Cystathionine Beta-Synthase Deficiency Causes Fat Loss in Mice

    Get PDF
Cystathionine beta synthase (CBS) is the rate-limiting enzyme responsible for the de novo synthesis of cysteine. Patients with CBS deficiency have greatly elevated plasma total homocysteine (tHcy), decreased levels of plasma total cysteine (tCys), and often a marfanoid appearance characterized by thinness and low body-mass index (BMI). Here, we characterize the growth and body mass characteristics of CBS deficient TgI278T Cbs−/− mice and show that these animals have significantly decreased fat mass and tCys compared to heterozygous sibling mice. The decrease in fat mass is accompanied by a 34% decrease in liver glutathione (GSH) along with a significant decrease in liver mRNA and protein for the critical fat biosynthesizing enzyme Stearoyl CoA desaturase-1 (Scd-1). Because plasma tCys has been positively associated with fat mass in humans, we tested the hypothesis that decreased tCys in TgI278T Cbs−/− mice was the cause of the lean phenotype by placing the animals on water supplemented with N-acetyl cysteine (NAC) from birth to 240 days of age. Although NAC treatment in TgI278T Cbs−/− mice caused significant increase in serum tCys and liver GSH, there was no increase in body fat content or in liver Scd-1 levels. Our results show that lack of CBS activity causes loss of fat mass, and that this effect appears to be independent of low serum tCys

Impact of the Algebra I End of Course Examination on African American Students Obtaining a Standard High School Diploma

The state of Florida requires all students complete Algebra I and pass the End of Course Examination (EOCE) to graduate with a standard high school diploma. Algebra I EOCE results indicate that many African American students do not pass the examination. This research sought to determine if there is a relationship between African American students’ failure to pass the Algebra I EOCE and graduate with a standard diploma. Four hypotheses, null and alternative were tested. Two ninth-grade cohorts, 2013-2014 and 2014-2015 comprised the sample. Data were analyzed using t test and one-way analyses of variance (ANOVA). Results indicate a significant relationship between African American students’ failure to pass the Algebra I EOCE and graduation with a standard high school diploma

Radius constraints from high-speed photometry of 20 low-mass white dwarf binaries

We carry out high-speed photometry on 20 of the shortest-period, detached white dwarf binaries known and discover systems with eclipses, ellipsoidal variations (due to tidal deformations of the visible white dwarf), and Doppler beaming. All of the binaries contain low-mass white dwarfs with orbital periods less than 4 hr. Our observations identify the first eight tidally distorted white dwarfs, four of which are reported for the first time here, which we use to put empirical constraints on the mass-radius relationship for extremely low-mass (<0.30 Msun) white dwarfs. We also detect Doppler beaming in several of these binaries, which confirms the high-amplitude radial-velocity variability. All of these systems are strong sources of gravitational radiation, and long-term monitoring of those that display ellipsoidal variations can be used to detect spin-up of the tidal bulge due to orbital decay.Comment: 14 pages, 5 figures, accepted for publication in The Astrophysical Journa

Lung volumes identify an at-risk group in persons with prolonged secondhand tobacco smoke exposure but without overt airflow obstruction.

IntroductionExposure to secondhand smoke (SHS) is associated with occult obstructive lung disease as evident by abnormal airflow indices representing small airway disease despite having preserved spirometry (normal forced expiratory volume in 1 s-to-forced vital capacity ratio, FEV1/FVC). The significance of lung volumes that reflect air trapping in the presence of preserved spirometry is unclear.MethodsTo investigate whether lung volumes representing air trapping could determine susceptibility to respiratory morbidity in people with SHS exposure but without spirometric chronic obstructive pulmonary disease, we examined a cohort of 256 subjects with prolonged occupational SHS exposure and preserved spirometry. We elicited symptom prevalence by structured questionnaires, examined functional capacity (maximum oxygen uptake, VO2max) by exercise testing, and estimated associations of those outcomes with air trapping (plethysmography-measured residual volume-to-total lung capacity ratio, RV/TLC), and progressive air trapping with exertion (increase in fraction of tidal breathing that is flow limited on expiration during exercise (per cent of expiratory flow limitation, %EFL)).ResultsRV/TLC was within the predicted normal limits, but was highly variable spanning 22%±13% and 16%±8% across the increments of FEV1/FVC and FEV1, respectively. Respiratory complaints were prevalent (50.4%) with the most common symptom being ≥2 episodes of cough per year (44.5%). Higher RV/TLC was associated with higher OR of reporting respiratory symptoms (n=256; r2=0.03; p=0.011) and lower VO2max (n=179; r2=0.47; p=0.013), and %EFL was negatively associated with VO2max (n=32; r2=0.40; p=0.017).ConclusionsIn those at risk for obstruction due to SHS exposure but with preserved spirometry, higher RV/TLC identifies a subgroup with increased respiratory symptoms and lower exercise capacity

    Carbonylchlorido(1-methylsulfanylpenta-1,3-dien-1-yl-5-ylidene)bis(triphenylphosphane)osmium(II)

The crystal structure of the title compound, [Os(C6H7S)Cl(C18H15P)2(CO)], confirms the formulation as an osmabenzene. There is a slightly distorted octahedral coordination environment at the OsII ion, with the triphenylphosphane ligands mutually trans and the chloride cis to the carbon bearing the –SMe substituent. Within the metallacyclic ring, the C—C distances are appropriate for aromatic bonds and the two Os—C distances are shorter than typical Os—C single bonds. The maximum deviation from the least-squares plane through the osmabenzene ring occurs for the carbon bearing the SMe substituent [0.1037 (18) Å]
