    Taxonomy and evolutionary relationships of Passiflora subg. Decaloba supersect. Decaloba sect. Xerogona (Passifloraceae): contributions of palynological, morphological and molecular studies

    Passiflora subg. Decaloba supersect. Decaloba sect. Xerogona (Passifloraceae) is a tropical and subtropical group comprising 14 species that occur in tropical biomes throughout Latin America, including the Atlantic Forest. The section Xerogona comprises herbaceous vines characterized by a lack of petiole glands on their leaves, of bracts and of ocelli on their leaf blades, as well as by their capsular fruits. We analyzed the phylogeny on the basis of morphological characters (including pollen characters) and molecular data. The inferred phylogeny was used in order to characterize, circumscribe and delimit the section and the species. We examined the phylogenetic relationships among the species, evaluating the circumscription of the section on the basis of the trnL-trnF intergenic spacer region of chloroplast DNA and the internal transcribed spacer region of nuclear ribosomal DNA. We constructed phylogenetic trees on the basis of the morphological and molecular data. We found that P. subg. Decaloba supersect. Decaloba sect. Xerogona appears to be monophyletic only in the molecular analyses. The phylogenetic analyses performed here also indicated that P. subg. Decaloba is monophyleti

    Trapping of normal EB1 ligands in aggresomes formed by an EB1 deletion mutant

    Background EB1 is a microtubule tip-associated protein that interacts with the APC tumour suppressor protein and the p150glued subunit of dynactin. We previously reported that an EB1 deletion mutant that retains both of these interactions but does not directly associate with microtubules (EB1-ΔN2-GFP) spontaneously formed perinuclear aggregates when expressed in COS-7 cells. Results In the present study live imaging indicated that EB1-ΔN2-GFP aggregates underwent dynamic microtubule-dependent changes in morphology and appeared to be internally cohesive. EB1-ΔN2-GFP aggregates were phase-dense structures that displayed microtubule-dependent accumulation around the centrosome, were immunoreactive for both the 20s subunit of the proteasome and ubiquitin, and induced the collapse of the vimentin cytoskeleton. Fractionation studies revealed that a proportion of EB1-ΔN2-GFP was detergent-insoluble and ubiquitylated, indicating that EB1-ΔN2-GFP aggregates are aggresomes. Immunostaining also revealed that APC and p150glued were present in EB1-ΔN2-GFP aggregates, whereas EB3 was not. Furthermore, evidence for p150glued degradation was found in the insoluble fraction of EB1-ΔN2-GFP transfected cultures. Conclusion Our data indicate that aggresomes can be internally cohesive and may not represent a simple "aggregate of aggregates" assembled around the centrosome. Our observations also indicate that a partially misfolded protein may retain the ability to interact with its normal physiological ligands, leading to their co-assembly into aggresomes. This supports the idea that the trapping and degradation of co-aggregated proteins might contribute to human pathologies characterised by aggresome formation

    The snail density, levels of infection and the monthly production of Schistosoma mansoni cercariae by Biomphalaria glabrata were determined in a focus of Barreiro de Baixo (Belo Horizonte, MG, Brazil). During a period of 38 months (1984 to 1987) 5,366 snails were collected of which 324 (6.03%) were infected with S. mansoni. The total number of cercariae shed was 5,667,312. Each snail shed an average of 17,422 cercariae during the time that it was under study in the laboratory. The greatest longevity of infected snails was 218 days. Natural cure was observed in 42 (12.9%) of the infected specimens about 130 days after collection. The average snail density in the focus during the period of study was 16.3 snails per scoop. The shedding of cercariae by snails collected from the field was compared with laboratory bred specimens infected in mass with the LE strain of S. mansoni from Belo Horizonte. The laboratory infected snails shed an average of 6,061 cercariae each, a value 2.8 times less than the field specimens due to a shorter life span. The prevalence of schistosomiasis in the focus was 14.3%.<br>A densidade planorbídica, as taxas de infecção e a produção mensal de cercárias de Schistosoma mansoni por Biomphalaria glabrata, foram determinadas em foco do Barreiro de Baixo (Belo Horizonte MG, Brasil). Durante 38 meses, de 1984 a 1987, foram capturados 5.366 moluscos dentre os quais 324 (6,03%) estavam infectados com o S. mansoni. O total de cercárias eliminadas foi de 5.667.312. Cada molusco eliminou em média 17.422 cercárias durante sua permanência no laboratório. A maior longevidade dos moluscos infectados foi de 218 dias. Foi observada a cura natural de 42 (12,9%) dos exemplares infectados, após cerca de 130 dias de captura. A densidade planorbídica média, no foco durante esse período foi de 16,3 moluscos por conchada. A eliminação de cercárias pelos moluscos do campo foi comparada com a de exemplares criados no laboratório, infectados em massa com a linhagem LE de S. mansoni de Belo Horizonte. Os moluscos do laboratório eliminaram em média 6.061 cercárias por exemplar, número 2,8 vezes menor do que os moluscos do campo, devido a longevidade menor. A taxa de prevalência da esquistossomose no foco foi de 14,3%

    Procurou-se observar o desenvolvimento pós-embrionário de Chrysomya albiceps, testando-se, comparativamente, dietas à base de sardinha e carne eqüina. Os substratos frescos foram previamente mantidos a 30&deg;C de temperatura durante duas horas. Foram inoculadas 50 neolarvas em 100g de dieta por repetição. Utilizaram-se quatro repetições por tratamento. O experimento foi conduzido em condições de laboratório. O peso médio das larvas após o abandono espontâneo das dietas foi de 91,19mg, no substrato à base de carne eqüina e 76,01mg, no substrato à base de sardinha, a diferença foi, portanto, significativa. A taxa de sobrevivência larval registrada foi superior a 80%, enquanto a taxa de sobrevivência pupal foi próxima a 100%. A razão sexual foi próxima a 0,5. A dieta à base de carne eqüina mostrou-se mais adequada ao desenvolvimento de C. albiceps, pois as larvas mantidas neste substrato apresentaram-se mais pesadas, o que potencializa a capacidade reprodutiva do adulto.<br>The objective of the present study was to observe the postembryonic development of Chrysomya albiceps reared on sardines or horse meat and to compare the results. Fresh substratos were maintened m an oven ai 30&deg;C for 2 hours. Fifty newly emerged larvas were inoculated into 100g diel per reptication. Four replications per treatment were used. The experiment was conducted under laboratory conditions. The mean weight of postfeeding larvas was 91.19mg for especimens reared on horse meat and 76.01mg for especimens reared on sardines, with a marked difference between diets. Larval viability was more than 80% for the two diets, while pupal viability was dose to 100%. The sex ratio was about 0.5. The horse meat diet proved to be more adequate for the development of C. albiceps since the larvas weighed more, with consequent potentiations of adult reproductive abilily
