    Topological Reorganization of Odor Representations in the Olfactory Bulb

    Odors are initially represented in the olfactory bulb (OB) by patterns of sensory input across the array of glomeruli. Although activated glomeruli are often widely distributed, glomeruli responding to stimuli sharing molecular features tend to be loosely clustered and thus establish a fractured chemotopic map. Neuronal circuits in the OB transform glomerular patterns of sensory input into spatiotemporal patterns of output activity and thereby extract information about a stimulus. It is, however, unknown whether the chemotopic spatial organization of glomerular inputs is maintained during these computations. To explore this issue, we measured spatiotemporal patterns of odor-evoked activity across thousands of individual neurons in the zebrafish OB by temporally deconvolved two-photon Ca2+ imaging. Mitral cells and interneurons were distinguished by transgenic markers and exhibited different response selectivities. Shortly after response onset, activity patterns exhibited foci of activity associated with certain chemical features throughout all layers. During the subsequent few hundred milliseconds, however, MC activity was locally sparsened within the initial foci in an odor-specific manner. As a consequence, chemotopic maps disappeared and activity patterns became more informative about precise odor identity. Hence, chemotopic maps of glomerular input activity are initially transmitted to OB outputs, but not maintained during pattern processing. Nevertheless, transient chemotopic maps may support neuronal computations by establishing important synaptic interactions within the circuit. These results provide insights into the functional topology of neural activity patterns and its potential role in circuit function

    Dynamical Systems in Categories

    In this article we establish a bridge between dynamical systems, including topological and measurable dynamical systems as well as continuous skew product flows and nonautonomous dynamical systems; and coalgebras in categories having all finite products. We introduce a straightforward unifying definition of abstract dynamical system on finite product categories. Furthermore, we prove that such systems are in a unique correspondence with monadic algebras whose signature functor takes products with the time space. We substantiate that the categories of topological spaces, metrisable and uniformisable spaces have exponential objects w.r.t. locally compact Hausdorff, σ-compact or arbitrary time spaces as exponents, respectively. Exploiting the adjunction between taking products and exponential objects, we demonstrate a one-to-one correspondence between monadic algebras (given by dynamical systems) for the left-adjoint functor and comonadic coalgebras for the other.     The evolution and structure of ignited high-pressure cryogenic hydrogen jets

    The anticipated upscaling of hydrogen energy applications will involve the storage and transport of hydrogen at cryogenic conditions. Understanding the potential hazard arising from leaks in high-pressure cryogenic storage is needed to improve hydrogen safety. The manuscript reports a series of numerical simulations with detailed chemistry for the transient evolution of ignited high-pressure cryogenic hydrogen jets. The study aims to gain insight of the ignition processes, flame structures and dynamics associated with the transient flame evolution. Numerical simulations were firstly conducted for an unignited jet released under the same cryogenic temperature of 80 K and pressure of 200 bar as the considered ignited jets. The predicted hydrogen concentrations were found to be in good agreement with the experimental measurements. The results informed the subsequent simulations of the ignited jets involving four different ignition locations. The predicted time series snapshots of temperature, hydrogen mass fraction and the flame index are analyzed to study the transient evolution and structure of the flame. The results show that a diffusion combustion layer is developed along the outer boundary of the jet and a side diffusion flame is formed for the near-field ignition. For the far-field ignition, an envelope flame is observed. The flame structure contains a diffusion flame on the outer edge and a premixed flame inside the jet. Due to the complex interactions between turbulence, fuel-air mixing at cryogenic temperature and chemical reactions, localized spontaneous ignition and transient flame extinguishment are observed. The predictions also captured the experimentally observed deflagration waves in the far-field ignited jets

    Myocardial aging as a T-cell–mediated phenomenon

    In recent years, the myocardium has been rediscovered under the lenses of immunology, and lymphocytes have been implicated in the pathogenesis of cardiomyopathies with different etiologies. Aging is an important risk factor for heart diseases, and it also has impact on the immune system. Thus, we sought to determine whether immunological activity would influence myocardial structure and function in elderly mice. Morphological, functional, and molecular analyses revealed that the age-related myocardial impairment occurs in parallel with shifts in the composition of tissue-resident leukocytes and with an accumulation of activated CD4+ Foxp3- (forkhead box P3) IFN-γ+ T cells in the heart-draining lymph nodes. A comprehensive characterization of different aged immune-deficient mouse strains revealed that T cells significantly contribute to age-related myocardial inflammation and functional decline. Upon adoptive cell transfer, the T cells isolated from the mediastinal lymph node (med-LN) of aged animals exhibited increased cardiotropism, compared with cells purified from young donors or from other irrelevant sites. Nevertheless, these cells caused rather mild effects on cardiac functionality, indicating that myocardial aging might stem from a combination of intrinsic and extrinsic (immunological) factors. Taken together, the data herein presented indicate that heart-directed immune responses may spontaneously arise in the elderly, even in the absence of a clear tissue damage or concomitant infection.     Impaired Axonal Transport in Motor Neurons Correlates with Clinical Prion Disease

    Prion diseases are fatal neurodegenerative disorders causing motor dysfunctions, dementia and neuropathological changes such as spongiosis, astroglyosis and neuronal loss. The chain of events leading to the clinical disease and the role of distinct brain areas are still poorly understood. The role of nervous system integrity and axonal properties in prion pathology are still elusive. There is no evidence of both the functional axonal impairments in vivo and their connection with prion disease. We studied the functional axonal impairments in motor neurons at the onset of clinical prion disease using the combination of tracing as a functional assay for axonal transport with immunohistochemistry experiments. Well-established and novel confocal and ultramicroscopy techniques were used to image and quantify labeled neurons. Despite profound differences in the incubation times, 30% to 45% of neurons in the red nucleus of different mouse lines showed axonal transport impairments at the disease onset bilaterally after intracerebral prion inoculation and unilaterally—after inoculation into the right sciatic nerve. Up to 94% of motor cortex neurons also demonstrated transport defects upon analysis by alternative imaging methods. Our data connect axonal transport impairments with disease symptoms for different prion strains and inoculation routes and establish further insight on the development of prion pathology in vivo. The alterations in localization of the proteins involved in the retrograde axonal transport allow us to propose a mechanism of transport disruption, which involves Rab7-mediated cargo attachment to the dynein-dynactin pathway. These findings suggest novel targets for therapeutic and diagnostic approaches in the early stages of prion disease

    Dimerization of melanocortin 4 receptor controls puberty onset and body size polymorphism

    Xiphophorus fish exhibit a clear phenotypic polymorphism in puberty onset and reproductive strategies of males. In X. nigrensis and X. multilineatus, puberty onset is genetically determined and linked to a melanocortin 4 receptor (Mc4r) polymorphism of wild-type and mutant alleles on the sex chromosomes. We hypothesized that Mc4r mutant alleles act on wild-type alleles by a dominant negative effect through receptor dimerization, leading to differential intracellular signaling and effector gene activation. Depending on signaling strength, the onset of puberty either occurs early or is delayed. Here, we show by Förster Resonance Energy Transfer (FRET) that wild-type Xiphophorus Mc4r monomers can form homodimers, but also heterodimers with mutant receptors resulting in compromised signaling which explains the reduced Mc4r signaling in large males. Thus, hetero- vs. homo- dimerization seems to be the key molecular mechanism for the polymorphism in puberty onset and body size in male fish

    CAST constraints on the axion-electron coupling

    In non-hadronic axion models, which have a tree-level axion-electron interaction, the Sun produces a strong axion flux by bremsstrahlung, Compton scattering, and axiorecombination, the "BCA processes." Based on a new calculation of this flux, including for the first time axio-recombination, we derive limits on the axion-electron Yukawa coupling gae and axion-photon interaction strength ga using the CAST phase-I data (vacuum phase). For ma <~ 10 meV/c2 we find ga gae < 8.1 × 10−23 GeV−1 at 95% CL. We stress that a next-generation axion helioscope such as the proposed IAXO could push this sensitivity into a range beyond stellar energy-loss limits and test the hypothesis that white-dwarf cooling is dominated by axion emission

    Autochthonous heritage languages and social media:writing and bilingual practices in Low German on Facebook

    This article analyses how speakers of an autochthonous heritage language (AHL) make use of digital media, through the example of Low German, a regional language used by a decreasing number of speakers mainly in northern Germany. The focus of the analysis is on Web 2.0 and its interactive potential for individual speakers. The study therefore examines linguistic practices on the social network site Facebook, with special emphasis on language choice, bilingual practices and writing in the autochthonous heritage language. The findings suggest that social network sites such as Facebook have the potential to provide new mediatized spaces for speakers of an AHL that can instigate sociolinguistic change
