Recent government sponsored research has demonstrated that there is a gap between the theory and practice of market intelligence gathering within the Australian micro, small and medium businesses (SMEs). Typically, there is a significant amount of information in literature about 'what needs to be done', however, there is little insight in terms of how market intelligence gathering should occur. This paper provides a novel insight and a comparison between the theory and practices of market intelligence gathering of micro-business and SMEs in Australia and demonstrates an anomoly in so far as typically the literature does not match what actually occurs in practice. A model for market intelligence gathering for micro-businesses and SMEs is also discussed

Differential hydrophobicity drives self-assembly in Huntington's disease
Identifying the driving forces and the mechanism of association of
huntingtin-exon1, a close marker for the progress of Huntington's disease, is
an important prerequisite towards finding potential drug targets, and
ultimately a cure. We introduce here a modelling framework based on a key
analogy of the physico-chemical properties of the exon1 fragment to block
copolymers. We use a systematic mesoscale methodology, based on Dissipative
Particle Dynamics, which is capable of overcoming kinetic barriers, thus
capturing the dynamics of significantly larger systems over longer times than
considered before. Our results reveal that the relative hydrophobicity of the
poly-glutamine block as compared to the rest of the (proline-based) exon1
fragment, ignored to date, constitutes a major factor in the initiation of the
self-assembly process. We find that the assembly is governed by both the
concentration of exon1 and the length of the poly-glutamine stretch, with a low
length threshold for association even at the lowest volume fractions we
considered. Moreover, this self-association occurs irrespective of whether the
glutamine stretch is in random coil or hairpin configuration, leading to
spherical or cylindrical assemblies, respectively. We discuss the implications
of these results for reinterpretation of existing research within this context,
including that the routes towards aggregation of exon1 may be distinct to those
of the widely studied homopolymeric poly-glutamine peptides
Novel splice variants associated with one of the zebrafish dnmt3 genes
BACKGROUND: DNA methylation and the methyltransferases are known to be important in vertebrate development and this may be particularly true for the Dnmt3 family of enzymes because they are thought to be the de novo methyltransferases. Mammals have three Dnmt3 genes; Dnmt3a, Dnmt3b, and Dnmt3L, two of which encode active enzymes and one of which produces an inactive but necessary cofactor. However, due to multiple promoter use and alternative splicing there are actually a number of dnmt3 isoforms present. Six different dnmt3 genes have recently been identified in zebrafish. RESULTS: We have examined two of the dnmt3 genes in zebrafish that are located in close proximity in the same linkage group and we find that the two genes are more similar to each other than they are to the other zebrafish dnmt3 genes. We have found evidence for the existence of several different splice variants and alternative splice sites associated with one of the two genes and have examined the relative expression of these genes/variants in a number of zebrafish developmental stages and tissues. CONCLUSION: The similarity of the dnmt3-1 and dnmt3-2 genes suggests that they arose due to a relatively recent gene duplication event. The presence of alternative splice and start sites, reminiscent of what is seen with the human DNMT3s, demonstrates strong parallels between the control/function of these genes across vertebrate species. The dynamic expression levels of these genes/variants suggest that they may well play a role in early development and this is particularly true for dnmt3-2-1 and dnmt3-1. dnmt3-2-1 is the predominantly expressed form prior to zygotic gene activation whereas dnmt3-1 predominates post zygotic gene activation suggesting a distinct developmental role for each
Dialog act guided contextual adapter for personalized speech recognition
Personalization in multi-turn dialogs has been a long standing challenge for
end-to-end automatic speech recognition (E2E ASR) models. Recent work on
contextual adapters has tackled rare word recognition using user catalogs. This
adaptation, however, does not incorporate an important cue, the dialog act,
which is available in a multi-turn dialog scenario. In this work, we propose a
dialog act guided contextual adapter network. Specifically, it leverages dialog
acts to select the most relevant user catalogs and creates queries based on
both -- the audio as well as the semantic relationship between the carrier
phrase and user catalogs to better guide the contextual biasing. On industrial
voice assistant datasets, our model outperforms both the baselines - dialog act
encoder-only model, and the contextual adaptation, leading to the most
improvement over the no-context model: 58% average relative word error rate
reduction (WERR) in the multi-turn dialog scenario, in comparison to the
prior-art contextual adapter, which has achieved 39% WERR over the no-context

Dual-Attention Neural Transducers for Efficient Wake Word Spotting in Speech Recognition
We present dual-attention neural biasing, an architecture designed to boost
Wake Words (WW) recognition and improve inference time latency on speech
recognition tasks. This architecture enables a dynamic switch for its runtime
compute paths by exploiting WW spotting to select which branch of its attention
networks to execute for an input audio frame. With this approach, we
effectively improve WW spotting accuracy while saving runtime compute cost as
defined by floating point operations (FLOPs). Using an in-house de-identified
dataset, we demonstrate that the proposed dual-attention network can reduce the
compute cost by for WW audio frames, with only increase in the
number of parameters. This architecture improves WW F1 score by relative
and improves generic rare word error rate by relative compared to the

Use or Abuse? A Qualitative Study of Emergency Physicians' Views on Use of Observation Stays at Three Hospitals in the United States and England.
STUDY OBJECTIVE: Accumulating evidence has shown increasing use of observation stays for patients presenting to emergency departments and requiring diagnostic evaluation or time-limited treatment plans, but critics suggest that this expansion arises from hospitals' concerns to maximize revenue and shifts costs to patients. Perspectives of physicians making decisions to admit, observe, or discharge have been absent from the debate. We examine the views of emergency physicians in the United States and England on observation stays, and what influences their decisions to use observation services. METHODS: We undertook in-depth, qualitative interviews with a purposive sample of physicians in 3 hospitals across the 2 countries and analyzed these using an approach based on the constant-comparison method. Limitations include the number of sites, whose characteristics are not generalizable to all institutions, and the reliance on self-reported interview accounts. RESULTS: Physicians used observation status for the specific presentations for which it is well evidenced but acknowledged administrative and financial considerations in their decisionmaking. They also highlighted an important role for observation not described in the literature: as a "safe space," relatively immune from the administrative gaze, where diagnostic uncertainties, sociomedical problems, and medicolegal challenges could be contained. CONCLUSION: Observation status increases the options available to admitting physicians in a way that they valued for its potential benefits to patient safety and quality of care, but some of these have been neglected in the literature to date. Reform to observation status should address these important but previously unacknowledged functions
Suppression of Epithelial to Mesenchymal Transitioning (EMT) Enhances Ex Vivo Reprogramming of Human Exocrine Pancreatic Tissue towards Functional Insulin Producing β-Like Cells
Because of the lack of tissue available for islet transplantation, new sources of β-cells have been sought for the treatment of type 1 diabetes. The aim of this study was to determine whether the human exocrine-enriched fraction from the islet isolation procedure could be reprogrammed to provide additional islet tissue for transplantation. The exocrine-enriched cells rapidly dedifferentiated in culture and grew as a mesenchymal monolayer. Genetic lineage tracing confirmed that these mesenchymal cells arose, in part, through a process of epithelial-to-mesenchymal transitioning (EMT). A protocol was developed whereby transduction of these mesenchymal cells with adenoviruses containing Pdx1, Ngn3, MafA, and Pax4 generated a population of cells that were enriched in glucagon-secreting α-like cells. Transdifferentiation or reprogramming toward insulin-secreting β-cells was enhanced, however, when using unpassaged cells in combination with inhibition of EMT by inclusion of Rho-associated kinase (ROCK) and transforming growth factor-β1 inhibitors. Resultant cells were able to secrete insulin in response to glucose and on transplantation were able to normalize blood glucose levels in streptozotocin diabetic NOD/SCID mice. In conclusion, reprogramming of human exocrine-enriched tissue can be best achieved using fresh material under conditions whereby EMT is inhibited, rather than allowing the culture to expand as a mesenchymal monolayer
A Phase 1 Randomized, Double Blind, Placebo Controlled Rectal Safety and Acceptability Study of Tenofovir 1% Gel (MTN-007)
Objective: Rectal microbicides are needed to reduce the risk of HIV acquisition associated with unprotected receptive anal intercourse. The MTN-007 study was designed to assess the safety (general and mucosal), adherence, and acceptability of a new reduced glycerin formulation of tenofovir 1% gel. Methods: Participants were randomized 1:1:1:1 to receive the reduced glycerin formulation of tenofovir 1% gel, a hydroxyethyl cellulose placebo gel, a 2% nonoxynol-9 gel, or no treatment. Each gel was administered as a single dose followed by 7 daily doses. Mucosal safety evaluation included histology, fecal calprotectin, epithelial sloughing, cytokine expression (mRNA and protein), microarrays, flow cytometry of mucosal T cell phenotype, and rectal microflora. Acceptability and adherence were determined by computer-administered questionnaires and interactive telephone response, respectively. Results: Sixty-five participants (45 men and 20 women) were recruited into the study. There were no significant differences between the numbers of ≥ Grade 2 adverse events across the arms of the study. Likelihood of future product use (acceptability) was 87% (reduced glycerin formulation of tenofovir 1% gel), 93% (hydroxyethyl cellulose placebo gel), and 63% (nonoxynol-9 gel). Fecal calprotectin, rectal microflora, and epithelial sloughing did not differ by treatment arms during the study. Suggestive evidence of differences was seen in histology, mucosal gene expression, protein expression, and T cell phenotype. These changes were mostly confined to comparisons between the nonoxynol-9 gel and other study arms. Conclusions: The reduced glycerin formulation of tenofovir 1% gel was safe and well tolerated rectally and should be advanced to Phase 2 development.
