    Valuable Business Knowledge Asset Discovery by Processing Unstructured Data.

    Modern organizations are challenged to enact a digital transformation and improve their competitiveness while contributing to the ninth Sustainable Development Goal (SGD), “Build resilient infrastructure, promote sustainable industrialization and foster innovation”. The discovery of hidden process data’s knowledge assets may help to digitalize processes. Working on a valuable knowledge asset discovery process, we found a major challenge in that organizational data and knowledge are likely to be unstructured and undigitized, constraining the power of today’s process mining methodologies (PMM). Whereas it has been proved in digitally mature companies, the scope of PMM becomes wider with the complement proposed in this paper, embracing organizations in the process of improving their digital maturity based on available data. We propose the C4PM method, which integrates agile principles, systems thinking and natural language processing techniques to analyze the behavioral patterns of organizational semi-structured or unstructured data from a holistic perspective to discover valuable hidden information and uncover the related knowledge assets aligned with the organization strategic or business goals. Those assets are the key to pointing out potential processes susceptible to be handled using PMM, empowering a sustainable organizational digital transformation.     Evaluación del crecimiento radial de Lactarius volemus en medio semisólido

    Objetivo. Evaluar el crecimiento radial de Lactarius volemus en cinco medios de cultivo semisólidos in vitro. Materiales y métodos. Cuerpos fructíferos de L. volemus provenientes de la Sierra Norte del estado de Oaxaca, México, se cultivaron en laboratorio en medios Agar Papa Dextrosa, Agar Czapek-Dox, Agar Extracto de Malta, Agar Papa Sacarosa y Agar Dextrosa Saboraud; mediante dos técnicas de sembrado. Se evaluaron las características morfológicas de colonias obtenidas de distintas muestras del cuerpo fructífero, así como el crecimiento radial de cada una. Resultados. El crecimiento colonial evaluado permitió seleccionar un medio que reúne las condiciones óptimas para el cultivo de Lactarius volemus in vitro. No todas las muestras utilizadas desarrollan un crecimiento abundante: la muestra proveniente del látex presenta un crecimiento escaso. Conclusiones. Con la evaluación del crecimiento radial de Lactarius volemus se obtiene una referencia directa del ciclo de crecimiento de esta especie; es posible identificar las fases exponencial y estacionaria pero las condiciones del medio no permiten evaluar la fase de muerte debido a la deshidratación y reducción del agar

    Insights of Active Extension Within a Collisional Orogen From GNSS (Central Betic Cordillera, S Spain)

    The coexistence of shortening and extensional tectonic regimes is a common feature in orogenic belts. The westernmost end of the Western Mediterranean is an area undergoing shortening related to the 5 mm/yr NNW‒SSE convergence of the Nubia and Eurasia Plates. In this region, the Central Betic Cordillera shows a regional ENE‒WSW extension. Here, we present GNSS-derived geodetic data along a 170 km-long transect orthogonal to the main active normal faults of the Central Betic Cordillera. Our data indicate that the total extension rate along the Central Betic Cordillera is 2.0 ± 0.3 mm/yr. Extension is accommodated in the eastern (0.8 ± 0.3 mm/yr in the Guadix-Baza Basin) and western (1.3 ± 0.3 mm/yr in the Granada Basin) parts of the Central Betic Cordillera, while no extension is recorded in the central part of the study area. Moreover, our data permit us to quantify, for the first time, short-term fault slip rates of the Granada Fault System, which is one of the main seismogenic sources of the Iberian Peninsula. We deduce a fault slip rate of ∼1.3 ± 0.3 mm/yr for the whole Granada Basin, with 0.9 ± 0.3 mm/yr being accommodated in the Granada Fault System and 0.4 ± 0.3 mm/yr being accommodated in the southwestern sector of the Granada Basin, where no active faults have been previously described at the surface. The heterogeneous extension in the Central Betic Cordillera could be accommodated by shallow high-angle normal faults that merge with a detachment at depth.     Crustal velocity field in Baza and Galera faults: A new estimation from GPS position time series in 2009 - 2018 time span

    The Baza and Galera faults are two active geologic structures located in the central area of the Betic Cordillera (Southern Spain). The goal of our research is to constrain the activity of this faults from high quality GPS measurements to obtain precise deformation rates. In 2008 a GPS survey – mode network was installed to monitor this area. In previous works, we presented a velocity field based on the analysis of some GPS campaigns. Here we show the new results computed from nine GPS campaigns in the timespan 2009-2018. The measurements were done in September 2009, 2010, 2011, 2012, 2013, 2014, 2015, 2017 and 2018. The data process and analysis were performed in Precise Point Positioning by using GIPSYX 1.6 software. GIPSY is a GNSS-inferred positioning software developed by the Jet Propulsion Laboratory. Then, the new estimation of the crustal velocity field is computed from the IGb14 time series by SARI software. The model applied to the original time series, using weighted least squares, consists of an intercept, a site rate and an offset to account for an antenna change. The error term is composed of white noise and temporally correlated random error. The colored noise is described by a random-walk process. We have assumed a typical magnitude for this process of 1.0 mm/√yr.     Impact of FLT3–ITD Mutation Status and Its Ratio in a Cohort of 2901 Patients Undergoing Upfront Intensive Chemotherapy: A PETHEMA Registry Study

    FLT3–ITD results in a poor prognosis in terms of overall survival (OS) and relapse-free survival (RFS) in acute myeloid leukemia (AML). However, the prognostic usefulness of the allelic ratio (AR) to select post-remission therapy remains controversial. Our study focuses on the prognostic impact of FLT3–ITD and its ratio in a series of 2901 adult patients treated intensively in the pre-FLT3 inhibitor era and reported in the PETHEMA registry. A total of 579 of these patients (20%) harbored FLT3–ITD mutations. In multivariate analyses, patients with an FLT3–ITD allele ratio (AR) of >0.5 showed a lower complete remission (CR rate) and OS (HR 1.47, p = 0.009), while AR > 0.8 was associated with poorer RFS (HR 2.1; p 0.5). Using the maximally selected log-rank statistics, we established an optimal cutoff of FLT3–ITD AR of 0.44 for OS, and 0.8 for RFS. We analyzed the OS and RFS according to FLT3–ITD status in all patients, and we found that the group of FLT3–ITD-positive patients with AR 0.44, allo-HSCT was superior to auto-HSCT in terms of OS and RFS.     NGS-Based Molecular Karyotyping of Multiple Myeloma: Results from the GEM12 Clinical Trial

    Simple Summary Multiple Myeloma (MM) is considered an incurable chronic disease, which prognosis depends on the presence of different genomic alterations. To accomplish a complete molecular diagnosis in a single essay, we have designed and validated a capture-based NGS approach to reliably identify pathogenic mutations (SNVs and indels), genomic alterations (CNVs and chromosomic translocations), and IGH rearrangements. We have observed a good correlation of the results obtained using our capture panel with data obtained by both FISH and WES techniques. In this study, the molecular classification performed using our approach was significantly associated with the stratification and outcome of MM patients. Additionally, this panel has been proven to detect specific IGH rearrangements that could be used as biomarkers in patient follow-ups through minimal residual disease (MRD) assays. In conclusion, we think that MM patients could benefit from the use of this capture-based NGS approach with a more accurate, single-essay molecular diagnosis. Next-generation sequencing (NGS) has greatly improved our ability to detect the genomic aberrations occurring in multiple myeloma (MM); however, its transfer to routine clinical labs and its validation in clinical trials remains to be established. We designed a capture-based NGS targeted panel to identify, in a single assay, known genetic alterations for the prognostic stratification of MM. The NGS panel was designed for the simultaneous study of single nucleotide and copy number variations, insertions and deletions, chromosomal translocations and V(D)J rearrangements. The panel was validated using a cohort of 149 MM patients enrolled in the GEM2012MENOS65 clinical trial. The results showed great global accuracy, with positive and negative predictive values close to 90% when compared with available data from fluorescence in situ hybridization and whole-exome sequencing. While the treatments used in the clinical trial showed high efficacy, patients defined as high-risk by the panel had shorter progression-free survival (p = 0.0015). As expected, the mutational status of TP53 was significant in predicting patient outcomes (p = 0.021). The NGS panel also efficiently detected clonal IGH rearrangements in 81% of patients. In conclusion, molecular karyotyping using a targeted NGS panel can identify relevant prognostic chromosomal abnormalities and translocations for the clinical management of MM patients
