    In-vitro application of pentoxifylline preserved ultrastructure of spermatozoa after vitrification in asthenozoospermic patients

    Abstract PURPOSE: To evaluate the effect of in vitro application of pentoxifylline (PX) on sperm parameters and ultrastructure after vitrification in asthenozoospermic patients. MATERIALS AND METHODS: A total of 30 asthenozoospermic semen samples (aged 25-45 years) were divided into four groups before vitrification, after vitrification, control (without PX) and experimental (with PX). In experimental group, each sample was exposed for 30 min to 3.6mmol/l PX and the control group without any treatment apposing in 370C for 30 min. After incubation, the samples were washed and analyzed again. Vitrification was done according to straw method. Eosin-nigrosin and Papanicolaou staining were applied for assessment of sperm viability and morphology, respectively. The samples without PX and post treatment with PX were assessed by transmission electron microscopy (TEM). RESULTS: A significant decrease in sperm motility (P ≤ .001), morphology (11.47 ± 2.9 versus 6.73 ± 2.01) and viability (73.37 ± 6.26 versus 54.67 ± 6.73) was observed post vitrification, but sperm motility (19.85 ± 4.75 versus 32.07 ± 5.58, P ≤ .001) was increased significantly following application of PX. This drug had no significant (P >.05) detrimental neither negative effect on ultrastructure acrosome, plasma membrane and coiled tail statues of spermatozoa. CONCLUSION: Vitrification had detrimental effects on sperm parameters, but PX reversed detrimental effects on sperm motility. However, PX had no alteration on ultrastructure morphology of human spermatozoa after vitrification

    The regulatory G4 motif of the Kirsten ras (KRAS) gene is sensitive to guanine oxidation: Implications on transcription

    KRAS is one of the most mutated genes in human cancer. It is controlled by a G4 motif located upstream of the transcription start site. In this paper, we demonstrate that 8-oxoguanine (8-oxoG), being more abundant in G4 than in non-G4 regions, is a new player in the regulation of this oncogene. We designed oligonucleotides mimicking the KRAS G4-motif and found that 8-oxoG impacts folding and stability of the G-quadruplex. Dimethylsulphate-footprinting showed that the G-run carrying 8-oxoG is excluded from the G-tetrads and replaced by a redundant G-run in the KRAS G4-motif. Chromatin immunoprecipitation revealed that the base-excision repair protein OGG1 is recruited to the KRAS promoter when the level of 8-oxoG in the G4 region is raised by H2O2. Polyacrylamide gel electrophoresis evidenced that OGG1 removes 8-oxoG from the G4-motif in duplex, but when folded it binds to the G-quadruplex in a non-productive way. We also found that 8-oxoG enhances the recruitment to the KRAS promoter of MAZ and hnRNP A1, two nuclear factors essential for transcription. All this suggests that 8-oxoG in the promoter G4 region could have an epigenetic potential for the control of gene expression

    Fatigue properties of nano-reinforced bituminous mixtures: A viscoelastic continuum damage approach

    The experimental investigation described in this paper focused on the effects of nanoclays on the fatigue behaviour of bituminous mixtures. Damage characteristics of a bituminous mixture produced by making use of a nano-reinforced binder were compared to those of a reference mixture obtained by employing the same neat bitumen used as a base in the preparation of the nanoclay–bitumen blend. Dynamic modulus tests and direct tension cyclic fatigue tests were carried out to determine the linear viscoelastic properties and the damage evolution characteristics of materials. Corresponding results were modelled by means of a viscoelastic continuum damage approach and by making use of a more empirical evaluation based on the classical Wöhler representation. It was found that the use of nanoclays produced a reinforcement of bituminous mixtures, the benefits of which were observed both in the progression of damage and in the occurrence of ultimate failure condition

    Chemically exfoliated graphene detects NO2 at the ppb level

    Abstract The high sensitivity of graphene to the adsorption/desorption of gas molecule, is at the very beginning of its exploitation. This sensitivity relies on the two-dimensional nature of graphene allowing a total exposure of all its atoms to the adsorbing gas molecules, thus providing the greatest sensor area per unit volume. Indeed several technological limits weigh on the synthesis and manipulation of the material for the device fabrication. Herein a simple approach to fabricate conductometric sensors based on chemically exfoliated natural graphite is presented. The devices were tested upon sub-ppm concentrations of NO 2 in environmental conditions and show the ability to detect this toxic gas down to few ppb at room temperature

    Fate of soil organic carbon and polycyclic aromatic hydrocarbons in a vineyard soil treated with biochar

    The effect of biochar addition on the levels of black carbon (BC) and polcyclic aromatic hydrocarbons (PAHs) in a vineyard soil in central Italy was investigated within a two year period. Hydropyrolysis (HyPy) was used to determine the contents of BC (BCHyPy) in the amended and control soils while the hydrocarbon composition of the semi-labile (non-BCHyPy) fraction released by HyPy was determined by gas chromatography-mass spectrometry, together with the solvent-extractable PAHs. The concentrations of these three polycyclic aromatic carbon reservoirs, changed and impacted differently on the soil organic carbon over the period of the trial. The addition of biochar (33 ton dry biochar ha-1) gave rise to a sharp increase in soil organic carbon which could be accounted for by an increase of BCHyPy. Over time, the concentration of BCHyPy decreased significantly from 36 to 23 mg g-1, and as a carbon percentage from 79% to 61%. No clear time trends were observed for the non-BCHyPy PAHs varying from 39 to 34 µg g-1 in treated soils, not significantly different from control soils. However, the concentrations of extractable PAHs increased markedly in the amended soils, and decreased with time from 153 to 78 ng g-1 remaining always higher than those in untreated soil. The extent of the BCHyPy loss was more compatible with physical rather than chemical processes

    Performance Characteristics of Nano-Modified Asphalt Mixtures

    The growing need for high quality paving materials has aroused an increasing interest in innovative reinforcing agents, such as those characterized by nanometric dimensions. The experimental study presented in this paper focused on the use in asphalt mixtures of nanoclays and multiwall carbon nanotubes as bitumen modifiers. The performance characteristics of asphalt mixtures containing these nano-sized additives and those of a reference neat mixture were compared in a wide array of temperature and loading conditions. The testing program included the assessment of linear viscoelastic characteristics, anti-rutting potential and crack propagation resistance, by means of stiffness modulus, flow number and semi-circular bending tests, respectively.
