    University Students’ Preferences for Labour Conditions at a Mining Site: Evidence from Two Australian Universities

    The mining industry makes up a large portion of the gross domestic product (GDP) in Australia, although securing human resources remains a problem in that field. The aim of this paper is to identify Australian university mining students’ preferences, considering it as potential employees’ preferences, for labour conditions at mining sites by means of a discrete choice experiment to promote efficient improvements in labour conditions in the mining industry. The data of 93 respondents analysed in this paper was collected by survey carried out in two universities in Australia. The result of the study showed that students have preferences on several factors such as wage, fatality rate, working position, commuting style, and company. Students having specific sociodemographic characters were found to show specific preferences on labour conditions. The results of this study indicate the potential average of appropriate monetary compensation for each factor

    Nontypeable Haemophilus influenzae induces COX-2 and PGE2 expression in lung epithelial cells via activation of p38 MAPK and NF-kappa B

    <p>Abstract</p> <p>Background</p> <p>Nontypeable <it>Haemophilus influenzae </it>(NTHi) is an important respiratory pathogen implicated as an infectious trigger in chronic obstructive pulmonary disease, but its molecular interaction with human lung epithelial cells remains unclear. Herein, we tested that the hypothesis that NTHi induces the expression of cyclooxygenase (COX)-2 and prostaglandin E2 (PGE2) via activation of p38 mitogen-activated protein kinase (MAPK) and nuclear factor (NF)-kappa B in pulmonary alveolar epithelial cells.</p> <p>Methods</p> <p>Human alveolar epithelial A549 cells were infected with different concentrations of NTHi. The phosphorylation of p38 MAPK was detected by Western blot analysis, the DNA binding activity of NF-kappa B was assessed by electrophoretic mobility shift assay (EMSA), and the expressions of COX-1 and 2 mRNA and PGE2 protein were measured by reverse transcription-polymerase chain reaction (RT-PCR) and enzyme linked immunosorbent assay (ELISA), respectively. The roles of Toll-like receptor (TLR) 2 and TLR4, well known NTHi recognizing receptor in lung epithelial cell and gram-negative bacteria receptor, respectively, on the NTHi-induced COX-2 expression were investigated in the HEK293 cells overexpressing TLR2 and TLR4 <it>in vitro </it>and in the mouse model of NTHi-induced pneumonia by using TLR2 and TLR4 knock-out mice <it>in vivo</it>. In addition, the role of p38 MAPK and NF-kappa B on the NTHi-induced COX-2 and PGE2 expression was investigated by using their specific chemical inhibitors.</p> <p>Results</p> <p>NTHi induced COX-2 mRNA expression in a dose-dependent manner, but not COX-1 mRNA expression in A549 cells. The enhanced expression of PGE2 by NTHi infection was significantly decreased by pre-treatment of COX-2 specific inhibitor, but not by COX-1 inhibitor. NTHi induced COX-2 expression was mediated by TLR2 in the epithelial cell <it>in vitro </it>and in the lungs of mice <it>in vivo</it>. NTHi induced phosphorylation of p38 MAPK and up-regulated DNA binding activity of NF-kappa B. Moreover, the expressions of COX-2 and PGE2 were significantly inhibited by specific inhibitors of p38 MAPK and NF-kappa B. However, NTHi-induced DNA binding activity of NF-kappa B was not affected by the inhibition of p38 MAPK.</p> <p>Conclusion</p> <p>NTHi induces COX-2 and PGE2 expression in a p38 MAPK and NF-kappa B-dependent manner through TLR2 in lung epithelial cells <it>in vitro </it>and lung tissues <it>in vivo</it>. The full understanding of the role of endogenous anti-inflammatory PGE2 and its regulation will bring new insight to the resolution of inflammation in pulmonary bacterial infections.</p

    Failure Mechanisms and Local Scour at Coastal Structures induced by Tsunamis

    On March 11 2011, an exceptionally large tsunami event was triggered by a massive earthquake offshore, the northeast coast of Japan, which affected coastal infrastructure such as seawalls, coastal dikes and breakwaters in the Tohoku region. Such infrastructure was built to protect against the Level 1 tsunamis that previously hit the region, but not for events as significant as the 2011 Tohoku tsunami, which was categorized as a Level 2 tsunami [Shibayama et al. 2013]. The failure mechanisms of concrete-armoured dikes, breakwaters and seawalls due to Level 2 tsunamis are still not fully understood by researchers and engineers. This paper investigates the failure modes and mechanisms of damaged coastal structures in Miyagi and Fukushima Prefectures, following the authors' post-disaster field surveys carried out between 2011 and 2013. Six significant failure mechanisms were identified for the coastal dikes and seawalls affected by this tsunami: 1) Leeward toe scour failure, 2) Crown armour failure, 3) Leeward slope armour failure, 4) Seaward toe and armour failure, 5) Overturning failure, and 6) Parapet wall failure, in which leeward toe scour being recognized as the major failure mechanism in most surveyed locations. The authors also propose a simple practical mathematical model for predicting the scour depth at the leeward toe of the coastal dikes, by considering the effects of the tsunami hydrodynamics, the soil properties and the type of structure. The key advantage of this model is that it depends entirely on quantities that are measurable in the field. Furthermore this model was further refined by conducting a series of hydraulic model experiments aimed to understand the governing factors of the leeward toe scour failure. Finally, based on the results obtained, key recommendations are given for the design of resilient coastal defence structures that can survive a level 2 tsunami event

    Activation of Epidermal Growth Factor Receptor Is Required for NTHi-Induced NF-ÎșB-Dependent Inflammation

    Inflammation is a hallmark of many serious human diseases. Nontypeable Haemophilus influenzae (NTHi) is an important human pathogen causing respiratory tract infections in both adults and children. NTHi infections are characterized by inflammation, which is mainly mediated by nuclear transcription factor-kappa B (NF-ÎșB)-dependent production of proinflammatory mediators. Epidermal growth factor receptor (EGFR) has been shown to play important roles in regulating diverse biological processes, including cell growth, differentiation, apoptosis, adhesion, and migration. Its role in regulating NF-ÎșB activation and inflammation, however, remains largely unknown.In the present study, we demonstrate that EGFR plays a vital role in NTHi-induced NF-ÎșB activation and the subsequent induction of proinflammatory mediators in human middle ear epithelial cells and other cell types. Importantly, we found that AG1478, a specific tyrosine kinase inhibitor of EGFR potently inhibited NTHi-induced inflammatory responses in the middle ears and lungs of mice in vivo. Moreover, we found that MKK3/6-p38 and PI3K/Akt signaling pathways are required for mediating EGFR-dependent NF-ÎșB activation and inflammatory responses by NTHi.Here, we provide direct evidence that EGFR plays a critical role in mediating NTHi-induced NF-ÎșB activation and inflammation in vitro and in vivo. Given that EGFR inhibitors have been approved in clinical use for the treatment of cancers, current studies will not only provide novel insights into the molecular mechanisms underlying the regulation of inflammation, but may also lead to the development of novel therapeutic strategies for the treatment of respiratory inflammatory diseases and other inflammatory diseases

    Development and Validation of Virtual Reality Application in Mining Education

    Productivity, safety and its improvement is also an integral part of a good mining operation. In recent times, due to constraints on time and cost, it has become increasingly harder to conduct training and safety inductions at mine sites. For the purpose of overcoming these limitations, the use of virtual reality (VR) is proposed for mining education and training. VR has already been introduced in the education and training of miners overseas, and quantitative studies on the effects of using VR for miner&apos;s education and training have been made. However, Japan has only one such application of VR for mining education, namely, “Virtual Mining Practice System” which was produced by Akita University, and there are relatively few cases where VR has been introduced in the Japanese mining industry. Furthermore, there has been no quantitative study to date on the effects of education using VR for mining education. Therefore, the objective of this study is to investigate the effects of a class that utilizes a VR application developed for mining education (Mining VR), as well as evaluate its learning outcomes. In this study, a method called randomized controlled trial (RCT) is used for evaluating Mining VR&apos;s effectiveness. Study participants are divided two groups randomly where one class makes use of Mining VR and another class using other non-VR material. After the classes are completed, a test is conducted and the average results of each group are compared by T-test. The results of this experiment showed that there were no statistically significant differences in skill of “understanding” and “knowledge retention” comparing two groups. On the other hand, results suggested that Mining VR has improved students&apos;“ motivation” for class when using Mining VR

    Assessing Peruvian University Students’ Preferences for Labor Conditions in Mining Site

    The mining industry is one of the biggest industries and has a strong impact in Peru. Despite the mining industry’s importance, it faces labor shortages and environmental risks. For these situations, mining students are important sources of workers not only as labor forces but also as skilled workers who can contribute to solving the environmental issues of mining companies. Thus, the purpose of this paper is to assess Peruvian university mining students’ preferences for labor conditions in mining sites using a discrete choice experiment in order to promote efficient improvements in labor conditions in mining sites that contribute to stable employment and address environmental concerns. The number of respondents was 222 in two Peruvian universities, including males and females aged 16–35 years. The analysis’s findings indicate that labor conditions at mining sites can be optimized by adjusting them to specific individual characteristics of potential mining workers, resulting in a more efficient working environment for companies and workers
