The collection, storage and use of genomic and clinical data from patients and healthy individuals is a key component of personalised medicine enterprises such as the Precision Medicine Initiative, the Cancer Moonshot and the 100,000 Genomes Project. In order to maximise the value of this data, it is important to embed a culture within the scientific, medical and patient communities that supports the appropriate sharing of genomic and clinical information. However, this aspiration raises a number of ethical, legal and regulatory challenges that need to be addressed. The Global Alliance for Genomics and Health, a worldwide coalition of researchers, healthcare professionals, patients and industry partners, is developing innovative solutions to support the responsible and effective sharing of genomic and clinical data. Southern African summer-rainfall variability, and its teleconnections, on interannual to interdecadal timescales in CMIP5 models
23 pagesInternational audienceThis study provides the first assessment of CMIP5 model performances in simulating southern Africa (SA) rainfall variability in austral summer (Nov–Feb), and its teleconnections with large-scale climate variability at different timescales. Observed SA rainfall varies at three major timescales: interannual (2–8 years), quasi-decadal (8–13 years; QDV) and interdecadal (15–28 years; IDV). These rainfall fluctuations are, respectively, associated with El Niño Southern Oscillation (ENSO), the Interdecadal Pacific Oscillation (IPO) and the Pacific Decadal Oscillation (PDO), interacting with climate anomalies in the South Atlantic and South Indian Ocean. CMIP5 models produce their own variability, but perform better in simulating interannual rainfall variability, while QDV and IDV are largely underestimated. These limitations can be partly explained by spatial shifts in core regions of SA rainfall variability in the models. Most models reproduce the impact of La Niña on rainfall at the interannual scale in SA, in spite of limitations in the representation of ENSO. Realistic links between negative IPO are found in some models at the QDV scale, but very poor performances are found at the IDV scale. Strong limitations, i.e. loss or reversal of these teleconnections, are also noted in some simulations. Such model errors, however, do not systematically impact the skill of simulated rainfall variability. This is because biased SST variability in the South Atlantic and South Indian Oceans strongly impact model skills by modulating the impact of Pacific modes of variability. Using probabilistic multi-scale clustering, model uncertainties in SST variability are primarily driven by differences from one model to another, or comparable models (sharing similar physics), at the global scale. At the regional scale, i.e. SA rainfall variability and associated teleconnections, while differences in model physics remain a large source of uncertainty, the contribution of internal climate variability is increasing. This is particularly true at the QDV and IDV scales, where the individual simulations from the same model tend to differentiate, and the sampling error increase

Thrombospondin-1 Silencing Down-Regulates Integrin Expression Levels in Human Anaplastic Thyroid Cancer Cells with BRAFV600E: New Insights in the Host Tissue Adaptation and Homeostasis of Tumor Microenvironment
Background and Rationale: Anaplastic thyroid cancer (ATC) is characterized by pleomorphic cells, has a poor prognosis, is highly devastating disease, and is not curable. No reliable biomarkers of metastatic potential, helpful for early diagnosis of ATC and therapeutic response have been found yet. Thrombospondin-1 (TSP-1) plays a fundamental role in cancer progression by regulating cell stromal cross-talk in the tumor microenvironment. Goals: Our goal was to understand whether TSP-1 could affect protein levels of its integrin receptors (e.g., ITGα3, α6, and β1) and cell morphology in BRAFV600E-ATC cells in vitro and in vivo. Experimental Design: Anaplastic thyroid cancer-derived cell cultures and western blotting were used to assess integrin protein expression upon TSP-1 silencing. Immunohistochemistry was performed on orthotopic primary human ATC and metastatic ATC in lung tissue to compare TSP-1 and integrin protein expression levels. Results:: TSP-1 knock-down down-regulates ITGα3, α6, and β1 in BRAFV600E-human ATC cells. BRAFV600E-ATC cells with TSP-1 knock-down were rounded compared to control cells, which displayed a spread morphology. TSP-1 knock-down also reduced TSP-1, ITGα3, α6, and β1 protein expression levels in vivo in the ATC microenvironment, which is enriched in stromal and inflammatory cells. Conclusion:: TSP-1 silencing causes changes in ITG levels and ATC cell morphology. The assessment of TSP-1 and ITG levels might contribute to earlier metastatic potential of BRAFV600E-positive aggressive thyroid cancers, and allow improved patient selection for clinical trials

The role of an educational leader has changed in the last century. The principal of a school has more responsibility and accountability than ever before. Chief among them is being a role model for students and adults in the school and community, and a leader in the school\u27s progress. Always in the public eye, the principal must make choices based on good judgement and data, and must search for what is best for the stakeholders. The principal should be open to new ideas and pushing the school forward. As Wilmore said in Principal Leadership, a school that is not moving forward is standing still and stagnant, and some schools have done that for too long, which is an injustice to kids and the community (Wilmore, 2002)
