Visceral Adipose Tissue Inflammatory Factors (TNF-Alpha, SOCS3) in Gestational Diabetes (GDM): Epigenetics as a Clue in GDM Pathophysiology
Gestational diabetes (GDM) is among the most challenging diseases in westernized countries, affecting mother and child, immediately and in later life. Obesity is a major risk factor for GDM. However, the impact visceral obesity and related epigenetics play for GDM etiopathogenesis have hardly been considered so far. Our recent findings within the prospective 'EaCH' cohort study of women with GDM or normal glucose tolerance (NGT), showed the role, critical factors of insulin resistance (i.e., adiponectin, insulin receptor) may have for GDM pathophysiology with epigenetically modified expression in subcutaneous (SAT) and visceral (VAT) adipose tissues. Here we investigated the expression and promoter methylation of key inflammatory candidates, tumor necrosis factor-alpha (TNF-α) and suppressor of cytokine signaling 3 (SOCS3) in maternal adipose tissues collected during caesarian section (GDM, n = 19; NGT, n = 22). The mRNA expression of TNF-α and SOCS3 was significantly increased in VAT, but not in SAT, of GDM patients vs. NGT, accompanied by specific alterations of respective promoter methylation patterns. In conclusion, we propose a critical role of VAT and visceral obesity for the pathogenesis of GDM, with epigenetic alterations of the expression of inflammatory factors as a potential factor
DNA Extraction from Dry Museum Beetles without Conferring External Morphological Damage
BACKGROUND: A large number of dry-preserved insect specimens exist in collections around the world that might be useful for genetic analyses. However, until now, the recovery of nucleic acids from such specimens has involved at least the partial destruction of the specimen. This is clearly undesirable when dealing with rare species or otherwise important specimens, such as type specimens. METHODOLOGY: We describe a method for the extraction of PCR-amplifiable mitochondrial and nuclear DNA from dry insects without causing external morphological damage. Using PCR to amplify ≈220 bp of the mitochondrial gene cytochrome c oxidase I, and 250–345 bp fragments of the multi-copy, nuclear 28s ribosomal DNA gene, we demonstrate the efficacy of this method on beetles collected up to 50 years ago. CONCLUSIONS: This method offers a means of obtaining useful genetic information from rare insects without conferring external morphological damage

Unterhaltungsprogrammen wächst auch das Interesse der deutschen Werbeindustrie an einer Nutzung dieses Instruments zur werblichen Kommunikation. Bis dato mangelt es jedoch an wissenschaftlichen Untersuchungen, welche die in der Marketingpraxis bestehenden Unsicherheiten hinsichtlich eines effektiven Einsatzes
von Product Placements beseitigen. In der Forschungsliteratur existiert bislang noch
keine umfassende wissenschaftliche Betrachtung potentieller Determinanten und Konsequenzen der Werbewirkung von Product Placements. Das Erkenntnisziel dieser Studie liegt darin, einen wesentlichen Beitrag zur Schließung dieser
Forschungslücke beizusteuern.
Zunächst erfolgt eine ausführliche Darstellung des Forschungsstands zur Werbeeffektivität von Product Placements. Auf dieser Basis werden die
Determinanten der Einstellungswirkung zur platzierten Marke und der damit verbundenen Kaufabsicht identifiziert. Die erarbeiteten Wirkungszusammenhänge werden in ein komplexes Hypothesensystem integriert und anhand der Analyse von empirisch erhobenen Daten (n= 309) unter Verwendung eines linearen
Strukturgleichungsmodells überprüft.
Die Ergebnisse der Untersuchung belegen, dass die Einstellung zur platzierten Marke und die daraus resultierende Kaufabsicht in hohem Maße durch die Gestaltung der Produktplatzierung beeinflusst werden. Darunter erweisen sich
mehrere vom Marketing-Management direkt beeinflussbare Gestaltungsdeterminanten
der Werbeeffektivität als signifikant. Die Werbewirkung ist umso effektiver, je höher die wahrgenommene Kongruenz zwischen Marke und Film sowie zwischen Marke und Hauptfigur ausfällt. Weiterhin übt die wahrgenommene Attraktivität der Hauptfigur einen positiven Effekt auf die Einstellung zur platzierten Marke aus. Schließlich kann auch ein positiver Einfluss der durch den Film induzierten Stimmung auf die Werbeeffektivität von Product Placements

Astrophysics datamining in the classroom: Exploring real data with new software tools and robotic telescopes
Within the efforts to bring frontline interactive astrophysics and astronomy
to the classroom, the Hands on Universe (HOU) developed a set of exercises and
platform using real data obtained by some of the most advanced ground and space
observatories. The backbone of this endeavour is a new free software Web tool -
Such a Lovely Software for Astronomy based on Image J (Salsa J). It is
student-friendly and developed specifically for the HOU project and targets
middle and high schools. It allows students to display, analyze, and explore
professionally obtained astronomical images, while learning concepts on
gravitational dynamics, kinematics, nuclear fusion, electromagnetism. The
continuous evolving set of exercises and tutorials is being completed with real
(professionally obtained) data to download and detailed tutorials. The
flexibility of the Salsa J platform tool enables students and teachers to
extend the exercises with their own observations. The software developed for
the HOU program has been designed to be a multi-platform, multi-lingual
experience for image manipulation and analysis in the classroom. Its design
enables easy implementation of new facilities (extensions and plugins), minimal
in-situ maintenance and flexibility for exercise plugin. Here, we describe some
of the most advanced exercises about astrophysics in the classroom, addressing
particular examples on gravitational dynamics, concepts currently introduced in

Maternal but Not Paternal High-Fat Diet (HFD) Exposure at Conception Predisposes for 'Diabesity' in Offspring Generations
While environmental epigenetics mainly focuses on xenobiotic endocrine disruptors, dietary composition might be one of the most important environmental exposures for epigenetic modifications, perhaps even for offspring generations. We performed a large-scale rat study on key phenotypic consequences from parental (F0) high-caloric, high-fat diet (HFD) food intake, precisely and specifically at mating/conception, focusing on 'diabesity' risk in first- (F1) and second- (F2) generation offspring of both sexes. F0 rats (maternal or paternal, respectively) received HFD overfeeding, starting six weeks prior to mating with normally fed control rats. The maternal side F1 offspring of both sexes developed a 'diabesity' predisposition throughout life (obesity, hyperleptinemia, hyperglycemia, insulin resistance), while no respective alterations occurred in the paternal side F1 offspring, neither in males nor in females. Mating the maternal side F1 females with control males under standard feeding conditions led, again, to a 'diabesity' predisposition in the F2 generation, which, however, was less pronounced than in the F1 generation. Our observations speak in favor of the critical impact of maternal but not paternal metabolism around the time frame of reproduction for offspring metabolic health over generations. Such fundamental phenotypic observations should be carefully considered in front of detailed molecular epigenetic approaches on eventual mechanisms
Visceral adipose tissue alteration of PI3KR1 expression is associated with gestational diabetes but not promoter DNA methylation
Obesity and diabetes are at an epidemic rate, as well as growing incidences of gestational diabetes mellitus (GDM) which causes pregnancy risks, and harm in both maternal and child health. It remains unclear which molecular mechanisms are driving the functional differences between visceral and subcutaneous fat and how these types directly affect an individual's health outcome. Paired abdominal subcutaneous and omental visceral adipose tissue were collected from women with GDM (n = 20) and with normal glucose tolerance (NGT, n = 22) during planned caesarian section. Both groups had similar maternal age (average 32.5 years) and BMI at delivery (average 33.3 kg/m2). Adipose tissue mRNA expression analyses of insulin signalling genes: PI3KCA, PI3KR1, IRS1 and IRS2 showed significantly decreased PI3KR1 expression (-23%) in visceral fat in GDM with no association to promoter DNA methylation. Reduced visceral fat PI3KR1 expression appears to be a pathogenic factor in GDM but not through altered promoter methylation
Scrapie prevalence in sheep of susceptible genotype is declining in a population subject to breeding for resistance
<p>Abstract</p> <p>Background</p> <p>Susceptibility of sheep to scrapie infection is known to be modulated by the PrP genotype of the animal. In the Netherlands an ambitious scrapie control programme was started in 1998, based on genetic selection of animals for breeding. From 2002 onwards EU regulations required intensive active scrapie surveillance as well as certain control measures in affected flocks.</p> <p>Here we analyze the data on genotype frequencies and scrapie prevalence in the Dutch sheep population obtained from both surveillance and affected flocks, to identify temporal trends. We also estimate the genotype-specific relative risks to become a detected scrapie case.</p> <p>Results</p> <p>We find that the breeding programme has produced a steady increase in the level of genetic scrapie resistance in the Dutch sheep population. We also find that a significant decline in the prevalence of scrapie in tested animals has occurred a number of years after the start of the breeding programme. Most importantly, the estimated scrapie prevalence level per head of susceptible genotype is also declining significantly, indicating that selective breeding causes a population effect.</p> <p>Conclusions</p> <p>The Dutch scrapie control programme has produced a steady rise in genetic resistance levels in recent years. A recent decline in the scrapie prevalence per tested sheep of susceptible prion protein genotype indicates that selective breeding causes the desired population effect.</p
Formation and quantification of protein complexes between peroxisomal alcohol oxidase and GroEL
We have studied the use of yeast peroxisomal alcohol oxidase (AO) as a model protein for in vitro binding by GroEL. Dilution of denatured AO in neutral buffer leads to aggregation of the protein, which is prevented by the addition of GroEL. Formation of complexes between GroEL and denatured AO was demonstrated by a gel-shift assay using non-denaturing polyacrylamide gel electrophoresis, and quantified by laser-densitometry of the gels. In the presence of MgAMP-PNP or MgADP the affinity of GroEL for AO was enhanced. Under these conditions up to 70% of the purified GroEL formed a complex with this protein. Release was stimulated at room temperature by MgATP, and was further enhanced by addition of GroES.
Eradication of scrapie with selective breeding: are we nearly there?
<p>Abstract</p> <p>Background</p> <p>Following EU decision 2003/100/EC Member States have recently implemented sheep breeding programmes to reduce the prevalence of sheep with TSE susceptible prion genotypes. The present paper investigates the progress of the breeding programme in the Netherlands. The PrP genotype frequencies were monitored through time using two sets of random samples: one set covers the years 2005 to 2008 and is taken from national surveillance programme; the other is taken from 168 random sheep farms in 2007. The data reveal that although the level of compliance to the breeding programme has been high, the frequency of susceptible genotypes varies substantially between farms. The 168 sheep farms are a subset of 689 farms participating in a postal survey inquiring about management and breeding strategies. This survey aimed to identify how much these strategies varied between farms, in order to inform assessment of the expected future progress towards eradication of classical scrapie.</p> <p>Results</p> <p>On the one hand, we found that compliance to the national breeding program has been high, and the frequency of resistant genotypes is expected to increase further in the next few years. On the other hand, we observed a large variation in prevalence of the scrapie resistant PrP genotype ARR between farms, implicating a large variation of genetic resistance between farms. Substantial between-flock differences in management and breeding strategies were found in the postal survey, suggesting considerable variation in risk of scrapie transmission between farms.</p> <p>Conclusions</p> <p>Our results show that although there has been a good progress in the breeding for scrapie resistance and the average farm-level scrapie susceptibility in the Netherlands has been significantly reduced, still a considerable proportion of farms contain high frequencies of susceptible genotypes in their sheep population. Since 2007 the breeding for genetic resistance is voluntarily again, and participation to selective breeding can decrease as a result of this. This, together with the patterns of direct and indirect contact between sheep farms, might present a challenge of the aim of scrapie eradication. Communication to sheep owners of the effect of the breeding programme thus far, and of the prospects for classical scrapie eradication in The Netherlands might be essential for obtaining useful levels of participation to the voluntary continuation of the breeding programme.</p
