    Frequency of antimicrobial resistance among invasive and colonizing Group B Streptococcal isolates

    BACKGROUND: Group B Streptococcus (GBS) remains susceptible to penicillin, however, resistance to second-line antimicrobials, clindamycin and erythromycin, has increased since 1996. We describe the age-specific antibiotic susceptibility profile and capsular type distribution among invasive and colonizing GBS strains. METHODS: We tested 486 invasive GBS isolates from individuals of all ages collected by a Wisconsin surveillance system between 1998 and 2002 and 167 colonizing strains collected from nonpregnant college students during 2001 in Michigan. Antimicrobial susceptibility testing was performed by disk diffusion or Etest and capsular typing was performed using DNA dot blot hybridization RESULTS: 20.0% (97/486) of invasive and 40.7% (68/167) of colonizing isolates were resistant to clindamycin (P < .001) and 24.5% (119/486) of invasive and 41.9% (70/167) of colonizing isolates were resistant to erythromycin (P < .001). Similarly, 19.8% (96/486) of invasive and 38.3% (64/167) of colonizing isolates were resistant to both antimicrobial agents (P < .001). 29.4% (5/17) of invasive isolates from persons 18–29 years of age and 24.3% (17/70) of invasive isolates from persons 30–49 years of age were resistant to clindamycin. Similarly, 35.3% (6/17) of invasive isolates from persons 18–29 years of age and 31.4% (22/70) of invasive isolates from persons 30–49 years of age were resistant to erythromycin. 34.7% (26/75) of invasive isolates from persons < 1 year of age were capsular type Ia, whereas capsular type V predominated among isolates from adults. CONCLUSION: Clindamycin and erythromycin resistance rates were high among isolates colonizing nonpregnant college students and invasive GBS isolates, particularly among the colonizing isolates. Susceptibility profiles were similar by age although the proportion of clindamycin and erythromycin resistance among invasive isolates was highest among persons 18–49 years of age. Increasing antimicrobial resistance has implications for GBS disease treatment and intrapartum prophylaxis among penicillin intolerant patients

    Recruitment of latent pools of high-avidity CD8+ T cells to the antitumor immune response

    A major barrier to successful antitumor vaccination is tolerance of high-avidity T cells specific to tumor antigens. In keeping with this notion, HER-2/neu (neu)-targeted vaccines, which raise strong CD8+ T cell responses to a dominant peptide (RNEU420-429) in WT FVB/N mice and protect them from a neu-expressing tumor challenge, fail to do so in MMTV-neu (neu-N) transgenic mice. However, treatment of neu-N mice with vaccine and cyclophosphamide-containing chemotherapy resulted in tumor protection in a proportion of mice. This effect was specifically abrogated by the transfer of neu-N–derived CD4+CD25+ T cells. RNEU420-429-specific CD8+ T cells were identified only in neu-N mice given vaccine and cyclophosphamide chemotherapy which rejected tumor challenge. Tetramer-binding studies demonstrated that cyclophosphamide pretreatment allowed the activation of high-avidity RNEU420-429-specific CD8+ T cells comparable to those generated from vaccinated FVB/N mice. Cyclophosphamide seemed to inhibit regulatory T (T reg) cells by selectively depleting the cycling population of CD4+CD25+ T cells in neu-N mice. These findings demonstrate that neu-N mice possess latent pools of high-avidity neu-specific CD8+ T cells that can be recruited to produce an effective antitumor response if T reg cells are blocked or removed by using approaches such as administration of cyclophosphamide before vaccination

    The ESPOSALLES database: An ancient marriage license corpus for off-line handwriting recognition

Automatic processing of historical documents, however, has mostly been focused on single works of literature and less on social records, which tend to have a distinct layout, structure, and vocabulary. Such information is usually collected by expert demographers that devote a lot of time to manually transcribe them. This paper presents a new database, compiled from a marriage license books collection, to support research in automatic handwriting recognition for historical documents containing social records. Marriage license books are documents that were used for centuries by ecclesiastical institutions to register marriage licenses. Books from this collection are handwritten and span nearly half a millennium until the beginning of the 20th century. In addition, a study is presented about the capability of state-of-the-art handwritten text recognition systems, when applied to the presented database. Baseline results are reported for reference in future studies. © 2012 Elsevier Ltd.     Characterization and exploitation of community structure in cover song networks

    The use of community detection algorithms is explored within the framework of cover song identification, i.e. the automatic detection of different audio renditions of the same underlying musical piece. Until now, this task has been posed as a typical query-by-example task, where one submits a query song and the system retrieves a list of possible matches ranked by their similarity to the query. In this work, we propose a new approach which uses song communities to provide more relevant answers to a given query. Starting from the output of a state-of-the-art system, songs are embedded in a complex weighted network whose links represent similarity (related musical content). Communities inside the network are then recognized as groups of covers and this information is used to enhance the results of the system. In particular, we show that this approach increases both the coherence and the accuracy of the system. Furthermore, we provide insight into the internal organization of individual cover song communities, showing that there is a tendency for the original song to be central within the community. We postulate that the methods and results presented here could be relevant to other query-by-example tasks

    Young Adult Counselors with Diabetes at Diabetes Camps: The Effect of Being a Peer Mentor on Counselors’ Health Behavior

    Adolescents and young adults (AYA) with type I diabetes (T1D) often struggle with illness management. Although diabetes camps have been shown to improve blood sugar control among campers, the effect of the camp experience on counselors’ diabetes self-management has never been studied. In addition to the camp environment, it was hypothesized that peer factors among counselors, such as diabetes role modeling, would positively influence diabetes self-care behaviors and that counselors would be able to select diabetes role models based on these appropriate self-management behaviors. Counselors with T1D working at 2 summer camps were recruited to participate. Participants completed questionnaires including the Diabetes Behavior Rate Scale (DBRS) and a peer-assessment form in which participants were asked to nominate friends and diabetes role models, and to assign a role model score for each counselor. Hemoglobin A1C (HbA1c), a measure of blood sugar control, was obtained pre- and post-camp. Thirty-three participants completed the study. The average HbA1c score decreased 0.4% (p < .01) over the 6 to 10 weeks of camp indicating improved metabolic control at camp. The number of nominations for diabetes role model was associated with diabetes self-care (r = 0.351, p = 0.027). Diabetes role model scores were not associated with diabetes self-care (r = 0.272, p = 0.074). There was no correlation between HbA1c and diabetes self-care. The findings suggest that the camp counselor role is a potential target for intervention to promote positive outcomes for AYA with T1D. Lessons learned from how AYA with diabetes support one another at camp can be applied to community-based interventions for youth with T1D or other chronic illnesses

    Elastic properties and secondary structure formation of single-stranded DNA at monovalent and divalent salt conditions

    Single-stranded DNA (ssDNA) plays a major role in several biological processes. It is therefore of fundamental interest to understand how the elastic response and the formation of secondary structures are modulated by the interplay between base pairing and electrostatic interactions. Here we measure force-extension curves (FECs) of ssDNA molecules in optical tweezers set up over two orders of magnitude of monovalent and divalent salt conditions, and obtain its elastic parameters by fitting the FECs to semiflexible models of polymers. For both monovalent and divalent salts, we find that the electrostatic contribution to the persistence length is proportional to the Debye screening length, varying as the inverse of the square root of cation concentration. The intrinsic persistence length is equal to 0.7 nm for both types of salts, and the effectivity of divalent cations in screening electrostatic interactions appears to be 100-fold as compared with monovalent salt, in line with what has been recently reported for single-stranded RNA. Finally, we propose an analysis of the FECs using a model that accounts for the effective thickness of the filament at low salt condition and a simple phenomenological description that quantifies the formation of non-specific secondary structure at low forces
