    An assessment of the mechanical strengths of aluminide-based thin coatings

    Titanium aluminide and nickel aluminide-based thin coatings were synthesized by magnetron sputtering from intermetallic TiAl and Ni3Al alloy targets on nickel substrates. Both types of aluminide coating exhibited high surface hardness values that varied with the degree of heat treatment. The hardness of the coatings was investigated using micro- and nano- indentation techniques. In order to estimate the intrinsic strength of the films, the indentation size effects of the apparent hardness were analyzed by the Jönsson-Hogmark model and a model recently proposed by the authors. The analysis indicated that the strengths of the aluminide coatings may considerably exceed their strengths in bulk.published_or_final_versio

    p21-Activated Kinases 1, 2 and 4 in Endometrial Cancers: Effects on Clinical Outcomes and Cell Proliferation

    p21-activated kinases (Paks) are serine/threonine protein kinases involved in biological events linked to malignant tumor progression. In this study, expression of Pak1, p-Pak2 Ser20, Pak4, pPak4 Ser474 in 21 normal endometrium, 16 hyperplastic endometrium without atypia, 17 atypical complex hyperplasia and 67 endometrial cancers was assessed by immunohistochemistry and correlated with clinicopathological parameters. We also accessed the proliferative role and downstream targets of Pak1 in endometrial cancer. Pak1 was expressed in cytoplasm whereas Pak4 and p-Pak4 were expressed in both cytoplasm and nucleus of endometrial tissues. In normal endometrium, significantly higher Pak1 (P = 0.028) and cytoplasmic p-Pak2 (P = 0.048) expression was detected in proliferative endometrium than secretory endometrium. Pak1, cytoplasmic and nuclear Pak4 and nuclear p-Pak4 was significantly overexpressed in endometrial cancer when compared to atrophic endometrium (all P<0.05). Moreover, type I endometrioid carcinomas showed significantly higher Pak1 expression than type II non-endometrioid carcinomas (P<0.001). On the other hand, Pak1, Pak4 and p-Pak4 expression negatively correlated with histological grade (all P<0.05) while p-Pak2 and cytoplasmic Pak4 expression inversely correlated with myometrial invasion (all P<0.05). Furthermore, patients with endometrial cancers with lower cytoplasmic Pak4 expression showed poorer survival (P = 0.026). Multivariate analysis showed cytoplasmic Pak4 is an independent prognostic factor. Functionally, knockdown of Pak1, but not Pak4, in endometrial cancer cell line led to reduced cell proliferation along with reduced cyclin D1, estrogen receptor (ERα) and progestogen receptor (PR) expression. Significant correlation between Pak1 and PR expression was also detected in clinical samples. Our findings suggest that Pak1 and cytoplasmic p-Pak2 may promote cell proliferation in normal endometrium during menstral cycle. Pak1, cytoplasmic and nuclear Pak4 and nuclear p-Pak4 are involved in the pathogenesis of endometrial cancer especially in postmenopausal women. Pak1 promote endometrial cancer cell proliferation, particular in type I endometrioid carcinoma. Cytoplasmic Pak4 can be potential prognostic marker in endometrial cancer.published_or_final_versio

    Functional polymorphisms in the BRCA1 promoter influence transcription and are associated with decreased risk for breast cancer in Chinese women

    Background: The BRCA1 gene is an important breast-cancer susceptibility gene. Promoter polymorphisms can alter the binding affinity of transcription factors, changing transcriptional activity and may affect susceptibility to disease. Methods and Results: Using direct sequencing of the BRCA1 promoter region, we identified four polymorphisms c.-2804T→C (rs799908:T→C), c.-2265C→T (rs11655505:C→T), c.-2004A→G (rs799906:A→G) and c.-1896(ACA) 1→(ACA) 2 (rs8176071:(ACA) 1→(ACA) 2) present in Hong Kong Chinese. Each polymorphism was studied independently and in combination by functional assays. Although all four variants significantly altered promoter activity, the c.-2265T allele had stronger binding than the C allele, and the most common mutant haplotype, which contains the c.-2265T allele, increased promoter activity by 70%. Risk association first tested in Hong Kong Chinese women with breast cancer and age-matched controls and replicated in a large population-based study of Shanghai Chinese, together totalling >3000 participants, showed that carriers of the c.-2265T allele had a reduced risk for breast cancer (combined odd ratio (OR) = 0.80, 95% Cl 0.69 to 0.93; p = 0.003) which was more evident among women aged ≥45 years at first diagnosis of breast cancer and without a family history of breast cancer (combined OR = 0.75, 95% Cl 0.61 to 0.91; p = 0.004). The most common haplotype containing the c.-2265T allele also showed significant risk association for women aged ≥45 years without a family history of breast cancer (OR = 0.64, 95% Cl 0.46 to 0.89; p = 0.008). Conclusion: This comprehensive study of BRCA1 promoter polymorphisms found four variants that altered promoter activity and with the most significant contribution from c.-2265C→T, which could affect susceptibility to breast cancer in the Chinese population.     Clinical spectrum of Exophiala infections and a novel Exophiala species, Exophiala hongkongensis

    Poster PresentationBackground: Exophiala species are saprophytic fungi which have been isolated from environments rich in hydrocarbons or from hot, humid, and oligotrophic environments. These fungi are considered as dematiaceous moulds; and due to their phenotypic characteristics at the beginning of colony formation, they are also often referred to as ‘black yeasts’, a misnomer which sometimes may mislead the choice of antifungal agents. When the cultures mature, brown hyphae are formed bearing conidiogenous cells referred to as annellides, a typical characteristic of this fungal genus. Although Exophiala species are environmental fungi, they should not be disregarded as contaminants when they are isolated from clinical specimens. These fungi are causative agents of skin and subcutaneous tissue infections and of systemic infections, such as prosthetic valve endocarditis, dialysis-associated peritonitis, and disseminated infections, especially in immunocompromised patients. Unfortunately, Exophiala species can often only be identified to the genus level by phenotypic characterisation. Objectives: The aims of this study were to study the clinical spectrum of Exophiala infections in Queen Mary Hospital, Hong Kong by a polyphasic approach, and to characterise a potentially novel Exophiala species, Exophiala hongkongensis (ex-type strain HKU32T). Methods: All Exophiala strains characterised in this study were isolated from patients during a 15-year period (1998-2012) and were retrieved from the collection in the clinical microbiology laboratory at Queen Mary Hospital, Hong Kong. The strains were characterised phenotypically by microscopic examination of fungal structure using the agar block smear preparation method and phylogenetically using the internal transcribed spacer (ITS) region and Rpb1 gene. In addition, a unique strain, HKU32T, was further characterised phenotypically by scanning electron microscopy, enzyme activity test using the API-ZYM system, and growth tests on different temperatures and culture media. HKU32T was also further phylogenetically characterised using β-tubulin and β-actin genes. All the phylogenetic analyses were performed by the maximum likelihood method using MEGA 5.0.5. Results: Microscopic examination of the young cultures of all the 12 strains showed subspherical, budding, yeast-like cells. Sequencing of the ITS region and partial Rpb1 gene showed 11 of the 12 strains were known Exophiala species, including E. oligosperma [n = 3], E. jeanselmei [n = 2], E. lecanii-corni [n = 2], E. bergeri [n = 1], E. cancerae [n = 1], E. dermatitidis [n = 1], and E. xenobiotica [n = 1]). As for HKU32T, it displayed unique morphological features and was positive for eight enzymes in the API-ZYM test. Optimal growth was observed at 30°C on potato dextrose agar or at 24°C on cornmeal agar. HKU32T also occupied unique phylogenetic positions in all the phylogenetic analyses, with Exophiala nishimurae being the most closely related species. Clinical spectrum of Exophiala infections in Hong Kong included chronic skin infection, colonisation of gastrointestinal tract, continuous ambulatory peritoneal dialysis (CAPD) peritonitis, onychomycosis, pneumonia, tinea pedis, and wrist or finger nodule. Conclusion: Exophiala species could cause a wide range of infections and the most frequent species isolated from patients in Hong Kong was Exophiala oligosperma.     Overexpression of proto-oncogene FBI-1 activates membrane type 1-matrix metalloproteinase in association with adverse outcome in ovarian cancers

    <p>Abstract</p> <p>Background</p> <p>FBI-1 (factor that binds to the inducer of short transcripts of human immunodeficiency virus-1) is a member of the POK (POZ and Kruppel) family of transcription factors and play important roles in cellular differentiation and oncogenesis. Recent evidence suggests that FBI-1 is expressed at high levels in a subset of human lymphomas and some epithelial solid tumors. However, the function of FBI-1 in human ovarian cancers remains elusive.</p> <p>Results</p> <p>In this study, we investigated the role of FBI-1 in human ovarian cancers, in particularly, its function in cancer cell invasion via modulating membrane type 1-matrix metalloproteinase (MT1-MMP). Significantly higher FBI-1 protein and mRNA expression levels were demonstrated in ovarian cancers samples and cell lines compared with borderline tumors and benign cystadenomas. Increased FBI-1 mRNA expression was correlated significantly with gene amplification (P = 0.037). Moreover, higher FBI-1 expression was found in metastatic foci (P = 0.036) and malignant ascites (P = 0.021), and was significantly associated with advanced stage (P = 0.012), shorter overall survival (P = 0.032) and disease-free survival (P = 0.016). <it>In vitro</it>, overexpressed FBI-1 significantly enhanced cell migration and invasion both in OVCA 420 and SKOV-3 ovarian carcinoma cells, irrespective of <it>p53 </it>status, accompanied with elevated expression of MT1-MMP, but not MMP-2 or TIMP-2. Moreover, knockdown of MT1-MMP abolished FBI-1-mediated cell migration and invasion. Conversely, stable knockdown of FBI-1 remarkably reduced the motility of these cells with decreased expression of MT1-MMP. Promoter assay and chromatin immunoprecipitation study indicated that FBI-1 could directly interact with the promoter spanning ~600bp of the 5'-flanking sequence of MT1-MMP and enhanced its expression in a dose-dependent manner. Furthermore, stable knockdown and ectopic expression of FBI-1 decreased and increased cell proliferation respectively in OVCA 420, but not in the p53 null SKOV-3 cells.</p> <p>Conclusions</p> <p>Our results suggested an important role of FBI-1 in ovarian cancer cell proliferation, cell mobility, and invasiveness, and that FBI-1 can be a potential target of chemotherapy.</p

    Enhancing biogas production by anaerobic codigestion of water hyacinth and pig manure

    The characteristics of anaerobic batch co-digestion of water hyacinth (WH) with pig manure (PM) under seven mixing ratio 100%WH; 80%WH : 20%PM; 60%WH : 40%PM; 50%WH : 50%PM; 40%WH : 60%PM; 20%WH : 80%PM and 100%PM were investigated, each treatment was conducted in five replications with daily loading rate at 1 gVS.L-1.day-1. During the anaerobic digestion process of 60 days, maximum biogas production occurred in two periods, the first stage from 12- 22 days and second stage from 30 - 35 days. The maximum daily biogas productions from each stage were 17.2 L.day-1 and 15.1 L.day-1, respectively. The cumulative biogas production varied between 60 L (100%PM) and 360 L (60%WH : 40%PM). The results showed that the biogas yields of co-digestion 40- 80%WH were higher from 34.6 to 56.1% in comparison with 100%PM and from 109 to 143% in comparison with 100%WH. When mixing with WH, treatments were received more methane and the methane contents were higher than 45% (v/v) that good for energy using purposes.Nghiên cứu được thực hiện nhằm khảo sát khả năng gia tăng lượng khí sinh học khi tiến hành đồng phân hủy yếm khí lục bình (WH) và phân heo (PM) ở các tỉ lệ phối trộn khác nhau gồm 100%WH; 80%WH : 20%PM; 60%WH : 40%PM; 50%WH : 50%PM; 40%WH : 60%PM; 20%WH : 80%PM và 100%PM. Các nghiệm thức được nạp lượng nguyên liệu là 1 gVS.L-1.ngày-1 và bố trí lặp lại 5 lần. Theo dõi quá trình phân hủy của các nghiệm thức trong 60 ngày ghi nhận có 2 khoảng thời gian lượng khí sản sinh nhiều nhất - giai đoạn 1 từ ngày 12 đến 22, giai đoạn 2 từ ngày 30 đến 35. Lượng khí sản sinh cao nhất tương ứng trong mỗi giai đoạn là 17.2 L.ngày-1 và 15.1 L.ngày-1. Lượng khí tích lũy trong suốt thời gian thí nghiệm ghi nhận thấp nhất ở nghiệm thức 100%PM đạt 60 L, và cao nhất ở nghiệm thức 60%WH : 40%PM đạt 360 L. Năng suất khí sinh ra của các nghiệm thức phối trộn lục bình từ 40 đến 80% cao hơn từ 34,6 đến 56,1% so với nghiệm thức 100%PM và cao hơn từ 109% đến 143% so với nghiệm thức 100%WH. Hàm lượng mê-tan sinh ra từ các nghiệm thức có phối trộn lục bình ổn định trong khoảng > 45% đảm bảo nhiệt lượng cho nhu cầu sử dụng năng lượng
