    Morphological changes induced by testosterone in the mammary glands of female Wistar rats

    Increased levels of androgens in postmenopausal women are considered to be a risk factor for breast cancer. Testosterone, alone or in combination with estrogen, induces epithelial dysplasia and mammary tumors in Noble rats. Since this model of hormone-induced neoplasia has not been reported in other rat strains, we studied the effect of testosterone on the mammary gland morphology of female Wistar rats. Sixty adult, non-castrated, female Wistar rats were implanted in the dorsum midline with a silicone tube containing 50 mg testosterone (testosterone propionate in 30 animals and non-esterified testosterone in the remaining 30 animals) and 20 additional animals were implanted with empty tubes and used as control. Five animals per group were killed 30, 60, 90, 120, 150, and 180 days after implantation, and the mammary glands were dissected, fixed and embedded in paraffin. Histological sections were then stained with hematoxylin and eosin and picrosyrius red for collagen visualization. Morphological and morphometric analysis demonstrated ductal proliferation and acinotubular differentiation with secretory activity in all treated animals, peaking at 90 days of androgen exposure. After 90 days the proliferation of acinar epithelial cells was evident, but there was a progressive reduction of secretory differentiation and an increase in intralobular collagen fibers. There was no morphological evidence of dysplastic changes or other pre-neoplastic lesions. Testosterone treatment applied to adult, non-castrated female Wistar rats induced a mammary gland hyperplasia resembling the lactating differentiation, with progressive reduction in secretory differentiation

    Objetivou-se, neste trabalho, avaliar as alterações físico-químicas proporcionadas pelo tratamento de centros tendinosos diafragmáticos homólogos em solução alcalina seguida de liofilização, para implantação na fáscia interna do músculo reto do abdome de eqüinos. As amostras foram tratadas em períodos de 24, 48, 72, 120 e 144 horas, liofilizadas e analisadas quanto à homogeneidade, flexibilidade e resistência à sutura das amostras. Posteriormente foram caracterizadas por calorimetria exploratória diferencial e microscopia eletrônica de varredura. Para a implantação nos eqüinos, foram utilizadas amostras tratadas por 72 horas seguidas de liofilização, amostras conservadas em glicerina 98% e amostras apenas liofilizadas, que foram retiradas após uma, nove e 18 semanas para avaliar a existência de aderências. Verificou-se que a homogeneidade e a flexibilidade são diretamente proporcionais ao aumento do tempo de tratamento em solução alcalina, enquanto que a resistência é inversamente proporcional ao aumento de tempo, sendo o tratamento por 72 horas intermediário para estas características. A calorimetria exploratória diferencial mostrou que o tratamento não desnatura o colágeno presente nas amostras. Na microscopia eletrônica de varredura, observou-se que o aumento de tempo de tratamento proporciona expansão de zonas menos densas do material. Em relação à formação de aderências, as amostras apenas liofilizadas apresentaram grau máximo na formação da classificação proposta, seguida pelas amostras conservadas em glicerina 98% com grau médio e as amostras tratadas em solução alcalina e liofilizadas, que foram classificadas em grau mínimo. Concluiu-se que o tratamento por 72 horas seria mais apropriado para implantação e que a integração tissular com a parede abdominal foi melhor em relação às amostras apenas liofilizadas e às conservadas em glicerina.<br>The objectives in this research were to evaluate physicist-chemistries alterations in equines diaphragmatic tendineous centers implants submitted to alkaline solution and lyophilization treatment. The samples had been treated in alkaline solution for 24, 48, 72, 120 and 144 hours and lyophilized. It was analyzed the homogeneity, flexibility and resistance to the suture. After that, they had been submitted to the differential scanner calorimetry and to the scanner electronic microscopy. Samples of 72 hours in alkaline treatment, conserved in glycerin 98% and only lyophilized were implanted in recto abdominis internal fascia of equines. They were removed one, nine and 18 weeks post-implantation to evaluate the existence of tacks. It was verified that the homogeneity and flexibility are directly proportional to the increase of time of treatment in alkaline solution while the resistance is inversely proportional to the time increase, being 72 hour treatment intermediate for these characteristics. The differential scanner calorimetry showed that the treatment do not denature the present collagen in samples. The electronic microscopy showed that the increase of treatment time provides expansion of less dense zones of the material. In relation to the formation of tacks, the samples only lyophilized had presented maximum degree in the proposal classification followed for the conserved in glycerin 98% samples with average degree and the treated in alkaline solution and lyophilized samples had been classified in minimum degree. It was concluded that the treatment for 72 hours would be more appropriate for implantation and that the tissue integration with abdominal wall was better in relation to the samples lyophilized only and to the conserved in glycerin ones

    Alterações morfométricas no plexo mioentérico do cólon menor equino distendido experimentalmente Morphometrical alterations in myoenteric plexus of experimentally distended equine small colon

    O cólon menor dos equinos é frequentemente acometido por afecções obstrutivas, sendo a disfunção da motilidade uma complicação comum após o tratamento cirúrgico. Este transtorno pode estar relacionado com lesões no plexo mioentérico ocorridas durante a distensão intestinal, contudo pouco se sabe sobre sua fisiopatologia. O objetivo deste estudo foi avaliar as alterações morfológicas na inervação mioentérica em segmentos de cólon menor de eqüinos submetidos à distensão intraluminal com pressão suficiente para promover redução da perfusão microvascular (isquemia parcial) da parede intestinal. Nove eqüinos foram submetidos à distensão do cólon menor por 4h. Fragmentos da parede intestinal foram colhidos antes e ao final da distensão, após 1,5 e 12 horas de reperfusão no segmento experimental e ao final do procedimento em segmento distante. As amostras foram fixadas e processadas rotineiramente e secções histológicas foram coradas com cresil violeta para a morfometria. Por meio de um software de análise de imagens, obtiveram-se a área, o perímetro e os diâmetros mínimo e máximo do corpo neuronal, do núcleo e do nucléolo dos neurônios e as áreas do citoplasma e do nucleoplasma. Verificou-se redução significativa (P<0,05) das áreas do corpo neuronal e do citoplasma ao final da distensão, retornando aos valores equivalentes aos iniciais durante a reperfusão. Conclui-se que a distensão intraluminal alterou morfologicamente os neurônios do plexo mioentérico. Essas modificações morfológicas podem estar associadas e contribuir para explicar a disfunção da motilidade freqüentemente observada em casos clínicos.<br>The equine small colon is frequently affected by obstruction, and intestinal motility dysfunction is a common complication after its surgical treatment. This fact may be related to myoenteric plexus lesion caused by distention; however, little is known about the pathophysiology of this condition. The objective of this study was to evaluate the morphological alterations in the myoenteric inervation of segments of small colon of horses subjected to intraluminal distension with reduction of the microvascular perfusion (partial ischemia) of the intestinal wall. Nine horses were used to promote distension of on segment of small colon for 4 hours. Samples of intestinal wall were collected before and at the end of the distension, after 1.5 and 12 hours of reperfusion in the experimental segment and at the end of the procedure in a different distant segment. Samples were processed and histological sections were stained with cresyl violet for the morphometric studies. An image analyzer software was used to measure perimeter, diameter, and area of the neuronal body, nucleus and nucleolus of the neurons and the areas of the cytoplasm and nucleoplasm. Significant reductions (P<0.05) in the areas of the neuronal body and cytoplasm were detected at the end of intestinal distension, returning to the basal values during the reperfusion. In conclusion, intraluminal distension promoted changes in the morphology of the neurons of myoenteric plexus. These morphological modifications may be associated to the motility dysfunction frequently observed in clinical cases
