    Simultaneous multiple responses modelling, optimisation and correlation of Asian type peanuts (Arachis hypogaea L.) roasting using response surface methodology

    The effect of hot-air roasting temperatures and time on colour (L*), moisture content (% d.b.), hardness (N), and fracturability (mm) attributes of two different type of peanuts (China and India origin) were investigated using response surface methodology (RSM). The central composite design (CCD) was used to investigate temperatures ranging from 130–170 °C (China) and 130–200 °C (India), while roasting time was from 20–80 min (China) and 15–50 min (India). The results revealed that temperature and time have significant effect on all the responses. Increase in roasting time and temperature caused a decrease in all the responses for both peanuts. A feasible experimental condition of peanut roasting obtained from the optimisation of simultaneous multiple attributes’ response for the China and India peanuts was 152 °C for 60 min and 158 °C for 45 min, respectively. The strongest linearrelationship was found between hardness and fracturability, while the weakest was between colour and fracturability for both peanuts

    Effect of temperature on rheology behaviour of banana peel pectin extracted using hot compressed water

    Banana peel pectin is extracted from banana peel waste using a hot compressed water extraction (140-160°C, 5 minutes, 1.18 mm particle size). Physicochemical contents of banana peel pectin were found to be in a similar range with commercial pectin, and is comprised of moisture (7.44-8.47%), ash (3.45-4.98%), protein (1.08-1.92%), fat (0.04-3.42), carbohydrate (83-86%), total sugar (1.77-3.41%), energy (353-369 kcal/100g) and specific heat (1.42-1.62 kJ/kg°C). These contents possibly related to their flow deformation of rheological behaviour. Regression analysis displayed good agreements in all models applied, apart from the Casson Model. Flow behaviour indices, n<1 and decreasing of apparent viscosity within increasing of shear rate indicates that banana peel pectin has excellent shear thinning behaviour with a presence of yield stress

    Activated carbon and halloysite nanotubes membrane for CO2 and CH4 separation

    In this study, mixed matrix membranes (MMMs) were prepared where activated carbon and halloysite nanotubes with a loading of 1 wt% is incorporated into the polysulfone membrane, and the performance of each membrane was investigated. The morphological properties, mechanical strength and their correlations with the gas separation performance for CO2 and CH4 for halloysite-mixed matrix membrane (MMMs-HNT) and activated carbon-mixed matrix membrane (MMMs-AC) was studied by using Scanning Electron Microscopy (SEM-EDX), tensile test and gas permeation test. From the characterization of MMMs, SEM shows an increase of 30.77% on the thickness of the dense layer of MMMs-HNTs compared to neat membrane and MMMs-AC. The EDX results also showed that HNTs evenly distributed in the polymeric matrix without any sign of agglomeration. Elongation at the break for MMMs-HNTs also decreases to 11.38%. The gas separation performance for MMMs-HNTs increased by about 55.43% compared to MMMs-AC at 2 bar. Furthermore, MMMs-HNTs also showed in increase in the selectivity of membrane towards CO2 and CH4 from 0.82 to 15.83 at 2 bar. In conclusion, the addition of 1 wt% of HNTs into PSF polymeric matrix showed a better permeance of CO2 and a greater selectivity compared to the neat membrane and the MMMs-AC and thus is the optimum inorganic filler for the mixed matrix membrane

    Graphene oxide deposition on neodymium doped zinc borotellurite glass surface: optical and polarizability study for future fiber optics

    Neodymium oxide doped tellurite-based glass has been widely documented for potential uses in optoelectronics, but graphene oxide (GO)-coated tellurite-based glass has rarely been reported. In this work, we compare two sets of glass series which were GO-coated and uncoated tellurite-based glass series denoted as ZBTNd-GO and ZBTNd, respectively. The two sets of glasses were fabricated via melt-quenched process. A set of glass was coated with GO using low-cost spray coating method. The structural and morphological properties of the glass samples were investigated to confirm the type of structure in glass and formation of graphene oxide on glass surface. The X-ray diffraction (XRD) pattern confirmed the amorphous structural arrangement in both sets of glass series. The morphological study proved the existence of GO layers on top of the ZBTNd-GO surface. The optical bandgap energy of ZBTNd-GO glass was found in the range of 3.253 eV–3.381 eV which was higher than ZBTNd glass. Meanwhile, the refractive index of ZBTNd-GO glass varies from 2.301 to 2.332 which was higher than ZBTNd glass due to the presence of functionalized oxygenated groups in GO structure. The oxide ion polarizability of ZBTNd-GO glass was found decreased due to the shift of optical band gap when coated with GO. This work offers a new form of glass that could be used as a new strategy to upgrade the current photonic materials

    Sentinel surveillance for human enterovirus 71 in Sarawak, Malaysia: lessons from the first 7 years

    BACKGROUND: A major outbreak of human enterovirus 71-associated hand, foot and mouth disease in Sarawak in 1997 marked the beginning of a series of outbreaks in the Asia Pacific region. Some of these outbreaks had unusually high numbers of fatalities and this generated much fear and anxiety in the region. METHODS: We established a sentinel surveillance programme for hand, foot and mouth disease in Sarawak, Malaysia, in March 1998, and the observations of the first 7 years are described here. Virus isolation, serotyping and genotyping were performed on throat, rectal, vesicle and other swabs. RESULTS: During this period Sarawak had two outbreaks of human enterovirus 71, in 2000 and 2003. The predominant strains circulating in the outbreaks of 1997, 2000 and 2003 were all from genogroup B, but the strains isolated during each outbreak were genetically distinct from each other. Human enterovirus 71 outbreaks occurred in a cyclical pattern every three years and Coxsackievirus A16 co-circulated with human enterovirus 71. Although vesicles were most likely to yield an isolate, this sample was not generally available from most cases and obtaining throat swabs was thus found to be the most efficient way to obtain virological information. CONCLUSION: Knowledge of the epidemiology of human enterovirus 71 transmission will allow public health personnel to predict when outbreaks might occur and to plan interventions in an effective manner in order to reduce the burden of disease

    Investigation of highly unsaturated fatty acid metabolism in the Asian sea bass, Lates calcarifer

    Lates calcarifer, commonly known as the Asian sea bass or barramundi, is an interesting species that has great aquaculture potential in Asia including Malaysia and also Australia. We have investigated essential fatty acid metabolism in this species, focusing on the endogenous highly unsaturated fatty acid (HUFA) synthesis pathway using both biochemical and molecular biological approaches. Fatty acyl desaturase (Fad) and elongase (Elovl) cDNAs were cloned and functional characterization identified them as ∆6 Fad and Elovl5 elongase enzymes, respectively. The ∆6 Fad was equally active towards 18:3n-3 and 18:2n-6, and Elovl5 exhibited elongation activity for C18-20 and C20-22 elongation and a trace of C22-24 activity. The tissue profile of gene expression for ∆6 fad and elovl5 genes, showed brain to have the highest expression of both genes compared to all other tissues. The results of tissue fatty acid analysis showed that the brain contained more docosahexaenoic acid (DHA, 22:6n-3) than flesh, liver and intestine. The HUFA synthesis activity in isolated hepatocytes and enterocytes using [1-14C]18:3n-3 as substrate was very low with the only desaturated product detected being 18:4n-3. These findings indicate that L. calcarifer display an essential fatty acid pattern similar to other marine fish in that they appear unable to synthesize HUFA from C18 substrates. High expression of ∆6 fad and elovl5 genes in brain may indicate a role for these enzymes in maintaining high DHA levels in neural tissues through conversion of 20:5n-3

    Functional Desaturase Fads1 (Δ5) and Fads2 (Δ6) Orthologues Evolved before the Origin of Jawed Vertebrates

    Long-chain polyunsaturated fatty acids (LC-PUFAs) such as arachidonic (ARA), eicosapentaenoic (EPA) and docosahexaenoic (DHA) acids are essential components of biomembranes, particularly in neural tissues. Endogenous synthesis of ARA, EPA and DHA occurs from precursor dietary essential fatty acids such as linoleic and α-linolenic acid through elongation and Δ5 and Δ6 desaturations. With respect to desaturation activities some noteworthy differences have been noted in vertebrate classes. In mammals, the Δ5 activity is allocated to the Fads1 gene, while Fads2 is a Δ6 desaturase. In contrast, teleosts show distinct combinations of desaturase activities (e.g. bifunctional or separate Δ5 and Δ6 desaturases) apparently allocated to Fads2-type genes. To determine the timing of Fads1-Δ5 and Fads2-Δ6 evolution in vertebrates we used a combination of comparative and functional genomics with the analysis of key phylogenetic species. Our data show that Fads1 and Fads2 genes with Δ5 and Δ6 activities respectively, evolved before gnathostome radiation, since the catshark Scyliorhinus canicula has functional orthologues of both gene families. Consequently, the loss of Fads1 in teleosts is a secondary episode, while the existence of Δ5 activities in the same group most likely occurred through independent mutations into Fads2 type genes. Unexpectedly, we also establish that events of Fads1 gene expansion have taken place in birds and reptiles. Finally, a fourth Fads gene (Fads4) was found with an exclusive occurrence in mammalian genomes. Our findings enlighten the history of a crucially important gene family in vertebrate fatty acid metabolism and physiology and provide an explanation of how observed lineage-specific gene duplications, losses and diversifications might be linked to habitat-specific food web structures in different environments and over geological timescales

    Cloning, Functional Characterization and Nutritional Regulation of Delta 6 Fatty Acyl Desaturase in the Herbivorous Euryhaline Teleost Scatophagus Argus

    Marine fish are generally unable or have low ability for the biosynthesis of long-chain polyunsaturated fatty acids (LC-PUFA) from C18 PUFA precursors, with some notable exceptions including the herbivorous marine teleost Siganus canaliculatus in which such a capability was recently demonstrated. To determine whether this is a unique feature of S. canaliculatus or whether it is common to the herbivorous marine teleosts, LC-PUFA biosynthetic pathways were investigated in the herbivorous euryhaline Scatophagus argus. A putative desaturase gene was cloned and functionally characterized, and tissue expression and nutritional regulation were investigated. The full-length cDNA was 1972 bp, containing a 1338 bp open-reading frame encoding a polypeptide of 445 amino acids, which possessed all the characteristic features of fatty acyl desaturase (Fad). Functional characterization by heterologous expression in yeast showed the protein product of the cDNA efficiently converted 18:3n-3 and 18:2n-6 to 18:4n-3 and 18:3n-6, respectively, indicating D6 desaturation activity. Quantitative real-time PCR showed that highest D6 fad mRNA expression was detected in liver followed by brain, with lower expression in other tissues including intestine, eye, muscle, adipose, heart kidney and gill, and lowest expression in stomach and spleen. The expression of D6 fad was significantly affected by dietary lipid and, especially, fatty acid composition, with highest expression of mRNA in liver of fish fed a diet with a ratio of 18:3n-3/18:2n-6 of 1.72:1. The results indicated that S. argus may have a different LC-PUFA biosynthetic system from S. canaliculatus despite possessing similar habitats and feeding habits suggesting that LC-PUFA biosynthesis may not be common to all marine herbivorous teleosts
