    Beliefs and attitudes of profissionals about marital violence: studies with health profissionals, policemen and teachers

    Este artigo apresenta um conjunto de investigações cujo objectivo foi caracterizar as crenças dos profissionais mais directamente implicados na resposta e prevenção da violência conjugal face a este fenómeno. Para tal, um instrumento de avaliação das crenças sobre a violência conjugal foi administrado a um conjunto de 226 profissionais de saúde, 85 agentes de segurança e 280 professores. Os resultados evidenciam uma tendência para a discordância moderada, nas três amostras estudadas, relativamente aos mitos e crenças legitimadoras do abuso conjugal. Subsistem, contudo, alguns mitos relacionados com a raridade do problema, a sua atribuição a causas externas e a protecção da privacidade familiar. Os sujeitos mais velhos e de sexo masculino exibem, tendencialmente, uma maior legitimação da violência.This article presents a set of three research projects that aimed to characterize the beliefs about marital violence of the professionals most directly implied in the response and prevention of this problem. A research questionnaire that evaluates beliefs about marital violence was administered to a sample of 226 health professionals, 85 law enforcement agents and 280 teachers. On the global results show a tendency to moderate disagreement with beliefs that legitimize this form of abuse. Some myths, however, persist in these samples, namely those related to the privacy of the problem, its rarity and attribution to external causes.     Novel insights into enzymes inhibitory responses and metabolomic profile of supercritical fluid extract from chestnut shells upon intestinal permeability

    The health benefits of chestnut (Castanea sativa) shells (CSs) have been ascribed to phytochemicals, mainly phenolic compounds. Nevertheless, an exhaustive assessment of their intestinal absorption is vital considering a possible nutraceutical application. This study evaluated the bioactivity of CSs extract prepared by Supercritical Fluid Extraction and untargeted metabolomic profile upon in-vitro intestinal permeation across a Caco-2/HT29- MTX co-culture model. The results demonstrated the neuroprotective, hypoglycemic, and hypolipidemic properties of CSs extract by inhibition of acetylcholinesterase, α-amylase, and lipase activities. The untargeted metabolic profiling by LC-ESI-LTQ-Orbitrap-MS unveiled almost 60 % of lipids and 30 % of phenolic compounds, with 29 metabolic pathways indicated by enrichment analysis. Among phenolics, mostly phenolic acids, flavonoids, and coumarins permeated the intestinal barrier with most metabolites arising from phase I reactions (reduction, hydrolysis, and hydrogenation) and a minor fraction from phase II reactions (methylation). The permeation rates enhanced in the following order: ellagic acid <o-coumaric acid <p-coumaric acid <ferulaldehyde ≤hydroxyferulic acid ≤dihydroferulic acid <ferulic acid <trans-caffeic acid <trans-cinnamic acid <dihydrocaffeic acid, with better outcomes for 1000 µg/mL of extract concentration and after 4 h of permeation. Taken together, these findings sustained a considerable in-vitro intestinal absorption of phenolic compounds from CSs extract, enabling them to reach target sites and exert their biological effects. 

    Detection and quantification of EGFR T790M mutation in liquid biopsies by droplet digital PCR

In patients with advanced non-small cell lung cancer (NSCLC), droplet digital PCR (ddPCR) is increasingly used to genotype the epidermal growth factor receptor (EGFR) gene in circulating cell-free DNA (cfDNA). However, the sensitivity of this method is still under debate. The aim of this study was to implement and assess the performance of a ddPCR assay for detecting the EGFR T790M mutation in liquid biopsies. Methods: A ddPCR assay was optimized to detect the EGFR T790M mutation in plasma samples from 77 patients with NSCLC in progression. Results: Our ddPCR assay enabled the detection and quantification of the EGFR T790M mutation at cfDNA allele frequency as low as 0.5%. The mutation was detected in 40 plasma samples, corresponding to a positivity rate of 52%. The number of mutant molecules per mL of plasma ranged from 1 to 6,000. A re-biopsy was analyzed for 12 patients that had a negative plasma test and the mutation was detected in 2 cases. A second liquid biopsy was performed for 6 patients and the mutation was detected in 3 cases. Conclusions: This study highlights the value of ddPCR to detect and quantify the EGFR T790M mutation in liquid biopsies in a real-world clinical setting.     MAMMALS IN PORTUGAL : A data set of terrestrial, volant, and marine mammal occurrences in P ortugal

    Mammals are threatened worldwide, with 26% of all species being includedin the IUCN threatened categories. This overall pattern is primarily associatedwith habitat loss or degradation, and human persecution for terrestrial mam-mals, and pollution, open net fishing, climate change, and prey depletion formarine mammals. Mammals play a key role in maintaining ecosystems func-tionality and resilience, and therefore information on their distribution is cru-cial to delineate and support conservation actions. MAMMALS INPORTUGAL is a publicly available data set compiling unpublishedgeoreferenced occurrence records of 92 terrestrial, volant, and marine mam-mals in mainland Portugal and archipelagos of the Azores and Madeira thatincludes 105,026 data entries between 1873 and 2021 (72% of the data occur-ring in 2000 and 2021). The methods used to collect the data were: live obser-vations/captures (43%), sign surveys (35%), camera trapping (16%),bioacoustics surveys (4%) and radiotracking, and inquiries that represent lessthan 1% of the records. The data set includes 13 types of records: (1) burrowsjsoil moundsjtunnel, (2) capture, (3) colony, (4) dead animaljhairjskullsjjaws, (5) genetic confirmation, (6) inquiries, (7) observation of live animal (8),observation in shelters, (9) photo trappingjvideo, (10) predators dietjpelletsjpine cones/nuts, (11) scatjtrackjditch, (12) telemetry and (13) vocalizationjecholocation. The spatial uncertainty of most records ranges between 0 and100 m (76%). The data setincludes records of species classified by the IUCN as threatened(e.g.,Oryctolagus cuniculus[n=12,159],Monachus monachus[n=1,512],andLynx pardinus[n=197]). We believe that this data set may stimulate thepublication of other European countries data sets that would certainly contrib-ute to ecology and conservation-related research, and therefore assisting onthe development of more accurate and tailored conservation managementstrategies for each species.     Prospective individual patient data meta-analysis of two randomized trials on convalescent plasma for COVID-19 outpatients

    Data on convalescent plasma (CP) treatment in COVID-19 outpatients are scarce. We aimed to assess whether CP administered during the first week of symptoms reduced the disease progression or risk of hospitalization of outpatients. Two multicenter, double-blind randomized trials (NCT04621123, NCT04589949) were merged with data pooling starting when = 50 years and symptomatic for <= 7days were included. The intervention consisted of 200-300mL of CP with a predefined minimum level of antibodies. Primary endpoints were a 5-point disease severity scale and a composite of hospitalization or death by 28 days. Amongst the 797 patients included, 390 received CP and 392 placebo; they had a median age of 58 years, 1 comorbidity, 5 days symptoms and 93% had negative IgG antibody-test. Seventy-four patients were hospitalized, 6 required mechanical ventilation and 3 died. The odds ratio (OR) of CP for improved disease severity scale was 0.936 (credible interval (CI) 0.667-1.311); OR for hospitalization or death was 0.919 (CI 0.592-1.416). CP effect on hospital admission or death was largest in patients with <= 5 days of symptoms (OR 0.658, 95%CI 0.394-1.085). CP did not decrease the time to full symptom resolution

    O ensino do vocabulário e compreensão na leitura: contributo para abordagens didácticas numa perspectiva de actualização recíproca

    Com o título atribuído a este trabalho, pretende-se realçar a relação intrínseca existente entre o vocabulário e a compreensão na leitura, a mais valia que cada um deles constitui no processo de desenvolvimento do outro, a função fulcral que ambos desempenham no crescimento “inteiro” do aluno. Assim, este trabalho tem como finalidade poder contribuir para a construção de conhecimento didáctico sobre o ensino orientado para a aquisição/desenvolvimento do vocabulário, partindo do que se faz e de novas propostas resultantes de trabalhos teóricos. Poderá, assim, constituir-se como um elemento de uma linha diacrónica sobre o ensino do vocabulário e servirá para motivar o aparecimento de propostas de abordagem inovadoras. Nessa perspectiva, este projecto, tem como objectivos apresentar as concepções de professores de português, do 7º ano de escolaridade, aqui e agora, face ao ensino do vocabulário, determinar que estratégias e materiais propõem os manuais escolares que utilizam, definir quais as linhas orientadoras do programa em que se sustentam. Da maior ou menor proximidade de um casamento ou de um divórcio desta diversidade, surgirá uma perspectiva da situação na realidade nacional. Por se tratar de um estudo que interpreta e dá lugar às interpretações dos sujeitos investigados, que apresenta uma realidade, circunscrita no espaço e no tempo, sob múltiplas perspectivas, a metodologia de investigação adoptada foi o estudo etnográfico. Será, também, da maior ou menor proximidade com as perspectivas teóricas, inicialmente apresentadas, que, em conclusão, se propõem princípios didácticos que possam contribuir para a renovação das estratégias e materiais a utilizar na abordagem do ensino do vocabulário.With the title given to this work we intend to enhance the intrinsic relationship existing between vocabulary and reading comprehension, the increase in value each of them represents in the development process of the other, the fulcral role they play in the full growing up of the student. So, this work aims at being able to contribute to the construction of a didactic knowledge on a teaching process whose objective is the acquisition/development of the vocabulary, starting up from what we usually do and from new proposals, resulting from theorical works. This way, it may become an element of a diachronic line about the teaching of vocabulary and it will be useful to motivate the appearance of proposals with some innovating approaches. Following that perspective, this project has its aim to introduce the conceptions of some teachers of Portuguese, in charge of the 7th Form, in what concerns the teaching of the vocabulary, to determinate what strategies and materials are proposed by the course books they use and to define the guiding lines of the program which supports their work. From the closer or farther distance this diversity has to a wedding or to a divorce, a perspective of the situation in its national reality will arise. Because we are dealing with a study which makes an interpretation and gives origin to the interpretation of the investigated subjects, a study which introduces a reality circumscribed by space and time, under various perspectives, the adopted methodology for this investigation was the ethnographic study. It will also depend on the greater or smaller proximity to the theoretical perspectives introduced at the beginning of this work that, as a conclusion, we are going to propose some didactic principles which may contribute to the renewal of the strategies and materials to be used when approaching the teaching of vocabulary.Mestrado em Ensino de Língua
