Emergent quality of service - a bacterial approach
A possible model for future network quality of service control is proposed. This is based on a community of bacterial strains, each organism handling network requests in the same way as bacteria metabolise energy sources. This model makes use of the unique methods that bacteria use to transfer and share genetic material, to create a more robust solution to the service provision problems associated with future data networks
Adaptive management of an active services network
The benefits of active services and networks cannot be realised unless the associated increase in system complexity can be efficiently managed. An adaptive management solution is required. Simulation results show that a distributed genetic algorithm, inspired by observations of bacterial communities, can offer many key management functions. Performance of PCA3 and TMPRSS2:ERG urinary biomarkers in prediction of biopsy outcome in the Canary Prostate Active Surveillance Study (PASS).
BackgroundFor men on active surveillance for prostate cancer, biomarkers may improve prediction of reclassification to higher grade or volume cancer. This study examined the association of urinary PCA3 and TMPRSS2:ERG (T2:ERG) with biopsy-based reclassification.MethodsUrine was collected at baseline, 6, 12, and 24 months in the multi-institutional Canary Prostate Active Surveillance Study (PASS), and PCA3 and T2:ERG levels were quantitated. Reclassification was an increase in Gleason score or ratio of biopsy cores with cancer to â„34%. The association of biomarker scores, adjusted for common clinical variables, with short- and long-term reclassification was evaluated. Discriminatory capacity of models with clinical variables alone or with biomarkers was assessed using receiver operating characteristic (ROC) curves and decision curve analysis (DCA).ResultsSeven hundred and eighty-two men contributed 2069 urine specimens. After adjusting for PSA, prostate size, and ratio of biopsy cores with cancer, PCA3 but not T2:ERG was associated with short-term reclassification at the first surveillance biopsy (ORâ=â1.3; 95% CI 1.0-1.7, pâ=â0.02). The addition of PCA3 to a model with clinical variables improved area under the curve from 0.743 to 0.753 and increased net benefit minimally. After adjusting for clinical variables, neither marker nor marker kinetics was associated with time to reclassification in subsequent biopsies.ConclusionsPCA3 but not T2:ERG was associated with cancer reclassification in the first surveillance biopsy but has negligible improvement over clinical variables alone in ROC or DCA analyses. Therapeutic targeting of integrin αvÎČ6 in breast cancer
BACKGROUND: Integrin ?v?6 promotes migration, invasion, and survival of cancer cells; however, the relevance and role of ?v?6 has yet to be elucidated in breast cancer.METHODS: Protein expression of integrin subunit beta6 (?6) was measured in breast cancers by immunohistochemistry (n > 2000) and ITGB6 mRNA expression measured in the Molecular Taxonomy of Breast Cancer International Consortium dataset. Overall survival was assessed using Kaplan Meier curves, and bioinformatics statistical analyses were performed (Cox proportional hazards model, Wald test, and Chi-square test of association). Using antibody (264RAD) blockade and siRNA knockdown of ?6 in breast cell lines, the role of ?v?6 in Human Epidermal Growth Factor Receptor 2 (HER2) biology (expression, proliferation, invasion, growth in vivo) was assessed by flow cytometry, MTT, Transwell invasion, proximity ligation assay, and xenografts (n ? 3), respectively. A student's t-test was used for two variables; three-plus variables used one-way analysis of variance with Bonferroni's Multiple Comparison Test. Xenograft growth was analyzed using linear mixed model analysis, followed by Wald testing and survival, analyzed using the Log-Rank test. All statistical tests were two sided.RESULTS: High expression of either the mRNA or protein for the integrin subunit ?6 was associated with very poor survival (HR = 1.60, 95% CI = 1.19 to 2.15, P = .002) and increased metastases to distant sites. Co-expression of ?6 and HER2 was associated with worse prognosis (HR = 1.97, 95% CI = 1.16 to 3.35, P = .01). Monotherapy with 264RAD or trastuzumab slowed growth of MCF-7/HER2-18 and BT-474 xenografts similarly (P < .001), but combining 264RAD with trastuzumab effectively stopped tumor growth, even in trastuzumab-resistant MCF-7/HER2-18 xenografts.CONCLUSIONS: Targeting ?v?6 with 264RAD alone or in combination with trastuzumab may provide a novel therapy for treating high-risk and trastuzumab-resistant breast cancer patients.<br/

The effect of pH on the self-assembly of a collagen derived peptide amphiphile
Transitions in nanostructure driven by pH are observed for a self-assembling peptide amphiphile (PA) with a cationic pentapeptide headgroup. At pH 3, the PA forms flat tape-like structures, while at pH 4 the PA assembles into twisted right handed structures. These twisted structures transform again to flat tape-like structures at pH 7. In complete contrast, spherical micelles are observed at pH 2. These changes in response to pH may be relevant to biological and pharmaceutical applications of this PA in skincare
Extensions of the matrix Gelfand-Dickey hierarchy from generalized Drinfeld-Sokolov reduction
The matrix version of the -KdV hierarchy has been recently
treated as the reduced system arising in a Drinfeld-Sokolov type Hamiltonian
symmetry reduction applied to a Poisson submanifold in the dual of the Lie
algebra . Here a
series of extensions of this matrix Gelfand-Dickey system is derived by means
of a generalized Drinfeld-Sokolov reduction defined for the Lie algebra
using the natural
embedding for any positive integer. The
hierarchies obtained admit a description in terms of a matrix
pseudo-differential operator comprising an -KdV type positive part and a
non-trivial negative part. This system has been investigated previously in the
case as a constrained KP system. In this paper the previous results are
considerably extended and a systematic study is presented on the basis of the
Drinfeld-Sokolov approach that has the advantage that it leads to local Poisson
brackets and makes clear the conformal (-algebra) structures related to
the KdV type hierarchies. Discrete reductions and modified versions of the
extended -KdV hierarchies are also discussed.Comment: 60 pages, plain TE
Multiwavelength Monitoring of the BL Lacertae Object PKS 2155-304 in May 1994. II. The IUE Campaign
PKS 2155-304, the brightest BL Lac object in the ultraviolet sky, was
monitored with the IUE satellite at ~1 hour time-resolution for ten nearly
uninterrupted days in May 1994. The campaign, which was coordinated with EUVE,
ROSAT, and ASCA monitoring, along with optical and radio observations from the
ground, yielded the largest set of spectra and the richest short time scale
variability information ever gathered for a blazar at UV wavelengths. The
source flared dramatically during the first day, with an increase by a factor
~2.2 in an hour and a half. In subsequent days, the flux maintained a nearly
constant level for ~5 days, then flared with ~35% amplitude for two days. The
same variability was seen in both short- and long-wavelength IUE light curves,
with zero formal lag (~<2 hr), except during the rapid initial flare, when the
variations were not resolved. Spectral index variations were small and not
clearly correlated with flux. The flux variability observed in the present
monitoring is so rapid that for the first time, based on the UV emission alone,
the traditional Delta L/Delta t limit indicating relativistic beaming is
exceeded. The most rapid variations, under the likely assumption of synchrotron
radiation, lead to a lower limit of 1 G on the magnetic field strength in the
UV emitting region. These results are compared with earlier intensive
monitoring of PKS 2155-304 with IUE in November 1991, when the UV flux
