    Did political constraints bind during transition? Evidence from Czech elections 1990 - 2002

    Many theoretical models of transition are driven by the assumption that economic decision making is subject to political constraints. In this paper we empirically test whether the winners and losers of economic reform determined voting behaviour in the first five national elections in the Czech Republic. We propose that voters, taking stock of endowments from the planning era, could predict whether they would become “winners” or “losers” of transition. Using survey data we measure the percentage of individuals by region who were “afraid” and “not afraid” of economic reform in 1990. We define the former as potential “winners” who should vote for pro-reform parties, while latter are potential “losers” who should support left-wing parties. Using national election results and regional economic indicators, we demonstrate that there is persistence in support for pro-reform and communist parties driven by prospective voting based on initial conditions in 1990. As a result, we show that regional unemployment rates in 2002 are good predictors of regional voting patterns in 1990.

    Students’ Perceptions of their First Practice Placement in Early Childhood Care and Education

    It is widely accepted in higher education that practice placements facilitate students’ professional development by providing a beneficial learning environment, the chance to experience the ‘world of work’ and to develop the skills of a competent practitioner. However, the transition from college to practice placement, adjusting to new learning environments and managing workload demands can be challenging for students. This paper explores some of these issues. It investigates students’ perceptions regarding their first practice placement on an Early Childhood Care and Education programme in a third-level college in the Republic of Ireland. The study examines students’ views on specific aspects of placement, including preparation, management of the assessment portfolio and reflective journal, experience of supervision and opportunities for acquiring new skills and competencies.A self-administered, non-experimental survey, comprising closed and open questions, was utilised to allow the gathering of specific statistical information (quantitative) as well as students’ views and opinions (qualitative) of placement experiences. The findings indicate that the first practice placement was, in general, a positive experience, whereby students enjoyed opportunities to work with children and link theory to practice. However, it was found that there were difficulties in a) writing regular journal entries and reflective writing, b) completing child observations and c) implementing a planned intervention.     Spiritan faith friends: an evaluation of faith development in a peer-ministry programme

    This thesis sought to evaluate how a Christian Peer-ministry programme entitled ‘Spiritan Faith Friends’ (SFF) impacted the faith development of male adolescents in three second-level Spiritan Schools in Ireland. What faith development meant in the context of this programme was explored, as well as how the faith development of the participants was impacted by a sense of connectedness that may have arisen from belonging to such a programme within a denominational school setting. The definition of ‘connectedness’ used is in agreement with that of Karcher and Lee (2002, p.93), who define it as ‘a response to relatedness and belonging. …. Connectedness, then, reflects one’s perception of his or her own involvement in and affection for others, activities and organisations.’ There are two elements to draw from this definition: first, connectedness and belonging are distinct from each other yet related, and, second, connectedness which in this sense arises out of one’s sense of belonging, has a reciprocal nature, received as well as given. Many questions were asked, among them: How might this programme impact the faith development of the male adolescent participants? How might it connect with them during this stage of development? How might it connect with their search for meaning within a religious identity? Does it offer a sense of belonging [to the Christian community] by affording the experience of a role in the Catholic Church? Does the denominational setting impact the outcomes of the programme? The research was conducted in two stages: an action research stage and a lasting impact stage. Both stages employed ‘a wide range of interconnected interpretative methods,’ as the researcher’s ontology filtered the lens through which she guided and effected the final ‘bricolage’ (Denzin and Lincoln 2003, p.5)

    Hepatitis C quasispecies adaptation in the setting of a variable fidelity polymerase

    Hepatitis C (HCV) is a virus characterized by an RNA-dependent RNA polymerase that lacks a proofreading mechanism and, as a result, generates a quasispecies. There is emerging evidence that this RNA-dependent RNA polymerase may in fact have variable fidelity. Here, we review the relevant concepts, including fitness landscapes, clonal interference, robustness, selection, adaptation, mutation rates, and their optimization, and provide a unique interpretation of a number of relevant theoretical models, evolving the theory of replicative homeostasis in light of their findings. We suggest that a variable fidelity polymerase can find its own optimal mutation rate, which is governed by the sequence itself and certain population dynamics. We propose that this concept can explain features of viral kinetics and clearance, both spontaneously and following treatment of chronic HCV. We point to evidence that supports this theory and explain how it refines replicative homeostasis and conclude by discussing particular areas of potential research that might augment our understanding of viral host interactions at an individual cellular level

    In an investigation of 4-amino-3,4-dihydronaphthalen-1(2H)-ones as novel modulators of allergic and inflammatory phenomena, we have investigated a series of cyclic analogues. Tertiary amines of structural types 9, 10, 20 and 21 were synthesised and evaluated for mast cell stabilising activity. In vitro and in vivo studies showed that of these compounds, the cyclohexenylamino derivatives of tetralone and benzosuberone of series 20 and 21 exhibited interesting activity both in vitro and in vivo

    Effects of Feeding Bt Maize to Sows during Gestation and Lactation on Maternal and Offspring Immunity and Fate of Transgenic Material

    peer-reviewedBackground: We aimed to determine the effect of feeding transgenic maize to sows during gestation and lactation on maternal and offspring immunity and to assess the fate of transgenic material. Methodology/Principal Findings: On the day of insemination, sows were assigned to one of two treatments (n = 12/treatment); 1) non-Bt control maize diet or 2) Bt-MON810 maize diet, which were fed for ~143 days throughout gestation and lactation. Immune function was assessed by leukocyte phenotyping, haematology and Cry1Ab-specific antibody presence in blood on days 0, 28 and 110 of gestation and at the end of lactation. Peripheral-blood mononuclear cell cytokine production was investigated on days 28 and 110 of gestation. Haematological analysis was performed on offspring at birth (n = 12/treatment). Presence of the cry1Ab transgene was assessed in sows' blood and faeces on day 110 of gestation and in blood and tissues of offspring at birth. Cry1Ab protein presence was assessed in sows' blood during gestation and lactation and in tissues of offspring at birth. Blood monocyte count and percentage were higher (P<0.05), while granulocyte percentage was lower (P<0.05) in Bt maize-fed sows on day 110 of gestation. Leukocyte count and granulocyte count and percentage were lower (P<0.05), while lymphocyte percentage was higher (P<0.05) in offspring of Bt maize-fed sows. Bt maize-fed sows had a lower percentage of monocytes on day 28 of lactation and of CD4+CD8+ lymphocytes on day 110 of gestation, day 28 of lactation and overall (P<0.05). Cytokine production was similar between treatments. Transgenic material or Cry1Ab-specific antibodies were not detected in sows or offspring. Conclusions/Significance: Treatment differences observed following feeding of Bt maize to sows did not indicate inflammation or allergy and are unlikely to be of major importance.     Species classifier choice is a key consideration when analysing low-complexity food microbiome data

    peer-reviewedBackground The use of shotgun metagenomics to analyse low-complexity microbial communities in foods has the potential to be of considerable fundamental and applied value. However, there is currently no consensus with respect to choice of species classification tool, platform, or sequencing depth. Here, we benchmarked the performances of three high-throughput short-read sequencing platforms, the Illumina MiSeq, NextSeq 500, and Ion Proton, for shotgun metagenomics of food microbiota. Briefly, we sequenced six kefir DNA samples and a mock community DNA sample, the latter constructed by evenly mixing genomic DNA from 13 food-related bacterial species. A variety of bioinformatic tools were used to analyse the data generated, and the effects of sequencing depth on these analyses were tested by randomly subsampling reads. Results Compositional analysis results were consistent between the platforms at divergent sequencing depths. However, we observed pronounced differences in the predictions from species classification tools. Indeed, PERMANOVA indicated that there was no significant differences between the compositional results generated by the different sequencers (p = 0.693, R2 = 0.011), but there was a significant difference between the results predicted by the species classifiers (p = 0.01, R2 = 0.127). The relative abundances predicted by the classifiers, apart from MetaPhlAn2, were apparently biased by reference genome sizes. Additionally, we observed varying false-positive rates among the classifiers. MetaPhlAn2 had the lowest false-positive rate, whereas SLIMM had the greatest false-positive rate. Strain-level analysis results were also similar across platforms. Each platform correctly identified the strains present in the mock community, but accuracy was improved slightly with greater sequencing depth. Notably, PanPhlAn detected the dominant strains in each kefir sample above 500,000 reads per sample. Again, the outputs from functional profiling analysis using SUPER-FOCUS were generally accordant between the platforms at different sequencing depths. Finally, and expectedly, metagenome assembly completeness was significantly lower on the MiSeq than either on the NextSeq (p = 0.03) or the Proton (p = 0.011), and it improved with increased sequencing depth. Conclusions Our results demonstrate a remarkable similarity in the results generated by the three sequencing platforms at different sequencing depths, and, in fact, the choice of bioinformatics methodology had a more evident impact on results than the choice of sequencer did.This research was funded by Science Foundation Ireland in the form of a centre grant (APC Microbiome Institute grant number SFI/12/RC/2273).
