    Epoxysuccinyl peptide-derived cathepsin B inhibitors: Modulating membrane permeability by conjugation with the C-terminal heptapeptide segment of penetratin

    Besides its physiological role in lysosomal protein breakdown, extralysosomal cathepsin B has recently been implicated in apoptotic cell death. Highly specific irreversible cathepsin B inhibitors that are readily cellpermeant should be useful tools to elucidate the effects of cathepsin B in the cytosol. We have covalently functionalised the poorly cellpermeant epoxysuccinyl based cathepsin B inhibitor [RGlyGlyLeu(2S, 3S)tEpsLeuProOH; R=OMe] with the C-terminal heptapeptide segment of penetratin (R=εAhxArg ArgNleLysTrpLysLysNH(2)). The high inhibitory potency and selectivity for cathepsin B versus cathepsin L of the parent compound was not affected by the conjugation with the penetratin heptapeptide. The conjugate was shown to efficiently penetrate into MCF-7 cells as an active inhibitor, thereby circumventing an intracellular activation step that is required by other inhibitors, such as the prodruglike epoxysuccinyl peptides E64d and CA074Me

    Strategische Allianzen, Zu- und Verkäufe, die Schaffung neuer Produktionsanlagen und sowie Produktionsverlagerungen als unternehmerische Handlungsfelder füllen die Tagespresse. Unternehmen rechtfertigen ihren Aktivismus oft mit der Erschließung neuer Märkte und die Konzentration auf Kerngeschäftsfelder. Das verleitet dazu, anzunehmen, dass der Personalbestand konzernweit steigt (um weitere Märkte abzudecken), er sich nur dadurch verändert, dass Unternehmen zu- und verkaufen oder Geschäftsfelder aufgeben und Zukäufe nur in den deklarierten Kerngeschäftsfeldern stattfinden. Diese Annahmen bilden die Grundlage der vorliegenden Marktanalyse, die folgende Informationen erhebt:Wie haben sich die Beschäftigtenzahlen der einzelnen DAX 30-Unternehmen zwischen den Jahren 2000 und 2006 tatsächlich verändert? Und - sofern dies aus den Geschäftsberichten hervorgeht - welche Zu- bzw. Verkäufe sind in diesem Zeitraum von den einzelnen Unternehmen getätigt und wie viele Arbeitnehmer damit transferiert worden? Der Text hat diese Zahlen akribisch herausgefiltert und interpretiert. Er liefert einen - teils verblüffenden - Eindruck in die Entwicklung der Beschäftigtenzahlen über einen Zeitraum von 6 Jahren

    The Metallicity of the Intracluster Medium Over Cosmic Time: Further Evidence for Early Enrichment

    We use Chandra X-ray data to measure the metallicity of the intracluster medium (ICM) in 245 massive galaxy clusters selected from X-ray and Sunyaev-Zel'dovich (SZ) effect surveys, spanning redshifts 0<z<1.20<z<1.2. Metallicities were measured in three different radial ranges, spanning cluster cores through their outskirts. We explore trends in these measurements as a function of cluster redshift, temperature, and surface brightness "peakiness" (a proxy for gas cooling efficiency in cluster centers). The data at large radii (0.5--1 r500r_{500}) are consistent with a constant metallicity, while at intermediate radii (0.1-0.5 r500r_{500}) we see a late-time increase in enrichment, consistent with the expected production and mixing of metals in cluster cores. In cluster centers, there are strong trends of metallicity with temperature and peakiness, reflecting enhanced metal production in the lowest-entropy gas. Within the cool-core/sharply peaked cluster population, there is a large intrinsic scatter in central metallicity and no overall evolution, indicating significant astrophysical variations in the efficiency of enrichment. The central metallicity in clusters with flat surface brightness profiles is lower, with a smaller intrinsic scatter, but increases towards lower redshifts. Our results are consistent with other recent measurements of ICM metallicity as a function of redshift. They reinforce the picture implied by observations of uniform metal distributions in the outskirts of nearby clusters, in which most of the enrichment of the ICM takes place before cluster formation, with significant later enrichment taking place only in cluster centers, as the stellar populations of the central galaxies evolve.Comment: 13 pages.
