    Mixed Data and Classification of Transit Stops

    An analysis of the characteristics and behavior of individual bus stops can reveal clusters of similar stops, which can be of use in making routing and scheduling decisions, as well as determining what facilities to provide at each stop. This paper provides an exploratory analysis, including several possible clustering results, of a dataset provided by the Regional Transit Service of Rochester, NY. The dataset describes ridership on public buses, recording the time, location, and number of entering and exiting passengers each time a bus stops. A description of the overall behavior of bus ridership is followed by a stop-level analysis. We compare multiple measures of stop similarity, based on location, route information, and ridership volume over time

    Minute-of-Arc Resolution Gamma ray Imaging Experiment—MARGIE

    MARGIE (Minute-of-Arc Resolution Gamma-ray Imaging Experiment) is a large area(∼104 cm2), wide field-of-view (∼1 sr), hard X-ray/gamma-ray (∼20–600 keV) coded-mask imaging telescope capable of performing a sensitive survey of both steady and transient cosmic sources. MARGIE has been selected for a NASA mission-concept study for an Ultra Long Duration (100 day) Balloon flight. We describe our program to develop the instrument based on new detector technology of either cadmium zinc telluride (CZT) semiconductors or pixellated cesium iodide (CsI) scintillators viewed by fast-timing bi-directional charge-coupled devices (CCDs). The primary scientific objectives are to image faint Gamma-Ray Bursts (GRBs) in near-real-time at the low intensity (high-redshift) end of the logN-logS distribution, thereby extending the sensitivity of present observations, and to perform a wide field survey of the Galactic plane

    MARGIE: A gamma-ray burst ultra-long duration balloon mission

    We are designing MARGIE as a 100 day ULDB mission to: a) detect and localize gamma-ray bursts; and b) survey the hard X-ray sky. MARGIE will consist of one small field-of-view (FOV) and four large FOV coded mask modules mounted on a balloon gondola. The burst position will be calculated onboard and disseminated in near-real time, while information about every count will be telemetered to the ground for further analysis. In a 100-day mission we will localize ∼40 bursts with peak photon fluxes from 0.14 to ∼5 ph cm−2 s−1 using 1 s integrations; the typical localization resolution will be better than ∼2 arcminutes

    COPII-dependent export of cystic fibrosis transmembrane conductance regulator from the ER uses a di-acidic exit code

    Cystic fibrosis (CF) is a childhood hereditary disease in which the most common mutant form of the CF transmembrane conductance regulator (CFTR) ΔF508 fails to exit the endoplasmic reticulum (ER). Export of wild-type CFTR from the ER requires the coat complex II (COPII) machinery, as it is sensitive to Sar1 mutants that disrupt normal coat assembly and disassembly. In contrast, COPII is not used to deliver CFTR to ER-associated degradation. We find that exit of wild-type CFTR from the ER is blocked by mutation of a consensus di-acidic ER exit motif present in the first nucleotide binding domain. Mutation of the code disrupts interaction with the COPII coat selection complex Sec23/Sec24. We propose that the di-acidic exit code plays a key role in linking CFTR to the COPII coat machinery and is the primary defect responsible for CF in ΔF508-expressing patients

    Quantifying the potential for consumer-oriented policy to reduce European and foreign carbon emissions

    The EU Carbon-CAP project assembled a comprehensive portfolio of consumer initiatives in order to assess the potential total impact of consumer options on national carbon footprints. Existing evaluations of behavioural change have focused primarily on direct energy reductions, typically in households and buildings. However, changes in consumer demand have deeper impacts via their upstream supply chains. The consumer behaviour options considered in the portfolio focus on green household initiatives. Combining existing micro-level studies with a multiregional input-output economic model, we estimated the potential efficacy and uptake of each behaviour across the European Union (EU). The results suggest that adopting these consumer options could reduce carbon footprints by approximately 25%. While 75% of this is delivered as reductions in emissions within Europe, one-quarter of the effect is delivered as a reduced imported carbon footprint, due to changes in the composition of imports

    "An Impediment to Living Life": Why and How Should We Measure Stiffness in Polymyalgia Rheumatica?

    Objectives: To explore patients’ concepts of stiffness in polymyalgia rheumatica (PMR), and how they think stiffness should be measured. Methods: Eight focus groups were held at three centres involving 50 patients with current/previous PMR. Each group had at least one facilitator and one rapporteur making field notes. An interview schedule was used to stimulate discussion. Interviews were recorded, transcribed and analysed using an inductive thematic approach. Results: Major themes identified were: symptoms: pain, stiffness and fatigue; functional impact; impact on daily schedule; and approaches to measurement. The common subtheme for the experience of stiffness was “difficulty in moving”, and usually considered as distinct from the experience of pain, albeit with a variable overlap. Some participants felt stiffness was the “overwhelming” symptom, in that it prevented them carrying out “fundamental activities” and “generally living life”. Diurnal variation in stiffness was generally described in relation to the daily schedule but was not the same as stiffness severity. Some participants suggested measuring stiffness using a numeric rating scale or a Likert scale, while others felt that it was more relevant and straightforward to measure difficulty in performing everyday activities rather than about stiffness itself. Conclusions: A conceptual model of stiffness in PMR is presented where stiffness is an important part of the patient experience and impacts on their ability to live their lives. Stiffness is closely related to function and often regarded as interchangeable with pain. From the patients’ perspective, visual analogue scales measuring pain and stiffness were not the most useful method for reporting stiffness; participants preferred numerical rating scales, or assessments of function to reflect how stiffness impacts on their daily lives. Assessing function may be a pragmatic solution to difficulties in quantifying stiffness
