Összefüggés a fehérjék stabilitása, konformációs flexibilitása és működése között = Relationship between stability, conformational flexibility and function of proteins
HomolĂłg hĹ‘kedvelĹ‘, mezofil Ă©s hidegtűrĹ‘ enzimek sorozatainak szerkezeti, szervezĹ‘dĂ©si (folding) Ă©s funkcionális (reakciĂł kinetikai) összehasonlĂtĂł vizsgálatát vĂ©geztĂĽk el. A katalitikus reakciĂł enzimkinetikai paramĂ©tereinek a hĹ‘mĂ©rsĂ©klet Ă©s a konformáciĂłs fluktuáciĂłk (relaxáciĂłs spektrum) fĂĽggvĂ©nyĂ©ben törtĂ©nĹ‘ analĂzisĂ©vel megállapĂtottuk, hogy a gliceraldehid-3-foszfát dehidrogenáz enzim alegysĂ©gei közötti allosztĂ©rikus jeltovábbĂtásban a molekula dinamikus tulajdonságainak meghatározĂł szerepe van. A több domĂ©nbĹ‘l állĂł modellfehĂ©rjĂ©k (IPMDH, PGK) esetĂ©ben feltártuk a domĂ©nzárĂłdás atomi szintű lĂ©pĂ©seinek összefĂĽggĂ©sĂ©t a katalitikus folyamat egyes elemeivel. A fehĂ©rjĂ©k folding-refolding folyamatainak kinetikai analĂzĂsĂ©vel több, a hĹ‘stabilitás mechanizmusát Ă©rtelmezĹ‘ Ăşj megállapĂtást tettĂĽnk. Vizsgáltuk a Thermotoga maritima eubaktĂ©riumbĂłl származĂł, hasonlĂłságot mutatĂł termofil Ă©s hipertermofil xilanázok hĹ‘stabilitása közötti kĂĽlönbsĂ©g szerkezeti hátterĂ©t. IrányĂtott family shuffling segĂtsĂ©gĂ©vel számos kimĂ©ra enzimet állĂtottunk elĹ‘ a kĂ©t vad tĂpusĂş xilanázbĂłl. A termofil xilanáz enzim terminális rĂ©giĂłit lecserĂ©lve a hipertermofil xilanáz enzim terminális rĂ©giĂłira, a termofil enzim hĹ‘stabilitását 12 oC-kal megnöveltĂĽk. A lĂ©trehozott kimĂ©ra konstrukciĂłk arra világĂtottak rá, hogy a terminális rĂ©giĂłk közötti kölcsönhatás stabilizálásával megnövelhetĹ‘ azon fehĂ©rjĂ©k hĹ‘stabilitása, amelyek terminális rĂ©giĂłi megfelelĹ‘ közelsĂ©gben találhatĂłak. | Comparative structural, folding and functional studies were completed using sets of homologous thermophilic, mesophilic, and psychrotropic enzymes. It was revealed that the dynamic features of the enzyme molecule are essential for the mechanism of intramolecular, allosteric signal transduction between subunits of glyceraldehyde-3-phosphate dehydrogenase. The steps of domain closure, at the atomic level, were related to individual elements of the catalytic process in the case of multidomain enzymes (IPMDH, PGK). A novel mechanistic interpretation was developed based on kinetic analyses of folding-refolding processes. The structural background of the difference in heat stabilities between homologous thermophilic and hyperthermophilic xylanases from Thermotoga maritima was studied. Several chimeric constructs of the two parental enzymes were produced by directed family shuffling. A significant increase in heat stability was achieved in the case of one construct. Exchanging the terminal regions of the thermophilic enzyme by the respective fragments of the hyperthermophilic xylanase, the heat stability of the enzyme increased by 12 oC. Characterization of chimeric constructs showed that the interactions between terminal regions have an important role in stabilizing protein structure, and based on this observation, heat stability can be increased in proteins in which the terminal regions are in close proximity
Lineáris Ă©s paramĂ©ter fĂĽggĹ‘ lineáris kapcsolĂł ĂĽzemmĂłdĂş rendszerek irányĂtása = Control of linear and parameter-varying switching mode systems
A kutatás során a hibrid rendszerek egy osztályára, amit lineáris kapcsoló üzemű rendszerekenek neveznek, rendszerelméleti erdményeket dolgoztunk ki. A munka során elsősorban az alapvető rendszer tulajdonságok, mint a megfigyelhetőség és irányithatóság feltételeivel és ellenőrzésük módszereivel foglalkoztunk. A kapcsoló üzemű rendszerek egy fontos osztályával, a bimodális rendszerekkel részletesen foglalkoztunk, itt a kapcsolási feltétel állapot függő. Kidolgoztunk egy olyan bimodális irányithatósági/megigyelhetőségi kritériumot és a hozzá tartozó ellenőrzési módszert amely az eredeti Kalman - féle kritérium bimodális általánositásának tekinthető. Az eredményeket időfüggő és nemlineáris rendszerek egy bizonyos gyakorlat szempontjából fontos osztályára, a linráris paraméterváltozós (LPV) rendszerek osztályára is ki kiterjesztettük és több jármű és közlekedési rendszer irányitási analizisében felhasználtuk. | This research focused on developing cotheory for investigation of controllability and observability problems of switching linear and linear parameter varying systems (LPV). An important class of this sort of hybrid systems is the class of bimodal systems where the switching condition is system state dependent. We elaborated a controllability/observability criterion and the associated test procedure that can be be considered as the generalization of the original Kalman - rank test for the case of switching systems. The results were extended to certain LPV systems and were used in the analysis of vehicle and traffic control systems
Az elmĂ©letileg elĂ©rhetĹ‘ legjobb irányĂtás algoritmusainak kutatása = Investigation of the theoretically reachable best control algorithms
MĂłdszert dolgoztunk ki lineáris, állandĂł paramĂ©terű folyamatok szabályozási köreiben a beavatkozĂł szervek Ă©s magának a folyamatnak a tulajdonságaibĂłl adĂłdĂł korlátozásoktĂłl fĂĽggĹ‘ határ-optimális szabályozĂłk tervezĂ©sĂ©re. Algoritmust fejlesztettĂĽnk ki a beavatkozĂł szerv amplitudĂł korlátozásátĂłl fĂĽggĹ‘ elĂ©rhetĹ‘ legjobb tervezĂ©si cĂ©l (referencia modell) meghatározására. Megadtuk a nominális Ă©s az elĹ‘zĹ‘ mĂłdszerrel kapott optimális referencia modellektĹ‘l fĂĽggĹ‘ optimális szabályozĂłk algoritmusait, amelyek a realizálhatĂłsági vesztesĂ©get minimalizálják. Az optimális szabályozĂłkat stabilis folyamatok Youla parametrizált szabályozási köreire határoztuk meg. Az optimalitás kritĂ©riumakĂ©nt a H2, Hinf Ă©s L2, Linf normákat alkalmaztuk. A realizálhatĂłsági vesztesĂ©g H2, L2 normák szerinti optimalitását a szabályozĂłk - speciális Diofantoszi egyenletek alapján számolt - belsĹ‘ szűrĹ‘inek felhasználásával biztosĂtottuk. A realizálhatĂłsági vesztesĂ©g Hinf, Linf normák szerinti optimalitását a szabályozĂłk - speciális Nevanlinna-Pick approximáciĂłs egyenletek alapján számolt - belsĹ‘ szűrĹ‘inek alkalmazásával biztosĂtottuk. A H2, Hinf normák alkalmazásával csak nem integrálĂł optimális szabályozĂł nyerhetĹ‘. Optimális integrálĂł szabályozĂłhoz az eredeti normákat speciális ""energia"" illetve ""supremum"" normákká kellett kiegĂ©szĂteni az L2, Linf normák egyidejű alkalmazásával Ă©s nem Dirac-delta alakĂş gerjesztĂ©s feltĂ©telezĂ©sĂ©vel. Ăšj iteratĂv mĂłdszert vezettĂĽnk be a modellezĂ©si vesztesĂ©g optimalizálására is. | A new method was developed for design of the (reachable) best controllers for LPI processes when the activator has an amplitude constraint and the process has invariant properties. A procedure was suggested to compute the best (reachable) reference model for tracking and disturbance rejection. A new approach was introduced to minimize the realizability degradation part of the sensitivity function using nominal process models. This optimization uses Youla parametrized regulators for open-loop stable plants. H2, Hinf and L2, Linf norms are used in this procedure. The H2, L2 norm based optimization uses special Diophantine equation to calculate the optimal embedded filter in the regulators. The Hinf, Linf norm based optimization uses special Nevanlinna-Pick approximation to calculate the optimal imbedded filter in the regulators. The original norm formulation was used in a combined way forming general ''energy'' and ''supremum'' norms assuming higher order excitations than the Dirac-delta to ensure integrating optimal regulators, too. New iterative algorithm was developed to minimize the modeling degradation part of the sensitivity function

Egy ismeretlen „őserdő” a Kelet-Mecsekben : 25 éve felhagyott bükkös aljnövényzetének térbeli mintázatai = A scarcely known old-growth forest in Eastern Mecsek hills : Spatial patterns of the herb layer of a beech forest abandoned 25 years ago
A 130-years old beech forest stand (Helleboro odori – Fagetum) in Eastern Mecsek hills (Southern Hungary), abandoned 22-25 years ago, provides excellent opportunities for the study of spontaneous forest dynamics. In 2010 intensive studies started, iniciated by Mecsekerdő Zrt: repeating an individuum-based tree-stand survey of 1986-87, and a gridbased herb layer survey according to the Forest Reserve Protocol. In the herb layer 95 species were found, amongst them 14 protected ones, most of them in the hillside-hilltop ecotone. Most important are the more thousand exemplars of Doronicum orientale. More than 30% percentage of specialist and competitor species indicates a very good naturalness state, comparable with forest reserves
PEAR, a flexible fluorescent reporter for the identification and enrichment of successfully prime edited cells
Prime editing is a recently developed CRISPR/Cas9 based gene engineering tool that allows the introduction of short insertions, deletions, and substitutions into the genome. However, the efficiency of prime editing, which typically achieves editing rates of around 10%-30%, has not matched its versatility. Here, we introduce the prime editor activity reporter (PEAR), a sensitive fluorescent tool for identifying single cells with prime editing activity. PEAR has no background fluorescence and specifically indicates prime editing events. Its design provides apparently unlimited flexibility for sequence variation along the entire length of the spacer sequence, making it uniquely suited for systematic investigation of sequence features that influence prime editing activity. The use of PEAR as an enrichment marker for prime editing can increase the edited population by up to 84%, thus significantly improving the applicability of prime editing for basic research and biotechnological applications

A Lineráris ParamĂ©terVáltozĂłs (LPV) rendszerek alkalmazása kĂĽlönösen lĂ©gi Ă©s földi járművek modellezĂ©si Ă©s irányĂtási feladatainak megoldásában vált nagy jelentĹ‘sĂ©gĂĽvĂ©. Ezek a rendszerek nemlineáris, idĹ‘fĂĽggĹ‘ dinamikájĂşak Ă©s az LPV modellezĂ©si paradigma lehetĹ‘sĂ©get ad a geometriai rendszerelmĂ©let, valamint a robusztus irányĂtások lineáris Ă©s affin nemlináris mĂłdszertanának alkalmazására. A kutatási program során sikerĂĽlt megalapozni az LPV rendszerek geometriai elmĂ©letĂ©t Ă©s több olyan algoritmust dolgoztunk ki amellyel rendszer analĂzis, hibadetektálás irányĂtás Ă©s átkonfigurálás megoldhatĂł. Ez utĂłbbira a University of Minnesota Aerospace and Mechanics tanszĂ©kĂ©vel közös kutatás keretĂ©n belĂĽl kerĂĽlt sor, az eredmĂ©nyeket folyĂłiratokban Ă©s konferencia kiadványokban publikáltuk. | Linear Parameter Varying (LPV) systems gained an increasing importance in the area of road and aerial vehicle modelling and control system design. These systems can be described by complex nonlinear and time-varying dynamics and the use of LPV modeling paradighms allows the designer to use results from geometric theory and robust control methods elaborated for linear certain nonlinear afine control systems. This research program resulted in the elaboration of the foundations of a geometric system theory for LPV systems and this lead to the implementation of new algorithms to solve analysis and design problems associated to fault detection, controller design and control system reconfiguration for certain class of time varying and nonlinear systems. A summary of the research results was made public in the form of a semiĂĽplenary lecture at the 16th IFAC Word Congress (2005) and the results were also veryfied in the desig of control systems for road and aerial vehicles.
