Response to comment on "Human-specific gain of function in a developmental enhancer"
Duret and Galtier argue that human-specific sequence divergence and gain of function in the HACNS1 enhancer result from deleterious biased gene conversion (BGC) with no contribution from positive selection. We reinforce our previous conclusion by analyzing hypothesized BGC
events genomewide and assessing the effect of recombination rates on human-accelerated conserved noncoding sequence ascertainment. We also provide evidence that AT â GC substitution bias can coexist with positive selection
MAES Service Case: Wetland ecosystem condition mapping (v.1.0)
SWOS Technical publicationThe MAES working group is preparing the workshop on âecosystem condition mappingâ to streamline the efforts done so far with regards to the mapping and assessment of the condition of Europeâs ecosystems. The MAES WG has requested directly to SWOS partners a specific document to support the mapping and assessing wetland ecosystem condition for this workshop. This document shall highlight the different elements to take into account for the mapping and assessment of wetland ecosystems with the aim of supporting Member States and the European Commission in their efforts to better describe the situation of wetland ecosystems in Europe.
This document represents the major output of the MAES Service case that shall show how SWOS outputs are useful to support the MAES WG with regards to wetland ecosystem mapping and assessment
The Status of Coastal Benthic Ecosystems in the Mediterranean Sea: Evidence From Ecological Indicators
The Mediterranean Sea is subject to multiple human pressures increasingly threatening its unique biodiversity. Spatially explicit information on the ecological status of marine ecosystems is therefore key to an effective maritime spatial planning and management, and to help the achievement of environmental targets. Here, we summarized scientific data on the ecological status of a selection of marine ecosystems based on a set of ecological indicators in more than 700 sites of the Mediterranean Sea. For Posidonia oceanica seagrass beds, rocky intertidal fringe, and coastal soft bottoms, more than 70% of investigated sites exhibited good to high ecological conditions. In contrast, about two-thirds of sites for subtidal rocky reefs were classified to be in moderate to bad conditions, stressing the need for prioritizing conservation initiatives on these productive and diverse environments. Very little quantitative information was available for the southern Mediterranean Sea, thus monitoring programs and assessments in this area are essential for a representative assessment of the health of marine coastal ecosystems in the whole basin. This overview represents a first step to implement a baseline that, through georeferenced data on ecological status, could help identifying information gaps, directing future research priorities, and supporting improvements to spatial models of expected cumulative impacts on marine ecosystems
Attribute and technology value mapping for conceptual product design phase

The new test-bed has the ability to define a technology value map to assess multiple technical options as a function of its attributes, whose precise values can be determined at a given cost. A mathematical model that incorporates a highly adaptable, data-driven and multi-attribute value approach to product specification and conceptual design is developed, novel to the concept design process. Enhanced antitumoral activity of TLR7 agonists via activation of human endogenous retroviruses by HDAC inhibitors
In this work, we are reporting that âShock and Killâ, a therapeutic approach designed to eliminate latent HIV from cell reservoirs, is extrapolatable to cancer therapy. This is based on the observation that malignant cells express a spectrum of human endogenous retroviral elements (HERVs) which can be transcriptionally boosted by HDAC inhibitors. The endoretroviral gene HERV-V2 codes for an envelope protein, which resembles syncytins. It is significantly overexpressed upon exposure to HDAC inhibitors and can be effectively targeted by simultaneous application of TLR7/8 agonists, triggering intrinsic apoptosis. We demonstrated that this synergistic cytotoxic effect was accompanied by the functional disruption of the TLR7/8-NFÎșB, Akt/PKB, and Ras-MEK-ERK signalling pathways. CRISPR/Cas9 ablation of TLR7 and HERV-V1/V2 curtailed apoptosis significantly, proving the pivotal role of these elements in driving cell death. The effectiveness of this new approach was confirmed in ovarian tumour xenograft studies, revealing a promising avenue for future cancer therapies
High resolution AMI Large Array imaging of spinning dust sources: spatially correlated 8 micron emission and evidence of a stellar wind in L675
We present 25 arcsecond resolution radio images of five Lynds Dark Nebulae
(L675, L944, L1103, L1111 & L1246) at 16 GHz made with the Arcminute
Microkelvin Imager (AMI) Large Array. These objects were previously observed
with the AMI Small Array to have an excess of emission at microwave frequencies
relative to lower frequency radio data. In L675 we find a flat spectrum compact
radio counterpart to the 850 micron emission seen with SCUBA and suggest that
it is cm-wave emission from a previously unknown deeply embedded young
protostar. In the case of L1246 the cm-wave emission is spatially correlated
with 8 micron emission seen with Spitzer. Since the MIR emission is present
only in Spitzer band 4 we suggest that it arises from a population of PAH
molecules, which also give rise to the cm-wave emission through spinning dust

Evidence that links loss of cyclooxygenase-2 with increased asymmetric dimethylarginine : novel explanation of cardiovascular side effects associated with anti-inflammatory drugs

Cyclooxygenase-2 is expressed in the renal medulla where inhibition causes fluid retention and increased blood pressure. However, the mechanisms linking cyclooxygenase-2 inhibition and cardiovascular events are unknown and no biomarkers have been identified.METHODS AND RESULTS: Transcriptome analysis of wild-type and cyclooxygenase-2(-/-) mouse tissues revealed 1 gene altered in the heart and aorta, but >1000 genes altered in the renal medulla, including those regulating the endogenous nitric oxide synthase inhibitors asymmetrical dimethylarginine (ADMA) and monomethyl-l-arginine. Cyclo-oxygenase-2(-/-) mice had increased plasma levels of ADMA and monomethyl-l-arginine and reduced endothelial nitric oxide responses. These genes and methylarginines were not similarly altered in mice lacking prostacyclin receptors. Wild-type mice or human volunteers taking cyclooxygenase-2 inhibitors also showed increased plasma ADMA. Endothelial nitric oxide is cardio-protective, reducing thrombosis and atherosclerosis. Consequently, increased ADMA is associated with cardiovascular disease. Thus, our study identifies ADMA as a biomarker and mechanistic bridge between renal cyclooxygenase-2 inhibition and systemic vascular dysfunction with nonsteroidal anti-inflammatory drug usage.CONCLUSIONS: We identify the endogenous endothelial nitric oxide synthase inhibitor ADMA as a biomarker and mechanistic bridge between renal cyclooxygenase-2 inhibition and systemic vascular dysfunction.Peer reviewedFinal Published versio
Interactions between Magnetic Nanowires and Living Cells : Uptake, Toxicity and Degradation
We report on the uptake, toxicity and degradation of magnetic nanowires by
NIH/3T3 mouse fibroblasts. Magnetic nanowires of diameters 200 nm and lengths
comprised between 1 {\mu}m and 40 {\mu}m are fabricated by controlled assembly
of iron oxide ({\gamma}-Fe2O3) nanoparticles. Using optical and electron
microscopy, we show that after 24 h incubation the wires are internalized by
the cells and located either in membrane-bound compartments or dispersed in the
cytosol. Using fluorescence microscopy, the membrane-bound compartments were
identified as late endosomal/lysosomal endosomes labeled with lysosomal
associated membrane protein (Lamp1). Toxicity assays evaluating the
mitochondrial activity, cell proliferation and production of reactive oxygen
species show that the wires do not display acute short-term (< 100 h) toxicity
towards the cells. Interestingly, the cells are able to degrade the wires and
to transform them into smaller aggregates, even in short time periods (days).
This degradation is likely to occur as a consequence of the internal structure
of the wires, which is that of a non-covalently bound aggregate. We anticipate
that this degradation should prevent long-term asbestos-like toxicity effects
related to high aspect ratio morphologies and that these wires represent a
promising class of nanomaterials for cell manipulation and microrheology.Comment: 21 pages 12 figure
