Functional anatomy of female sex pheromone gland of Mayetiola destructor Say (Diptera: Cecidomyiidae)
FUNCTIONAL ANATOMY OF FEMALE SEX PHEROMONE GLAND OF MAYETIOLA DESTRUCTOR SAY (DIPTERA: CECIDOMYIIDAE) The Hessian Fly (Mayetiola destructor Say, Diptera Cecidomyiidae) sex calling of males by females, known from long ago, has been recently demonstrated of being mediated by a female produced sex pheromone that has been chemically identified lately. Following the investigation methods previously applied to study sex pheromone glands in other Cecidomyiidae species (i.e., Allocontarinia sorghicola (Coq.) Solinas and Dasineura brassicae Winn.), the female sex pheromone gland of the Hessian Fly has been identified through functional-anatomy, ultrastructural, and physiological observations here reported. The pheromone gland consists of the 8th-9th abdominal intersegmental membrane transformed epidermis. This appears quite different in virgin than mated females being in the former strongly hypertrophied and obviously atrophied in the latter. Seguendo lo stesso metodo messo a punto dagli stessi autori per lo studio delle ghiandole a feromoni di altre cecidomie (Allocontarinia sorghicola (Coq.) Solinas e Dasineura brassicae Winn.), è stata identificata la ghiandola a feromoni sessuali della Cecidomia distruttrice del grano, mediante indagini anatomo-funzionali, ultrastrutturistiche e fisiologiche qui riportate. Detta ghiandola a feromoni risulta costituita dall’epidermide trasformata della membrana intersegmentale tra 8° e 9° urite, la quale si presenta fortemente ipertrofica nelle femmine vergini sessualmente mature, mentre si riduce bruscamente di volume e si atrofizza in seguito all’accoppiamento.
Functional morphology of the antennal chemosensilla of Ceutorhynchus assimilis Payk. (Coleoptera: Curculionidae)
The cabbage seed weevil (Ceutorhynchus assimilis Payk.) is a serious pest of oilseed rape (Brassica napus L.) in Europe. Previous EAGs and behavioural investigations suggest an essential role of the antennal olfaction and taste in the chemical ecology of the weevil, but morpho- functional knowledge of its antennal sensilla is still lacking.To fill the gap, an anatomo-ultrastructural study of C assimilis antennal chemosensilla has been carried out and presented here. The sensilla in question have been found on the antennal club (9th + 10th + 11th antennomeres) only. Three types of chemosensilla have been identified and illustrateci: (i) sensilla chaetica (CH), the most prominent, arranged in three whorls, ultrastructurally gustative: (ii) peg-like sensilla (P), the most numerous but difficult to detect, ultrastructurally olfactive; (iii) hair-like (H) or sickle shaped sensilla, moderately numerous, ultrastructurally olfactive. The behavioural meaning of the men- tioned sensilla is discussed, and concluding suggestions are given to a fuller understanding of electrophysiological studies.MORFOLOGIA FUNZIONALE DEI CHEMIOSENSILLI ANTENNALI DI CEUTORHYNCHUS ASSIMILIS PAYK. (COLEOPTERA: CURCULIONIDAE) Il punteruolo delle silique del colza (Ceutorhynchus assimilis Payk.) è un fitofago seriamente dannoso alla oleaginosa in tutta Europa. Studi preliminari comportamentali ed elettrofisiologici hanno messo in evidenza un ruolo essenziale dell'olfatto e del gusto antennali nella ecologia chimica dell'insetto. Una base morfo-funzionale, indispensabile allo sviluppo di detti studi e tuttora mancante, viene qui fornita mediante ricerche anotomo-ultrastrutturistiche sui sensilli implicati nel ruolo in questione. Tali sensilli, presenti esclusivamente sulla clava antennale (antennomeri 9°+10°+11°), risultano essere di 3 tipi: l) "sensilla chaetica" (CH), i più vistosi, sistemati in tre verticilli, ultrastruttural- mente gustativi; 2) sensilli "peg-like" (P; esternamente a forma di piolo), i più numerosi ma difficili da individuare tra la folta peluria di rivestimento, ultrastrutturalmente olfattivi; 3) sensilli "hair- like", distalmente falciformi, moderatamente numerosi, ultrastrutturalmente pure olfattivi. Viene discusso il significato etologico dei sensilli menzionati e vengono fatte considerazioni conclusive utili, in particolare, ad ottimizzare la comprensione di ulteriori studi elettrofisiologici.
Eggshell fine morphology of Allocontarinia sorghicola (Coq.) (Diptera: Cecidomyiidae)
The fine morphology of the Sorghum midge Allocontarinia sorghicola (Coq.) eggshell has been investigated, in ovary and laid eggs, with scanning and transmission electron microscopes. The eggshell consists of three layers: exochorion, endochorion and vitelline membrane. The micropylar apparatus, situated at the egg anterior pole, has been studied in detail. It is formed by an exochorion invagination delimiting a sort of atrium with one external and several internal apertures. At the egg posterior pole stands out a long appendage becoming wrinkled few hours after oviposition and probably having mechanical function facilitating the egg entering and passing through the female reproductive ducts. Key words (in addition to those in the title): chorion, micropyle, vitelline membrane, ultrastructure.  MORFOLOGIA FINE DEGLI INVOLUCRI (CORION E MEMBRANA VITELLINA) DELL'UOVO DI ALLOCONTARINIA SORGHICOLA (COQ.) (DIPTERA: CECIDOMYIIDAE)  Sono riportati i risultati di una indagine condotta al microscopio elettronico a scansione ed a trasmissione relativi alla struttura del corion e della membrana vitellina di uova ovariche e deposte di Allocontarinia sorghicola. La regione micropilare, situata al polo anteriore dell'uovo, è stata particolarmente studiata. Il micropilo è costituito da una invaginazione del solo esocorion delimitante una sorta di atrio con imboccatura (apertura micropilare esterna) più stretta del fondo nel quale tutt'attorno si aprono diversi fori laceri (aperture micropilari interne). Nelle uova deposte, le superfici dell'esocorion delimitanti l'atrio finiscono per aderire l'una all'altra, occludendo tutte le aperture micropilari. Il lungo peduncolo situato al polo posteriore ha molto probabilmente la funzione di facilitare il passaggio dell'uovo lungo i gonodotti femminili
Ultrastructure of the anterior alimentary tract of Bursaphelenchus mucronatus Mamiya et Enda (Nematoda Aphelenchoididae)
In view of the phytosanitary importance of nematodes of the genus Bursaphelenchus and the limited information available on their ultrastructure, we carried out a study of the buccal cavity and oesophageal area of Bursaphelenchus mucronatus Mamiya et Enda, 1979. Like the quarantine species Bursaphelenchus xylophilus (Steiner et Bührer) Nickle, its development takes place in the resin canals of conifers. The specimens used in our study were taken from a laboratory population collected on Pinus pinaster Aiton in meso-thermophilic mixed forests of central Italy (Tuscany) and grown on Petri dishes in a climatic chamber for 20 generations. The ultrastructural features of the mouth opening, anterior intestine, labial sensilla, amphidial canals, stylet and metacorpus were investigated with a TEM. The oesophageal glands are also described. The observed structures are discussed and compared with those described for other nematodes
Antennal sensory structures and their ecological-behavioural meaning in Cecidomyiidae (Diptera) larvae
Cecidomyiidae larvai antennae, so far corisidered to be biarticulate (rarely triarticulate), are here indicated as uniarticulate on the basis of anatomicai evidence. Each of these monomerous antennae rises on a dome-shaped prominence (former first antennomere) and bears 5 kinds of sensilla: one large placoid sensillum (multiporous placoid sensillum), covering most of the antennai surface, a dorsal one (basai sensillum), and an apicai sensillum; an intermediate dorsai one (sensillum microampullaceum) and a basai, completely internai proprioceptor, both very difficult to detect with a light microscope. Functionai morphology of the mentioned sensilla, carried out in scanning and transmission electron microscopy, reveaied chat: the large placoid sensillum is a multiporous che- moreceptor, most probably olfactory; the basal s. is a mechanoreceptor, probably hygro- and thermoreceptor ; che apical s. is a uniporous chemoreceptor , most probably gustative ; the s. microampullaceum is very difficult to interpret on a mere morphological basis; the proprioceptor may be interpreted as aJohnston's organ reduced to a single scolopidium. Some considerations about ecological-behaviourai involvement of these antennai strucrures are here given. Key words: anatomy, chemoreceptors, convergent adaptation, functionai morphology, hygrothermoreceptor, scolopidium, ultrastructure.Le larve dei Cecidomiidi (Cecidomyiidae) hanno la testa minuscola ma bene strutturata, anoftalma, provvista di antenne relativamente ben sviluppate, in posizione di riposo completamente invaginata nel protorace, donde fuoriesce per esplorare l'ambiente circostante, per gli spostamenti della larva e per l'assunzione del cibo. La morfologia antennale è molto caratteristica, notevolmente omogenea nell'ambito della famiglia e piuttosto semplice, trattandosi di antenne sostanzialmente uniarticolate, apparentemente biarticolate (eccezionalmente triarticolate) per essere il torulo posto alla sommità di una vistosa prominenza membranosa, spesso interpretata, erroneamente, come «primo antennomero». Nonostante le ridotte o ridottissime dimensioni e l'apparente limitazione d'uso, dette antenne risultano sorprendentemente dotate di organi di senso tra i più complessi finora conosciuti: una «placca olfattiva» (Figs: MP) che ricopre in massima parte la superficie antennale; un chemiore- cettore apicale di tipo gustativo (AS); un recettore dorsale con apparato esterno assai minuto, di tipo ampullaceo (MA) ma difficilmente interpretabile su base esclusivamente morfologica; un mec- canorecettore basale (BS) con probabili funzioni igrorecettiva e termorecettiva; un propriorecettore basale interno, interpretabile come organo di]OHNSTON ridotto ad un singolo scolopidio. Dette strutture sensoriali appaiono nell'insieme idonee ad assicurare alla larva le informazioni ecologiche es- senziali per vivere nel suo microambiente. In particolare, lo sviluppo della placca olfattiva risulta strettamente correlato con la maggiore o minore difficoltà per le larve nel reperire il cibo e l o il rifugio idoneo per impuparsi e per svernare.
Antennal chemosensilla and glands, and their possible meaning in the reproductive behaviour of Trissolcus basalis (Woll.) (Hym. : Scelionidae)
The antennae of Trissolcus basalis, a solitary endoparasitoid of Nezara viridula (Heteroptera: Pen- tatomidae) eggs, have been investigated by SEM and TEM to map and describe the different types of chemosensilla and glands, some of which are reported for the first time in Parasitic Hymenoptera. A non chemoreceptor, sensillum campaniforme, has also been included since it is associateci with glands. A functional interpretation has been attempted correlating the antennal structures with mating and host recognition behaviours previously reported. SENSILLI CHEMIORECETTORI E GHIANDOLE ANTENNALI E LORO POSSIBILE SIGNIFICATO NEL COMPORTAMENTO RIPRODUTTIVO DI TRISSOLCUS BASALIS (WOLL.) (HYMENOPTERA, SCELIONIDAE)  Il Trissolcus basalis è un parassitoide solitario delle uova di Nezara viridula L. Dalle ovature parassitizzate sfarfallano per primi i Trissolcus maschi, i quali, essendo capaci di individuare la presenza delle proprie femmine (per via di un feromone sessuale volatile) all'interno delle uova dell'ospite, si fermano singolarmente davanti all'uovo parassitizzato, in attesa che sfarfalli l'individuata compagna. Alla fuoriuscita di questa seguono speditamente un rapido corteggiamento da parte del maschio, il riconoscimento e la reciproca accettazione del partner e quindi la copula: il tutto in poche decine di secondi. La femmina fecondata va subito in cerca di un'ovatura idonea di Nezara per ovideporre lasciando infine un feromone di marcatura sulle singole uova da essa parassitizzate. Detto comportamento riproduttivo risulta chiaramente e necessariamente mediato dalle antenne di ambo i sessi nella prima parte, e della femmina, ovviamente, nella seconda parte. Attraverso uno studio in stretta collaborazione tra etologi (Bin, Colazza, Vinson) e morfologi (Isidoro, Solinas) vengono identificati sensilli chemiorecettori e ghiandole antennali implicati nei meccanismi comportamentali sopra menzionati. In particolare, sono stati individuati, descritti e più o meno ampiamente interpretati: a) un solo tipo di sensilli olfattivi («sickle-shaped sensilla), presente in ambo i sessi, ma assai più rappresentato e potente nei maschi (recettori del feromone sessuale?) e più versatile nelle femmine (recettore di kairomone e sinomoni?); b) quattro tipi di sensilli gustativi, due dei quali sono presenti in ambo i sessi (sensilla chaetica, chiaramente implicati nel reciproco riconoscimento tra i sessi; e grooved peg sensilla, di difficile interpretazione), mentre un terzo tipo, del tutto particolare, è presente solo nelle femmine, (papillary sensilla» chiaramente implicati nel riconoscimento- accettazione dell'ospite), ed un quarto, pure particolare, ma meno facilmente interpretabile, è presente solo nei maschi (sensilla conica»); c) tre tipi di ghiandole di cui uno è presente in ambo i sessi (dorsal glands», chiaramente implicato nel reciproco - ma soprattutto da parte del maschio - riconoscimento tra i sessi), mentre degli altri due tipi, uno è associato ai papillary sensilla della femmina e dunque strettamente legato al funzionamento dei medesimi e l'altro, tipico del maschio, era già noto e qui se ne conferma la funzione di ghiandola a feromone sessuale maschile
Sexual size differences and colour polymorphism of Rhynchophorus phoenicis in the Southwest region of Cameroon
A total of 174 specimens of the African palm weevil Rhynchophorus phoenicis (F.) were sampled during 2010, in eight different
localities of the Southwest region of Cameroon. Male and female weevils were measured (body length, abdomen length, abdomen
width, pronotum length, pronotum width, head size, and length from tip of rostrum to antennal insertion), and the different pronotum
patterns are described. Statistical analysis shows that all morphometric parameters correlate with body length in both males
and females, and seven pronotum pattern types are identified, three of which are new. African palm weevil adults show sexual
dimorphism: abdomen and head size are significantly greater in females, while pronotum length is significantly greater in males.
No statistical differences in the frequencies of pronotum pattern types were seen between males and females, or among localities.
A significant difference was seen for the abdomen width between two localities
Exploitation of Tenebrio molitor larvae as biological factories for human probiotics, an exploratory study
The exploitation of yellow mealworm (Tenebrio molitor) larvae for the bioaugmentation of probiotic Bacillus
clausii strains was evaluated during a 7-day rearing period. qPCR was applied to evaluate the persistence
and growth of B. clausii in the rearing substrate and larvae (washed and non-washed). Moreover, the effect
of freeze-drying of larvae on B. clausii viability was evaluated. The results demonstrated the suitability of
yellow mealworm as biological factories for the multiplication of B. clausii through a simple and inexpensive
procedure, in view of the further application of larvae as foods and food ingredients. In more detail, an increase
in the load of B. clausii was observed during the 7-day rearing of larvae fed wheat middlings
spiked with 1 Log cells g−1. Further research is needed to evaluate the most suitable technologies and processing
parameters for obtaining yellow mealworm-based ingredients with a stable and active population of
probiotic B. clausii
Microbial dynamics in rearing trials of Hermetia illucens larvae fed coffee silverskin and microalgae
In the present study, Hermetia illucens larvae were reared on a main rearing substrate composed of a coffee
roasting byproduct (coffee silverskin, Cs) enriched with microalgae (Schizochytrium limacinum or Isochrysis galbana)
at various substitution levels. The microbial diversity of the rearing substrates, larvae, and frass (excrement
from the larvae mixed with the substrate residue) were studied by the combination of microbial culturing on
various growth media and metataxonomic analysis (Illumina sequencing). High counts of total mesophilic aerobes,
bacterial spores, presumptive lactic acid bacteria, coagulase-positive cocci, and eumycetes were detected.
Enterobacteriaceae counts were low in the rearing diets, whereas higher counts of this microbial family were
observed in the larvae and frass. The microbiota of the rearing substrates was characterized by the presence of
lactic acid bacteria, including the genera Lactobacillus, Leuconostoc and Weissella. The microbiota of the H. illucens
larvae fed Cs was characterized by the dominance of Paenibacillus. H. illucens fed diets containing I. galbana were
characterized by the presence of Enterococcus, Lysinibacillus, Morganella, and Paenibacillus, depending on the
algae inclusion level, while H. illucens fed diets containing S. limacinum were characterized by high relative
abundances of Brevundimonas, Enterococcus, Paracoccus, and Paenibacillus, depending on the algae inclusion level.
Brevundimonas and Alcaligenes dominated in the frass from larvae fed I. galbana; the predominance of Brevundimonas
was also observed in the frass from larvae fed Schyzochitrium-enriched diets. Based on the results of
the present study, an effect of algae nutrient bioactive substances (e.g. polysaccharides, high-unsaturated fatty
acids, taurine, carotenoids) on the relative abundance of some of the bacterial taxa detected in larvae may be
hypothesized, thus opening new intriguing perspectives for the control of the entomopathogenic species and
foodborne human pathogens potentially occurring in edible insects. Further studies are needed to support this
hypothesis. Finally, new information on the microbial diversity occurring in insect frass was also obtained
Association of acid phosphatase locus 1*C allele with the risk of cardiovascular events in rheumatoid arthritis patients
Introduction: Acid phosphatase locus 1 (ACP1) encodes a low molecular weight phosphotyrosine phosphatase
implicated in a number of different biological functions in the cell. The aim of this study was to determine the
contribution of ACP1 polymorphisms to susceptibility to rheumatoid arthritis (RA), as well as the potential
contribution of these polymorphisms to the increased risk of cardiovascular disease (CV) observed in RA patients.
Methods: A set of 1,603 Spanish RA patients and 1,877 healthy controls were included in the study. Information
related to the presence/absence of CV events was obtained from 1,284 of these participants. All individuals were
genotyped for four ACP1 single-nucleotide polymorphisms (SNPs), rs10167992, rs11553742, rs7576247, and
rs3828329, using a predesigned TaqMan SNP genotyping assay. Classical ACP1 alleles (*A, *B and *C) were imputed
with SNP data.
Results: No association between ACP1 gene polymorphisms and susceptibility to RA was observed. However,
when RA patients were stratified according to the presence or absence of CV events, an association between
rs11553742*T and CV events was found (P = 0.012, odds ratio (OR) = 2.62 (1.24 to 5.53)). Likewise, the ACP1*C
allele showed evidence of association with CV events in patients with RA (P = 0.024, OR = 2.43).
