Prospecção in silico de enzimas do complexo ligninocelulolĂtico em bacillus thuringiensis
A busca por fontes de energia alternativa avançam a medida que a disponibilidade de recursos petrĂłleo dependentes diminuem. As biomassas sĂŁo matrizes orgĂąnicas capazes de serem convertidas em energia. BactĂ©rias do gĂȘnero Bacillus spp. sĂŁo produtoras de enzimas do complexo ligninocelulolĂtico e apresentam grande potencial de uso na produção de biocombustĂveis. Algumas espĂ©cies do gĂȘnero ainda carecem de maiores investigaçÔes na busca destas enzimas, como Ă© o caso do Bacillus thuringiensis. Portanto, o objetivo do presente trabalho Ă© identificar e descrever a presença de enzimas do complexo ligninocelulolĂtico em B. thuringiensis. Os proteomas das bactĂ©rias utilizadas no estudo foram coletados no banco de dados NCBI e os dados foram prĂ© tratados utilizando linguagem de programação Python. Um script em VBA foi escrito para semi automatizar a procura das enzimas desejadas nos proteomas das bactĂ©rias via interface grĂĄfica Excel. Por fim, foi utilizado o programa Clustal Omega para construção de ĂĄrvore filogenĂ©tica das espĂ©cies coletadas. Foram encontradas 4 diferentes enzimas no proteoma de B. Thuringiensis: 6phospho ÎČ glucosidase, É glucosidase, É amilase e laccase. Todas estas com potencial de degradação de biomassa, principalmente amido e lignina. Ao analisar outras espĂ©cies do gĂȘnero, foi indentificado um maior nĂșmero e diversidade de enzimas do complexo ligninocelulolĂtico principalmente em B. amyloliquefaciens, B. licheniformis, B. velezensis e B. subtilis. Conclui se que apesar de B. thuringiensis apresentar um potencial na degradação de biomassa, outras espĂ©cies do gĂȘnero podem ser mais eficientes em aplicaçÔes reais. Objetivo: Investigar o processo de ensino e aprendizagem de slackline no contexto escolar de uma instituição pĂșblica. MĂ©todo: Participaram da pesquisa 33 discentes do Curso TĂ©cnico em Automação Industrial. Como procedimento de produção de dados, utilizamos o diĂĄrio de bordo e entrevistas acerca da questĂŁo pesquisada. Os resultados apontam a possibilidade do uso do slackline ampliando as possibilidades de participação dos discentes em aula, bem como a relação do esporte com as questĂ”es socioambientais. A tematização da aula via Educação Ambiental representa um caminho para (re)significar as aulas de Educação FĂsica no Ensino MĂ©dio
Impacts of Bar-driven Shear and Shocks on Star Formation
Bars drive gas inflow. As the gas flows inward, shocks and shear occur along the bar dust lanes. Such shocks and shear can affect the star formation (SF) and change the gas properties. For four barred galaxies, we present Hα velocity gradient maps that highlight bar-driven shocks and shear using data from the PHANGS-MUSE and PHANGS-ALMA surveys, which allow us to study bar kinematics in unprecedented detail. Velocity gradients are enhanced along the bar dust lanes, where shocks and shear are shown to occur in numerical simulations. Velocity gradient maps also efficiently pick up H ii regions that are expanding or moving relative to the surroundings. We put pseudo-slits on the regions where velocity gradients are enhanced and find that Hα and CO velocities jump up to âŒ170 km sâ1, even after removing the effects of circular motions due to the galaxy rotation. Enhanced velocity gradients either coincide with the peak of CO intensity along the bar dust lanes or are slightly offset from CO intensity peaks, depending on the objects. Using the BaldwinâPhilipsâTerlevich BPT diagnostic, we identify the source of ionization on each spaxel and find that SF is inhibited in the high-velocity gradient regions of the bar, and the majority of those regions are classified as a low-ionization nuclear emission-line region (LINER) or composite. This implies that SF is inhibited where bar-driven shear and shocks are strong. Our results are consistent with the results from the numerical simulations that show SF is inhibited in the bar where the shear force is strong
Luminous blue variables: An imaging perspective on their binarity and near environment
Luminous blue variables (LBVs) are rare massive stars with very high
luminosity. They are characterized by strong photo-metric and spectroscopic
variability related to transient eruptions. The mechanisms at the origin of
these eruptions is not well known. In addition, their formation is still
problematic and the presence of a companion could help to explain how they
form. Aims. This article presents a study of seven LBVs (about 20% of the known
Galactic population), some Wolf-Rayet stars, and massive binaries. We probe the
environments that surround these massive stars with near-, mid-, and
far-infrared images, investigating potential nebula/shells and the companion
stars. Methods. To investigate large spatial scales, we used seeing-limited and
near diffraction-limited adaptive optics images to obtain a differential
diagnostic on the presence of circumstellar matter and to determine their
extent. From those images, we also looked for the presence of binary companions
on a wide orbit. Once a companion was detected, its gravitational binding to
the central star was tested. Tests include the chance projection probability,
the proper motion estimates with multi-epoch observations, flux ratio, and star
separations. Results. We find that two out of seven of LBVs may have a wide
orbit companion. Most of the LBVs display a large circumstellar envelope or
several shells. In particular, HD168625, known for its rings, possesses several
shells with possibly a large cold shell at the edge of which the rings are

Progressive Motor Neuron Pathology and the Role of Astrocytes in a Human Stem Cell Model of VCP-Related ALS.
Motor neurons (MNs) and astrocytes (ACs) are implicated in the pathogenesis of amyotrophic lateral sclerosis (ALS), but their interaction and the sequence of molecular events leading to MN death remain unresolved. Here, we optimized directed differentiation of induced pluripotent stem cells (iPSCs) into highly enriched (> 85%) functional populations of spinal cord MNs and ACs. We identify significantly increased cytoplasmic TDP-43 and ER stress as primary pathogenic events in patient-specific valosin-containing protein (VCP)-mutant MNs, with secondary mitochondrial dysfunction and oxidative stress. Cumulatively, these cellular stresses result in synaptic pathology and cell death in VCP-mutant MNs. We additionally identify a cell-autonomous VCP-mutant AC survival phenotype, which is not attributable to the same molecular pathology occurring in VCP-mutant MNs. Finally, through iterative co-culture experiments, we uncover non-cell-autonomous effects of VCP-mutant ACs on both control and mutant MNs. This work elucidates molecular events and cellular interplay that could guide future therapeutic strategies in ALS
Do bone mineral content and density determine fracture in children? A possible threshold for physical activity
BackgroundRelations between bone parameters, physical exertion, and childhood fractures are complex. We aimed to estimate the associations between fracture history and bone mineral content (BMC) and areal bone mineral density (aBMD) at 7 years of age, by levels of physical activity, as a proxy for trauma frequency.MethodsWe used data collected from 2,261 children of the Generation XXI birth cohort, assembled in 2005/6 in Porto, Portugal. At the age of 7 years (2012/4), fracture history, time spent per week in active play, and sports practice were reported by parents. Subtotal and lumbar spine (LS) BMC and aBMD were measured using whole-body dual-energy X-ray absorptiometry.ResultsBoys and girls in the highest categories of time spent in sports practice or active play generally had higher BMC and aBMD. Among girls, BMC and aBMD were protective of fracture only in the highest quarter of active play (>660âmin/week)-odds ratios (OR; 95% confidence interval (95% CI)) for subtotal BMC=0.27 (0.11-0.67), subtotal aBMD=0.18 (0.06-0.49), and LS aBMD=0.41 (0.22-0.75). For boys in the highest quarter of sports practice (>240âmin/week), subtotal and LS BMC were protective of fracture-OR=0.39 (0.16-0.98) and 0.51 (0.27-0.96), respectively.ConclusionIn prepubertal children, BMC and aBMD predicted fracture history only in the highest levels of physical activity.info:eu-repo/semantics/publishedVersio

The unfinished agenda of communicable diseases among children and adolescents before the COVID-19 pandemic, 1990-2019: a systematic analysis of the Global Burden of Disease Study 2019
BACKGROUND: Communicable disease control has long been a focus of global health policy. There have been substantial reductions in the burden and mortality of communicable diseases among children younger than 5 years, but we know less about this burden in older children and adolescents, and it is unclear whether current programmes and policies remain aligned with targets for intervention. This knowledge is especially important for policy and programmes in the context of the COVID-19 pandemic. We aimed to use the Global Burden of Disease (GBD) Study 2019 to systematically characterise the burden of communicable diseases across childhood and adolescence. METHODS: In this systematic analysis of the GBD study from 1990 to 2019, all communicable diseases and their manifestations as modelled within GBD 2019 were included, categorised as 16 subgroups of common diseases or presentations. Data were reported for absolute count, prevalence, and incidence across measures of cause-specific mortality (deaths and years of life lost), disability (years lived with disability [YLDs]), and disease burden (disability-adjusted life-years [DALYs]) for children and adolescents aged 0-24 years. Data were reported across the Socio-demographic Index (SDI) and across time (1990-2019), and for 204 countries and territories. For HIV, we reported the mortality-to-incidence ratio (MIR) as a measure of health system performance. FINDINGS: In 2019, there were 3·0 million deaths and 30·0 million years of healthy life lost to disability (as measured by YLDs), corresponding to 288·4 million DALYs from communicable diseases among children and adolescents globally (57·3% of total communicable disease burden across all ages). Over time, there has been a shift in communicable disease burden from young children to older children and adolescents (largely driven by the considerable reductions in children younger than 5 years and slower progress elsewhere), although children younger than 5 years still accounted for most of the communicable disease burden in 2019. Disease burden and mortality were predominantly in low-SDI settings, with high and high-middle SDI settings also having an appreciable burden of communicable disease morbidity (4·0 million YLDs in 2019 alone). Three cause groups (enteric infections, lower-respiratory-tract infections, and malaria) accounted for 59·8% of the global communicable disease burden in children and adolescents, with tuberculosis and HIV both emerging as important causes during adolescence. HIV was the only cause for which disease burden increased over time, particularly in children and adolescents older than 5 years, and especially in females. Excess MIRs for HIV were observed for males aged 15-19 years in low-SDI settings. INTERPRETATION: Our analysis supports continued policy focus on enteric infections and lower-respiratory-tract infections, with orientation to children younger than 5 years in settings of low socioeconomic development. However, efforts should also be targeted to other conditions, particularly HIV, given its increased burden in older children and adolescents. Older children and adolescents also experience a large burden of communicable disease, further highlighting the need for efforts to extend beyond the first 5 years of life. Our analysis also identified substantial morbidity caused by communicable diseases affecting child and adolescent health across the world. Differential cross section measurements for the production of a W boson in association with jets in protonâproton collisions at âs = 7 TeV
Measurements are reported of differential cross sections for the production of a W boson, which decays into a muon and a neutrino, in association with jets, as a function of several variables, including the transverse momenta (pT) and pseudorapidities of the four leading jets, the scalar sum of jet transverse momenta (HT), and the difference in azimuthal angle between the directions of each jet and the muon. The data sample of pp collisions at a centre-of-mass energy of 7 TeV was collected with the CMS detector at the LHC and corresponds to an integrated luminosity of 5.0 fb[superscript â1]. The measured cross sections are compared to predictions from Monte Carlo generators, MadGraph + pythia and sherpa, and to next-to-leading-order calculations from BlackHat + sherpa. The differential cross sections are found to be in agreement with the predictions, apart from the pT distributions of the leading jets at high pT values, the distributions of the HT at high-HT and low jet multiplicity, and the distribution of the difference in azimuthal angle between the leading jet and the muon at low values.United States. Sex difference and intra-operative tidal volume: Insights from the LAS VEGAS study
BACKGROUND: One key element of lung-protective ventilation is the use of a low tidal volume (VT). A sex difference in use of low tidal volume ventilation (LTVV) has been described in critically ill ICU patients.OBJECTIVES: The aim of this study was to determine whether a sex difference in use of LTVV also exists in operating room patients, and if present what factors drive this difference.DESIGN, PATIENTS AND SETTING: This is a posthoc analysis of LAS VEGAS, a 1-week worldwide observational study in adults requiring intra-operative ventilation during general anaesthesia for surgery in 146 hospitals in 29 countries.MAIN OUTCOME MEASURES: Women and men were compared with respect to use of LTVV, defined as VT of 8âmlâkg-1 or less predicted bodyweight (PBW). A VT was deemed 'default' if the set VT was a round number. A mediation analysis assessed which factors may explain the sex difference in use of LTVV during intra-operative ventilation.RESULTS: This analysis includes 9864 patients, of whom 5425 (55%) were women. A default VT was often set, both in women and men; mode VT was 500âml. Median [IQR] VT was higher in women than in men (8.6 [7.7 to 9.6] vs. 7.6 [6.8 to 8.4] mlâkg-1 PBW, Pâ<â0.001). Compared with men, women were twice as likely not to receive LTVV [68.8 vs. 36.0%; relative risk ratio 2.1 (95% CI 1.9 to 2.1), Pâ<â0.001]. In the mediation analysis, patients' height and actual body weight (ABW) explained 81 and 18% of the sex difference in use of LTVV, respectively; it was not explained by the use of a default VT.CONCLUSION: In this worldwide cohort of patients receiving intra-operative ventilation during general anaesthesia for surgery, women received a higher VT than men during intra-operative ventilation. The risk for a female not to receive LTVV during surgery was double that of males. Height and ABW were the two mediators of the sex difference in use of LTVV.TRIAL REGISTRATION: The study was registered at Clinicaltrials.gov, NCT01601223
