    Molecular genetic methods for identifying raw materials in meat products: Diversity, opportunities and prospects

    In the current economic situation, after easing the Covid pandemic restrictions, almost all laboratories, which are focused on evaluation of the conformity of food products, have faced issues in supplying for their laboratories. In this regard, in the last years many laboratories have been forced to validate new approaches and introduce new methods for assessing conformity of the food products. Very often it is not possible to use only one method to resolve the issue of the food product ingredients, especially for the purpose of traceability of their names and the used raw materials, listed on the label. Survey of the raw food materials to determine whether they correspond to the type name is a simpler task, in contrast to survey of the multicomponent food product. Many researchers have to estimate the opportunities and feasibility of application of various methodologies in their workplaces. Therefore, this review is relevant for the researchers in this field, as it focuses on aspects and special features of similar methodologies. The prospect of molecular genetic methods for identification of the raw materials used for manufacturing of meat products is presented below. This review also represents characteristics of methods for identification of the sources of raw materials used for the manufacturing of the meat products, based on the recognition of species-specific sections within the nucleic acids structures. The variety of methods (hybridization methods, polymerase chain reaction, different types of isothermal amplifications, methods using CRISPR/Cas systems), the principles of their implementation, and achieved analytical characteristics are considered.     Comamonas testosteroni strain TI as a potential base for a microbial sensor detecting surfactants

    Strain Comamonas testosteroni TI, capable of degrading the nonionic surfactant (NIS) nonylphenolethoxylate (OP-10), was used for constructing a pilot cellular biosensor. The lower NIS detection limit for the biosensor was 0.25 mg/l. We studied the substrate specificity of the biosensor with respect to a wide range of organic compounds: surfactants, polyaromatic compounds (PAC), carbohydrates, alcohols, etc. It was shown that the biosensor based on Comamonas testosteroni TI did not respond to glucose, which was an advantage over the formerly described biosensor based on Pseudomonas rathonis T. The amplitude of the sensor response remained stable for 10 days

    The effect of spelt flour on the quality of puff pastry

    Croissant is one of the puff pastry products. This product is widely distributed in France. The assortment of croissants is very diverse, but in order to increase the assortment and improve the quality of this product, spelt grain flour has been used in our research. The purpose of the research is to study the effect of spelt flour of the Alcoran variety on the quality of puff pastry. The tasks are to consider the production technology of croissant; to investigate the organoleptic and physical and chemical parameters of croissant; to calculate the economic efficiency of the production of croissant. The methods used are the following: the regulatory framework of the research is the legislation of the Russian Federation on standardization and certification, regulatory documents (GOST, technical specifications). The assessment of organoleptic and physical and chemical parameters has been carried out according to the technical specifications 10.71.11-046-38826547-2016. The results are: trial baking has been conducted in the Volgograd State Agrarian University at the Department of Processing Technologies and Food Safety. Puff yeast dough for Croissant is divided into two main technological processes: kneading yeast dough; lamination of dough. The appearance of the croissant with spelt flour is vague, the color is brown, there is a taste of «nutty flavor», sweeter. The conclusion: when calculating the profitability of croissant production, it has been found that the raw material costs of spelt flour products are 1.6 times higher, which is associated with the cost of spelt grain flour. The calculation of profitability has shown an increase in costs by 2% in the production of croissants with spelt flour. The production of spelt culture refers to organic farming. Healthy natural products lead to an increase in the cost of raw materials, which cannot but affect the cost of further products

    Methodology for assessing the agroecological adaptability of genotypes under global climate warming

    Background. The relevance of the research is due to the fact that, despite the breeding achievements in releasing cultivars with high potential yield and resistance to the most common stressors in the regions, it is almost impossible to foresee and accurately predict the reaction of cultivars under production conditions. Besides, extreme weather events, including heat waves, droughts and downpours, will increasingly manifest themselves under global climate warming. To reduce the risk of a significant decrease in gross grain harvests in dry years and achieve high results under favorable conditions, the goal was to develop a comprehensive indicator for assessing the agroecological adaptability of genotypes to some consequences of global climate warming.Materials and methods. The results of a competitive test of 5 cultivars and 9 promising lines of spring wheat for their yield across six years contrasting in moisture availability by calculating the indicators of responsiveness to favorable conditions, grain yield depression under unfavorable conditions and the relative yield index were used to develop a complex indicator proposed to assess the agroecological adaptability of a genotype to contrasting environmental conditions.Results. The proposed method makes it possible to perform a comprehensive assessment that takes into account the responsiveness of an accession to improved agroclimatic conditions and the degree of its reaction to adverse environmental factors, taking into account the yield index in the studied set. The method is recommended for evaluating the source material and identifying the diversity of its responses to changes in agroclimatic conditions as well as for the stage of competitive variety testing in plant breeding for adaptability

    Highly Sensitive Immunochromatographic Identification of Tetracycline Antibiotics in Milk

    A rapid immunochromatographic assay was developed for the control of tetracycline (TC). The assay is based on the competition between immobilized TC-protein conjugate and TC in a tested sample for binding with polyclonal anti-TC antibodies conjugated to colloidal gold during the flow of the sample along a membrane strip with immobilized reactants. Conjugation of colloidal gold and the total immunoglobulin (IgG) fraction of polyclonal antibodies was used to increase the assay sensitivity to ensure low content of specific antibodies in the conjugate. This allowed effective inhibition of free TC and conjugate binding in the strip test zone. Photometric marker registration allows control of the reduction of binding, thereby enhancing detection sensitivity. The proposed assay allows TC to be detected at concentrations up to 20 ng/mL, exceeding the limit of detection of the known analogues, in a wide working range (more than two orders) of 60 pg/mL to 10 ng/mL, ensured through the use of polyclonal antibodies. The assay time is 10 min. The efficiency of the designed assay is shown to identify TC in milk; the degree of recovery of TC ranges from 90 to 112%.
