Cryo-EM structure of the nucleosome containing the ALB1 enhancer DNA sequence
Pioneer transcription factors specifically target their recognition DNA sequences within nucleosomes. FoxA is the pioneer transcription factor that binds to the ALB1 gene enhancer in liver precursor cells, and is required for liver differentiation in embryos. The ALB1 enhancer DNA sequence is reportedly incorporated into nucleosomes in cells, although the nucleosome structure containing the targeting sites for FoxA has not been clarified yet. In this study, we determined the nucleosome structure containing the ALB1 enhancer (N1) sequence, by cryogenic electron microscopy at 4.0 Å resolution. The nucleosome structure with the ALB1 enhancer DNA is not significantly different from the previously reported nucleosome structure with the Widom 601 DNA. Interestingly, in the nucleosomes, the ALB1 enhancer DNA contains local flexible regions, as compared to the Widom 601 DNA. Consistently, DNaseI treatments revealed that, in the nucleosome, the ALB1 enhancer (N1) DNA is more accessible than the Widom 601 sequence. The histones also associated less strongly with the ALB1 enhancer (N1) DNA than the Widom 601 DNA in the nucleosome. Therefore, the local histone–DNA contacts may be responsible for the enhanced DNA accessibility in the nucleosome with the ALB1 enhancer DNA

This theoretical article reviews old criticisms to and new issues of "Theory of Innate Infant Intersubjectivity (IS)" by Colwyn Trevarthen, which is recently getting other researchers\u27 attentions. Firstly, distinctions between rational, verbal, and interpretive IS in philosophy and IS to denote a communicative, sharing, and interacting IS in developmental psychology are made clear. Secondly, as old criticisms to IS, innateness of IS, objectivity of Trevarthen\u27s research methodology, underestimation of parent role in a developmental process, and developmental timing of self-other identification are listed and their theoretical limitations are discussed. Thirdly, new issues of IS, the main part of this article, which include developmental continuity/discontinuity between PIS and SIS, conceptual ambiguity between SIS and Joint-Attention, and the need to consider "We-space" and "Event and Uncertainty" to approach dynamic aspects of IS rather than static synchronized IS are reviewed with related recent studies. As a conclusion, it is proposed that establishing a methodology to approach shared manifold of IS to find dynamic aspects of IS is the key for future studies

Background : Bronchial occlusion using endobronchial Watanabe Spigot(EWS)is reported to be useful for treatment of secondary intractable pneumothorax and thoracic empyema, peripheral bronchial fistula. However, the methods of the bronchial occlusion are sometimes difficult and EWS sometimes fall off from plugged bronchus. Case : A 44 year old man presented hemosputum. He was diagnosed with Aspergilloma. We performed a resection of the right upper lobe and S6 partial resection. Air leak appeared at postoperative day 3. We performed EWS embolization with an application of push & slide method and the ropeway method, and the persistent air leak disappeared. Conclusion : Our method is useful when the bronchial occlusion is difficult
