Novel Approach to Increase the Energy-related Process Efficiency and Performance of Laser Brazing
AbstractAlthough laser brazing is well established, the energy-related efficiency of this joining method is quite low. That is because of low absorptivity of solid-state laser radiation, especially when copper base braze metals are used. Conventionally the laser beam is set close to the vertical axis and the filler wire is delivered under a flat angle. Therefore, the most of the utilized laser power is reflected and thus left unexploited. To address this situation an alternative processing concept for laser brazing, where the laser beam is leading the filler wire, has been investigated intending to make use of reflected shares of the laser radiation. Process monitoring shows, that the reflection of the laser beam can be used purposefully to preheat the substrate which is supporting the wetting and furthermore increasing the efficiency of the process. Experiments address a standard application from the automotive industry joining zinc coated steels using CuSi3Mn1 filler wire. Feasibility of the alternative processing concept is demonstrated, showing that higher processing speeds can be attained, reducing the required energy per unit length while maintaining joint properties
Spin-orbiton and quantum criticality in FeSc2S4
In FeSc2S4 spin-orbital exchange competes with strong spin-orbit coupling,
suppressing long-range spin and orbital order and, hence, this material
represents one of the rare examples of a spin-orbital liquid ground state.
Moreover, it is close to a quantum-critical point separating the ordered and
disordered regimes. Using THz and FIR spectroscopy we study low-lying
excitations in FeSc2S4 and provide clear evidence for a spin-orbiton, an
excitation of strongly entangled spins and orbitals. It becomes particularly
well pronounced upon cooling, when advancing deep into the quantum-critical
regime. Moreover, indications of an underlying structureless excitation

Coloring Mixed and Directional Interval Graphs
A mixed graph has a set of vertices, a set of undirected egdes, and a set ofdirected arcs. A proper coloring of a mixed graph is a function thatassigns to each vertex in a positive integer such that, for each edge in , and, for each arc in , G\chi(G)G$. A directional interval graph is a mixedgraph whose vertices correspond to intervals on the real line. Such a graph hasan edge between every two intervals where one is contained in the other and anarc between every two overlapping intervals, directed towards the interval thatstarts and ends to the right. Coloring such graphs has applications in routing edges in layered orthogonalgraph drawing according to the Sugiyama framework; the colors correspond to thetracks for routing the edges. We show how to recognize directional intervalgraphs, and how to compute their chromatic number efficiently. On the otherhand, for mixed interval graphs, i.e., graphs where two intersecting intervalscan be connected by an edge or by an arc in either direction arbitrarily, weprove that computing the chromatic number is NP-hard.<br
Time to Surgery Does Not Affect Overall or Disease-Free Survival of Patients with Primary Resectable PDAC
(1) Background: Delay in therapy for pancreatic ductal adenocarcinoma (PDAC) may contribute to a worse outcome. The aim of this study was to investigate the prognostic value of time from diagnosis to surgery in patients undergoing upfront surgery for primarily resectable pancreatic carcinoma. (2) Methods: This retrospective single-center study included 214 patients who underwent primary resection of PDAC from January 2000 to December 2018 at University Hospital Erlangen. Using a minimum p-value approach, patients were stratified according to time to surgery (TtS) into two groups: TtS ≤ 23 days and TtS > 23 days. Postoperative outcome and long-term survival were compared. (3) Results: Median TtS was 25 days. The best cut-off for TtS was determined as 23 days. There were no differences regarding postoperative outcome or overall survival (OS) and disease-free survival (DFS) (OS: 23.8 vs. 20.4 months, p = 0.210, respectively, and DFS: 15.8 vs. 13.6 months, p = 0.187). Multivariate analysis revealed age, lymph node metastasis, tumor differentiation and resection status as significant independent prognostic predictors for OS and DFS. (4) Conclusions: A delay of surgery > 23 days after first diagnosis does not affect overall or disease-free survival of patients with primary resectable PDAC. However, the psychological impact of a delay to patients waiting for surgery should not be underestimated
Colorectal cancer in Crohn’s colitis is associated with advanced tumor invasion and a poorer survival compared with ulcerative colitis: a retrospective dual-center study
Abstract
Purpose
Colorectal cancer is a well-recognized complication of inflammatory bowel diseases (IBD), such as ulcerative colitis (UC) and Crohn’s colitis (CC). In this study, we assess the clinico-pathological features and outcomes of patients with colorectal cancer from UC in comparison with CC.
Methods
Data of all patients with colitis-associated cancer (CAC) who underwent surgery at Erlangen or Würzburg University Clinic between 1995 and 2015 were selected. Clinical, histopathological, and survival data were analyzed retrospectively.
Results
Of all 88 patients with CAC, 20 patients had Crohn’s colitis and 68 patients had ulcerative colitis. We observed a young median age at tumor diagnosis (49.5 years UC; 45.5 years CC, p = 0.208) in both diseases and a long median disease duration before CAC (19 years UC; 18 years CC; p = 0.840). Patients with CC suffered more often from rectal cancer (14 (70.0%) in CC; 23 (33.8%) in UC; p = 0.005) and advanced tumor stages (8 (47.0%) pT4 in CC; 14 (25.0%) pT4/ypT4 in UC; p = 0.008). Five-year overall survival rate was 39.3% for CC and 67.1% for UC (p = 0.009 for difference between the groups). Survival did not differ significantly between UC and CC in the multivariate analysis after correction for UICC tumor stage.
Conclusion
CAC in CC showed advanced tumor stages associated with reduced survival compared with CAC in UC. This may be explained by less intense surveillance in patients with CC leading to delayed cancer diagnosis
Interleukin-3 protects against viral pneumonia in sepsis by enhancing plasmacytoid dendritic cell recruitment into the lungs and T cell priming
Rationale
Sepsis, a global health burden, is often complicated by viral infections leading to increased long-term morbidity and mortality. Interleukin-3 (IL-3) has been identified as an important mediator amplifying acute inflammation in sepsis; however, its function in the host response to viral infections during sepsis remains elusive.
Objectives
To investigate the role of IL-3 during viral pneumonia in sepsis.
Methods
We included septic patients from two different cohorts and used in vitro and in vivo assays. The obtained data were substantiated using a second model (SARS-CoV-2 infections).
Measurements and main results
Low plasma IL-3 levels were associated with increased herpes simplex virus (HSV) airway infections in septic patients, resulting in reduced overall survival. Likewise, Il-3-deficient septic mice were more susceptible to pulmonary HSV-1 infection and exhibited higher pulmonary inflammation than control mice. Mechanistically, IL-3 increases innate antiviral immunity by promoting the recruitment of circulating plasmacytoid dendritic cells (pDCs) into the airways and by enhancing pDC-mediated T cell activation upon viral stimulation. Interestingly, the ability of IL-3 to improve adaptive immunity was confirmed in patients with SARS-CoV-2 infections.
Conclusion
Our study identifies IL-3 as a predictive disease marker for viral reactivation in sepsis and reveals that IL-3 improves antiviral immunity by enhancing the recruitment and the function of pDCs
Exosomal ROR1 in peritoneal fluid identifies peritoneal disseminated PDAC and is associated with poor survival
BackgroundPancreatic ductal adenocarcinoma (PDAC) is one of the deadliest forms of cancer and peritoneal dissemination is one major cause for this poor prognosis. Exosomes have emerged as promising biomarkers for gastrointestinal cancers and can be found in all kinds of bodily fluids, also in peritoneal fluid (PF). This is a unique sample due to its closeness to gastrointestinal malignancies. The receptor tyrosine kinase-like orphan receptor 1 (ROR1) has been identified as a potential biomarker in human cancers and represents a promising target for an immunotherapy approach, which could be considered for future treatment strategies. Here we prospectively analyzed the exosomal surface protein ROR1 (exo-ROR1) in PF in localized PDAC patients (PER-) on the one hand and peritoneal disseminated tumor stages (PER+) on the other hand followed by the correlation of exo-ROR1 with clinical-pathological parameters.MethodsExosomes were isolated from PF and plasma samples of non-cancerous (NC) (n = 15), chronic pancreatitis (CP) (n = 4), localized PDAC (PER-) (n = 18) and peritoneal disseminated PDAC (PER+) (n = 9) patients and the surface protein ROR1 was detected via FACS analysis. Additionally, soluble ROR1 in PF was analyzed. ROR1 expression in tissue was investigated using western blots (WB), qPCR, and immunohistochemistry (IHC). Exosome isolation was proven by Nano Tracking Analysis (NTA), WB, Transmission electron microscopy (TEM), and BCA protein assay. The results were correlated with clinical data and survival analysis was performed.ResultsPDAC (PER+) patients have the highest exo-ROR1 values in PF and can be discriminated from NC (p <0.0001), PDAC (PER-) (p <0.0001), and CP (p = 0.0112). PDAC (PER-) can be discriminated from NC (p = 0.0003). In plasma, exo-ROR1 is not able to distinguish between the groups. While there is no expression of ROR1 in the exocrine pancreatic tissue, PDAC and peritoneal metastasis show expression of ROR1. High exo-ROR1 expression in PF is associated with lower overall survival (p = 0.0482).ConclusionWith exo-ROR1 in PF we found a promising diagnostic and prognostic biomarker possibly discriminating between NC, PDAC (PER-) and PDAC (PER+) and might shed light on future diagnostic and therapeutic concepts in PDAC
