells. However, their function and regulation remain considerably elusive, particularly those of less abundant variants. We demonstrate the human 20S proteasome recombinant assembly and confirmed the recombinant complex integrity biochemically and with a 2.6 Å resolution cryo-EM map. To assess its competence to form higher-order assemblies, we prepared and analyzed recombinant human 20S-PA200, a poorly characterized nuclear complex. Its 3.0 Å resolution cryo-EM structure reveals the PA200 unique architecture; the details of its intricate interactions with the proteasome, resulting in unparalleled proteasome α ring rearrangements; and the molecular basis for PA200 allosteric modulation of the proteasome active sites. Non-protein cryo-EM densities could be assigned to PA200-bound inositol phosphates, and we speculate regarding their functional role. Here we open extensive opportunities to study the fundamental properties of the diverse and distinct eukaryotic proteasome variants and to improve proteasome targeting under different therapeutic conditions
Associations between character strengths and life satisfaction: a study with college students
The aim of the study was to identify the relationship between character strengths and life satisfaction and the mean differences between participants’ gender and age. The sample consisted of 186 college students, 62.9% women, aged 17 to 45 years (M=21.55; SD=4.79), from two Brazilian states. The instruments used were the Character Strengths Scale and the Life Satisfaction Scale. Results showed significant differences for gender, and women obtained the highest scores in four character strengths. The age of participants did not show significant differences. Regarding the correlations among the constructs studied results showed that vitality, gratitude, hope, perseverance and love were the strengths presenting the highest correlation with life satisfaction, in moderate magnitudes. Findings indicated that individuals with higher scores in hope, vitality, gratitude, love, curiosity, perseverance and social intelligence strengths, tend to live a more satisfying life.
There is growing recognition within local and regional studies regarding the importance of institutions. A ‘soft institutionalism’ predominates, which is criticised here for leaving out the explicit role of the State and for having a bias towards synergies and positive externalities. We seek to contribute to an institutional political economic theory of local development. We bring together social and political conceptions of ‘old institutional economics’ and evolutionary economics, and the State is explicitly brought back into the analysis. The construction of new institutions is explored as a path dependent process in which institutions (re)shape the development path of an area in a particular direction. Using different kinds of power resources, different social groups struggle for control. We use the framework to analyze the divergent stories of two cases of local development in Brazil based on tourism. Prainha do Canto Verde developed community-based tourism, while in Jericoacoara local development culminated in mass tourism
A inovação é, nasua essência, complexa e multifactorial.
complexidade do tema.
Innovation has ever been an important lever for organizations to make the difference and even
to survive. Innovation is complex and multifactorial. In organizational context it has to do with
decisions, actions, organizational structure, processes and metrics used to promote, develop
and capture initiatives aligned with strategy and aiming to create value.
Looking into the organizational culture of the greatest portuguese energy company, this work
intends to be a contribution to the comprehension and understanding of the theme. Following
Schein’s model, the purpose is to identify values, artifacts and basic assumptions, trying to find
potential relationships and impacts on the innovation processes within the company. On the
other hand, exploring the archetypes of radical andincremental innovation – according to
Mclaughin, et al. (2005) - EDP organizational culture fits well in the model.
This is an exploratory, qualitative case study. Thefindings show that the combination of
different organizational forms, values and basic assumptions, can contribute to create a willing
environment to innovation. This study still shows that concerning analysis and policies, it is
important to consider innovation according to its degree of novelty, due to its complex nature.
The study shows evidence that the EDP organizational culture fits essentially in an incremental
model in which the focus is “doing better”, although some characteristics that facilitate radical
innovation have been found in the strategic dimensi ons of development and knowledge, and
external adaptation.
The results open some routes for future works. No way lessening the replication in other
Portuguese companies, it would be interesting to deepen the study of those with multiregional
experiences, carrying out longitudinal studies to evaluate how Portuguese culture impact
organizational culture
O comportamento de consumo e a ansiedade do cliente do El Corte Inglés Portugal no e ‐ Commerce
Project Work presented as the partial requirement for obtaining a Master's degree in Information Management, specialization in Marketing IntelligenceA forma como os consumidores compram está a evoluir a cada dia e, consequentemente, o modelo de
The consumers shopping way is evolving every day and, consequently, the retail model is adapting to
meet the needs and behavior of the online consumer, which has done companies such as the giant El
Corte Inglés, with two stores in Portugal, to promote continuous improvements in their
communication in this virtual medium. However, as technology has greatly assisted online shopping,
some problems have arisen, most of them related to customer anxiety during the purchase process,
caused by many reasons. This feeling can harm the company, leading the consumer to give up on the
purchase. Thus, the objective of this thesis was to analyze what makes the customer of the company
El Corte Inglés Portugal anxious during the pre-purchase, during-purchase and post-purchase process
and how it is possible to minimize this feeling of anxiety when purchasing online. Therefore, the
methodology used in this work was exploratory and bibliographical research. Through a case study
that sought patterns, ideas or hypotheses for the issue raised, 600 audios of calls made by customers
of the company El Corte Inglés Portugal were heard. We noticed that women are the ones who call the
most, with a 59.5% incidence, the main reasons for these calls being the need for exchanges, returns
and cancellations. Men call, most of the time, to check the delivery date. The during-purchase process
is the most representative of the occurrence of calls, with 45.83% (275 calls out of 600) of incidence.
The main reasons for the calls, in general, inquired about product stock, with 18.67% of the
occurrences; delivery date or order status, with 18%; and, finally, 17.67% related to aid in general. We
conclude that consumer anxiety is higher (High Anxiety) in the during-purchase phase, as stated, with
13.82%, while in other stages the high anxiety level appears with 12.87% for post-purchase, while 4
.55% are in the pre-purchase phase. Thus, it is visible that the anxiety that permeates an online
purchase is harmful to the company, as it generates discomfort, friction, insecurity, doubts when
making the purchase and even giving up, either for lack of information on the site, or for lack of product
stock, either by delay in delivery of the product, or even by error in delivery. The study pointed out
that this feeling of anxiety that plagues most of ECI's online customers can be minimized by adopting
strategies aimed at improving systems and providing more complete information within the online
sales environment, in order to satisfy and preserve the customer, in addition to providing the constant
growth of e-commerce in the field in question
Trabalhadores cuidadores: o novo paradigma familiar no contexto laboral
This study aims to arouse attention to a new labour phenomenon that is closely related to the current process of population aging and to gender stereotypes that still linger in today’s society: the situation of workers who are also caregivers of dependent relatives. These workers, mostly women, face numerous daily challenges. This is enhanced by a lack in what concerns employment protection laws, as the existing legal status of workers, who obviously have a family life, does not take into account their responsibilities towards their ascendant and collateral relatives.
Society continues to assign the families with the task of taking care of dependent relatives. Women's access to the labour market (paid work) did not release them from the caregiver’s tasks (unpaid work), which the family still attaches to them. Women are therefore overworked (be it paid or unpaid tasks), which affects their availability not only for work and training, but also for rest and leisure.
The current trend to flexible working conditions have primarily been developed in the interests of enterprises, neglecting their potential in terms of workers’ welfare and conciliation of their professional, personal and family areas.
The Portuguese Constitution establishes the family as an essential element of society, but such consecration is not reflected, at work, in a special protection to workers family caregivers.
Through the analysis of the current reconciliation mechanisms, gaps will be identified and solutions to the listed problems will be proposed in order to contribute to real equality in work and family
Numerosos
trabalhos reportaram a maior prevalência e incidência desta doença no sexo
masculino, relativamente ao sexo feminino. Estudos envolvendo a reposição
Parkinson´s disease is the second most common neurodegenerative
disorder after Alzheimer and is mainly characterized by a progressive and
selective depletion of dopamine neurons in the Substantia Nigra. Numerous
studies have reported a greater prevalence and incidence of PD in men than in
women. Studies involving estrogen treatment of ovariectomised rodents
attribute this largely to the neuroprotective effets of estrogen. However, a
neuroprotective role for physiologic levels of circulating estrogen in females is
less clear.
Estrogens have also been shown to regulate the expression of
neurotrophic factors, like glial cell line-derived neurotrophic factor (GDNF),
which might mediate their neuroprotective effects. GDNF produces
neuroprotective and regenerative effects in the nigrostriatal pathways, acting as
a potent survival factor for dopaminergic neurons that degenerate in
Parkinson’s disease.
In order to clarify the role of endogenous levels of estrogens in protecting
the nigrostriatal pathway, we used the 6-hydroxydopamine (6-OHDA) model of
Parkinson’s disease and tested how the removal of ovaries in fertile females
interferes with extent of the dopaminergic lesion induced by 6-OHDA. Female
Wistar rats were ovariectomised and 3 weeks after the surgery the animals
were stereotaxically injected in the striatum with 6-OHDA. The extent of the
lesion was assessed by counting the cells expressing the dopaminergic marker
tyrosine hydroxylase by imunohistochemistry and also the expression levels of this protein by Western blot in both the Substantia Nigra and the striatum. The
plasma levels of estradiol were also quantified. To determine if there was a
relationship between estradiol levels, the expression of GDNF and the extent of
the dopaminergic lesion, we also studied the expression of the neurotrophic
factor GDNF.
Our findings strongly suggest that endogenously produced estrogens and
GDNF are associated with increased levels of striatal tyrosine hydroxylase, a
marker of dopaminergic cell survival
This research is based on the study of a linguistic corpus - consisting in
narratives written by speakers of Portuguese Language (LP) from Guinea-Bissau – in
which we observed variations in the nominal reference, compared to European Portuguese (EP), such as: a) the absence and the generalization of definite articles, b)
the absence of indefinite articles and/or replacement of the indefinite article by the
definite article. We conclude that if these features can be explained as an inherent phase in the process of acquiring and learning a language, they are also explained as an inherent phenomenon to a variation, in training, of the Portuguese in Guinea-Bissau
(PGB), similar to that’s already been verified in other African variants of LP, as the Portuguese of Mozambique (PM).
The assertion of language variants, namely PGB, however, represents a dilemma
for many educators and researchers. For, in addition to overpower a strong prestige
attached to the European Portuguese, there is also a certain pragmatism to justify
inaction in the absence of a variant grammars PGB.
In this sense, we decided to study a language corpus of Non-Maternal Portuguese Language (NMPL) of Guinea-Bissau, integrating along with a theoretical
and methodological approach on language development and acquisition/ learning a
language (ADL). Finally, also note the application of a sociolinguistic questionnaire, developed by us, for the sociolinguistic characterization of our informants.
Along with the diversity inherent to the Portuguese in Guinea-Bissau, there is
also the issue of multilingualism in this context, which complicates the scenario of
options, for example, concerning the policy language or teaching methodologies
