Molecular Identification and Sequencing of Mannose Binding Protein (MBP) Gene of Acanthamoeba palestinensis
"nBackground: Acanthamoeba keratitis develops by pathogenic Acanthamoeba such as A. palestinensis. Indeed this species is one of the known causative agents of amoebic keratitis in Iran. Mannose Binding Protein (MBP) is the main pathogenicity factors for developing this sight threatening disease. We aimed to characterize MBP gene in pathogenic Acanthamoeba isolates such as A. palestinensis."nMethods: This experimental research was performed in the School of Public Health, Tehran University of Medical Sciences, Tehran, Iran during 2007-2008. A. palestinensis was grown on 2% non-nutrient agar overlaid with Escherichia coli. DNA extraction was performed using phenol-chloroform method. PCR reaction and amplification were done using specific primer pairs of MBP. The amplified fragment were purified and sequenced. Finally, the obtained fragment was deposited in the gene data bank."nResults: A 900 bp PCR-product was recovered after PCR reaction. Sequence analysis of the purified PCR product revealed a gene with 943 nucleotides. Homology analysis of the obtained sequence showed 81% similarity with the available MBP gene in the gene data bank. The fragment was deposited in the gene data bank under accession number EU678895"nConclusion: MBP is known as the most important factor in Acanthamoeba pathogenesis cascade. Therefore, characterization of this gene can aid in developing better therapeutic agents and even immunization of high-risk people
First Report of Vannellidae Amoebae (Vannella Spp.) Isolated From Biofilm Source
Background: Members of the Vannellidae family are free-living amoebae (FLA) distributed mainly in water and soil sources. The present study reports the first isolation of this genus in the biofilm source from hospital environment in Tehran, Iran.Methods: Biofilm samples were collected from hospital environment. Cultivation was performed in non-nutrient agar covered with a heat-killed Escherichia coli. Cloning of the suspected amoebae was done. PCR amplification and Homology analysis using the Basic Local Alignment Search Tool (BLASTn) was performed to search for the most similar reference sequences.Results: Microscopic examination showed numerous fan-shaped amoebae and peculiar cysts different to the usual shape of typical FLA. Sequence analysis of the PCR- product revealed that the suspected amoebae are highly homologous with Vannella spp. gene (99% identity and 100% query coverage) available in the gene bank database.Conclusion: Although Vannella spp. is not proved to be pathogenic itself, but they are capable of harboring pathogenic intracellular organisms such as Microsporidian parasites. Thus, identification of such amoebae can be of clinical importance, as they could lead to transmission of other pathogens to human
Anti-Acanthamoeba effects of silver and gold nanoparticles and contact lenses disinfection solutions
Background: This study aimed to investigate the anti-Acanthamoeba effects of the most used marketed disinfecting solutions in Iran. Moreover, the efficacy of some nano-compounds was tested against pathogenic Acanthamoeba. Methods: The present study was conducted in the School of Public Health, Tehran University of Medical sciences, Tehran, Iran during 2015-2016. Cysts of Acanthamoeba T4 genotype (7 x 104/ml) mixed at the equal volume with contact lens solutions including Opti-free, Ginza, ReNu, Maxima, Light, and Cyclean for the recommended time by the manufacturers. Nano-silver and nano-gold compounds were also treated with the amoebae. Chlorhexidine 0.02 and normal saline were used as positive and negative controls, respectively. Dead and alive amoebae were determined using vital stain and suspension was cultured in non-nutrient agar. The entire process was repeated at least three times. Results: In none of the solutions in the manufacturer's brochure recommended time, full cytotoxic effect was observed on the cysts of Acanthamoeba. Opti free express solution destroyed the cysts after 6 days. Nanosilver and nano-gold compounds showed no cytotoxic effect on the cysts of Acanthamoeba. Conclusion: None of the Nanoparticles compounds as well as contact lenses disinfecting solutions which studied was effective on Acanthamoeba cysts in the manufacturer's brochure recommended time. However, continuing study on Nano-silver and Nano-gold compounds to find effective ingredients against Acanthamoeba are highly recommended. © 2018, Tehran University of Medical Sciences (TUMS). Unusual Vermamoeba Vermiformis Strain Isolated from Snow in Mount Teide, Tenerife, Canary Islands, Spain
Background: Free-living amoebae (FLA) are protozoa that are widely distributed in the environment mainly in water and soil related habitats. Thermophilic amoebae are among the most common FLA in water bodies, being Vermamoeba vermiformis one of the most common species reported worldwide from these sources. Interestingly, V. vermiformis has often been reported to survive at high temperatures and osmotic pressure worldwide.Materials and Methods: In this study, snow samples were collected from Mount Teide, Tenerife, Canary Islands during the winter season of 2014. The samples were culture on non nutrient agar plates and checked daily for the presence of FLA. After a week, V. vermiformis amoebae were observed in the plates incubated at room temperature and 37ºC.Results: Molecular characterization was carried out by amplifying the 18S rDNA gene and DNA sequencing, confirmed that the isolated strain belonged to Vermamoeba vermiformis species.Conclusion: To the best of our knowledge, this is the first report of Vermamoeba vermiformis isolation from such an inusual habitat (low temperatures and high altitude) and the first report of these species in the Canary islands
First Report of Vermamoeba vermiformis in the Island of El Hierro, Canary Islands, Spain
Background: Free-living amoebae (FLA) are group of protozoa distributed worldwide in many habitats mainly water and soil related sources. Some members of FLA are able to act as opportunistic pathogens and are environmental carriers of other pathogenic agents such as bacteria and viruses. Vermamoeba vermiformis is a highly abundant FLA species in water bodies and has recently gained environmental importance as it acts as a vehicle of many pathogenic bacteria such as Legionella pneumophila.Cases Report: In this study, water samples were collected from the island of El Hierro, Canary Islands, Spain during 2015. El Hierro island was designated by UNESCO as a biosphere reserve and it is currently the less populated of the Canary Islands. The water samples were culture on 2 % Non-Nutrient Agar (NNA) plates covered with a thin layer of heat killed E. coli and checked daily for the presence of FLA. After a week, V. vermiformis amoebae were observed in the plates incubated at room temperature and 37 ºC. Molecular characterization was carried out by amplifying the 18S rDNA gene and DNA sequencing, confirming that the isolated strain belonged to Vermamoeba vermiformis species.Conclusion: To the best of our knowledge, this is the first report of Vermamoeba vermiformis isolation in the island of El Hierro and the second report of this species in the Canary Islands
Isolation of Acanthamoeba spp. from Different Environmental Sources
Background: Acanthamoeba spp. are free-living amebas found in a wide variety of natural habitats. The high percentage of Acanthamoeba in different environmental sources represents a sanitary risk for public health especially contact lens users and immunocompromised patients. The aim of this study was to determine the presence of Acanthamoeba spp. in different environments such as water, soil, dust and ophthalmology wards. Methods: From March to November 2007, 80 samples were collected from numerous localities in Tehran city including university campus, Laleh park and ophthalmology center. Sample types were water, soil, dust, cow faeces and medical instrument. Each sample was filtered through nitrate membrane and cultured on 1% non-nutrient agar. These plates were followed up daily for 2 weeks. Monitoring continued for two months on a weekly basis. Results: Overall, 46.25% of samples contained Acanthamoeba spp. All of the soil samples had shown positive culture in contrast to tap water. Of 61 dust samples, 28 were positive. Interestingly, we were able to isolate Acanthamoeba in treatment unit of an ophthalmology center in Tehran. It should be mentioned that two cow faeces showed positive culture as well.Conclusion: The widespread distribution of Acanthamoeba spp. across the environmental sources and increasing numbers of HIV+ patients and contact lens wearers, as well as its ability as a pathogen carrier for humans, demands more awareness and knowledge for public as a risk for human health
Isolation and genotyping of Acanthamoeba spp. from recreational soil of parks in Tehran, Iran
Background & aim: Acanthamoeba is a genus of free-living amoebae found in environmental sources. These amphizoic amoebae can lead to severe human disease such as encephalitis and keratitis. Acanthamoeba transmits to humans through contact with soil and dust from scratching the skin. The aim of the present study was to identify the genotypes of Acanthamoeba in parks of the city of Tehran using molecular and morphological - based methods.
Methods: In this study, 52 samples of soil were collected from 17 parks in Tehran. Samples were then filtered and cultured on 1.5% non-nutrient agar. DNA extraction and PCR amplification was performed using genus specific primers. Sequencing analysis and BLAST search were done for genotype identification.
Results: Out of 52 soil samples, 14 strain (26.9%) were positive for Acanthamoeba amoebae by microscopic observation. Out Of 14 positive isolates, 9 (17.3%) were positive for Acanthamoeba using genus specific primer pairs. Of 14 strains, 9 were sequenced successfully. Genotype identification was revealed that all strains were belonged to T4 type. T4 genotypes among strains are human pathogens.
Conclusions: Identification of pathogenic Acanthamoeba belonging to T4 genotype in recreational parks could be of utmost importance. Results of this study show that soil contamination, particularly in parks where children play and assemble is a sanitary risk for them.
