    IFN-γ and TNF-α synergize to inhibit CTGF expression in human lung endothelial cells.

    Connective tissue growth factor (CTGF/CCN2) is an angiogenetic and profibrotic factor, acting downstream of TGF-β, involved in both airway- and vascular remodeling. While the T-helper 1 (Th1) cytokine interferon-gamma (IFN-γ) is well characterized as immune-modulatory and anti-fibrotic cytokine, the role of IFN-γ in lung endothelial cells (LEC) is less defined. Tumour necrosis factor alpha (TNF-α) is another mediator that drives vascular remodeling in inflammation by influencing CTGF expression. In the present study we investigated the influence of IFN-γ and TNF-α on CTGF expression in human LEC (HPMEC-ST1.6R) and the effect of CTGF knock down on human LEC. IFN-γ and TNF-α down-regulated CTGF in human LEC at the promoter-, transcriptional- and translational-level in a dose- and time-dependent manner. The inhibitory effect of IFN-γ on CTGF-expression could be almost completely compensated by the Jak inhibitor AG-490, showing the involvement of the Jak-Stat signaling pathway. Besides the inhibitory effect of IFN-γ and TNF-α alone on CTGF expression and LEC proliferation, these cytokines had an additive inhibitory effect on proliferation as well as on CTGF expression when administered together. To study the functional role of CTGF in LEC, endogenous CTGF expression was down-regulated by a lentiviral system. CTGF silencing in LEC by transduction of CTGF shRNA reduced cell proliferation, but did not influence the anti-proliferative effect of IFN-γ and TNF-α. In conclusion, our data demonstrated that CTGF was negatively regulated by IFN-γ in LEC in a Jak/Stat signaling pathway-dependent manner. In addition, an additive effect of IFN-γ and TNF-α on inhibition of CTGF expression and cell proliferation could be found. The inverse correlation between IFN-γ and CTGF expression in LEC could mean that screwing the Th2 response to a Th1 response with an additional IFN-γ production might be beneficial to avoid airway remodeling in asthma

    We reinvestigate the competition between the Mott and the Anderson insulator state in a one-dimensional disordered fermionic system by a combination of instanton and renormalization group methods. Tracing back both the compressibility and the ac-conductivity to a vanishing kink energy of the electronic displacement field we do not find any indication for the existence of an intermediate (Mott glass) phase.Comment: 4 page

    Intermetallic γ-titanium aluminides are a promising material in high temperature technologies. Their high specific strength at temperatures above 700°C offers the possibility for their use as components of aerospace and automotive industries. With a specific weight of 50% of that of the widely used Ni-based superalloys TiAl is very suitable as material for fast rotating parts like turbine blades in aircraft engines and land based power stations or turbocharger rotors. Thus lower mechanical stresses and a reduced fuel consumption and CO2-emission are expected. To overcome the insufficient oxidation protection the halogen effect offers an innovative way. After surface doping using F-implantation or liquid phase-treatment with an F-containing solution and subsequent oxidation at high temperatures the formation of a protective alumina scale can be achieved. By using non-destructive ion beam analyses (PIGE, RBS) F was found at the metal/oxide interface. For analysis of large scale components a new vacuum chamber at the IKF was installed and became operative. With this prototype of in-situ quality assurance system for the F-doping of manufactured parts from TiAl some performance test measurements were done and presented in this paper.Received: 01 March 2013; Revised: 24 April 2013; Accepted: 25 April 201

    Joint interpretation of geoelectrical and soil-gas measurements for monitoring CO2 releases at a natural analogue

    The development and validation of hierarchic monitoring concepts is essential for detecting and assessing possible leakages from storage formations, especially for carbon capture and storage (CCS) applications. Joint interpretation of various techniques (such as carbon dioxide (CO2) concentration and flux measurements, self-potential (SP) and geoelectrical surveys) showed that the combination of geophysical methods with soil-gas analysis for mesoscale monitoring of the shallow subsurface above geologic CO2 storages can be a valuable tool for mapping and monitoring potential CO2 spread in the subsurface. Three measurement campaigns were undertaken - May 2011, July 2011 and April 2012 - at an analogue site in the Cheb Basin, Czech Republic, with the aim of studying CO2 leakages and their temporal and spatial behaviour. Results of geoelectrical investigations give an insight into the structural features of the subsurface. CO2 discharge into the atmosphere is mostly impeded by shallow, clay-rich, partly water-saturated zones, which can be seen in the electrical resistivity tomography (ERT) results. Several transport processes can be identified based on SP measurements. The SP results highlight the complex behaviour of temporal variations for the flow patterns. In particular, coupled migration of gas and water plays an important influencing role in this process. Site-specific, near surface geological features and meteorological conditions seem to exert great influence on the degassing pattern and measured CO 2 values. Therefore, soil-gas measurements represent a snapshot which illustrates both a distinct typical pattern of the soil-gas distribution in the near subsurface and certain differences caused by soil and meteorological conditions. Observed CO2 soil-gas anomalies and modelled results suggest that the occurrence of gas discharge is much more localized around restricted areas, often controlled by local permeability contrasts. Hence, our results show that a proposed monitoring concept should integrate SP, time-lapse ERT, meteorological parameters and soil-gas measurements to provide a comprehensive insight into the subsurface structures and processes.</p

    A Descent Method for Equality and Inequality Constrained Multiobjective Optimization Problems

    In this article we propose a descent method for equality and inequality constrained multiobjective optimization problems (MOPs) which generalizes the steepest descent method for unconstrained MOPs by Fliege and Svaiter to constrained problems by using two active set strategies. Under some regularity assumptions on the problem, we show that accumulation points of our descent method satisfy a necessary condition for local Pareto optimality. Finally, we show the typical behavior of our method in a numerical example

    PSA based multi objective evolutionary algorithms

    It has generally been acknowledged that both proximity to the Pareto front and a certain diversity along the front, should be targeted when using evolutionary multiobjective optimization. Recently, a new partitioning mechanism, the Part and Select Algorithm (PSA), has been introduced. It was shown that this partitioning allows for the selection of a well-diversified set out of an arbitrary given set, while maintaining low computational cost. When embedded into an evolutionary search (NSGA-II), the PSA has significantly enhanced the exploitation of diversity. In this paper, the ability of the PSA to enhance evolutionary multiobjective algorithms (EMOAs) is further investigated. Two research directions are explored here. The first one deals with the integration of the PSA within an EMOA with a novel strategy. Contrary to most EMOAs, that give a higher priority to proximity over diversity, this new strategy promotes the balance between the two. The suggested algorithm allows some dominated solutions to survive, if they contribute to diversity. It is shown that such an approach substantially reduces the risk of the algorithm to fail in finding the Pareto front. The second research direction explores the use of the PSA as an archiving selection mechanism, to improve the averaged Hausdorff distance obtained by existing EMOAs. It is shown that the integration of the PSA into NSGA-II-I and Δ p -EMOA as an archiving mechanism leads to algorithms that are superior to base EMOAS on problems with disconnected Pareto fronts. © 2014 Springer International Publishing Switzerland

    Chomsky’s principle of epistemological tolerance says that in theoretical linguistics contradictions between the data and the hypotheses may be temporarily tolerated in order to protect the explanatory power of the theory. The paper raises the following problem: What kinds of contradictions may be tolerated between the data and the hypotheses in theoretical linguistics? First a model of paraconsistent logic is introduced which differentiates between week and strong contradiction. As a second step, a case study is carried out which exemplifies that the principle of epistemological tolerance may be interpreted as the tolerance of week contradiction. The third step of the argumentation focuses on another case study which exemplifies that the principle of epistemological tolerance must not be interpreted as the tolerance of strong contradiction. The reason for the latter insight is the unreliability and the uncertainty of introspective data. From this finding the author draws the conclusion that it is the integration of different data types that may lead to the improvement of current theoretical linguistics and that the integration of different data types requires a novel methodology which, for the time being, is not available
