    Growth Response of Explants of Irvingia Gabonensis (O'rorke, Baill) to In Vitro Treatment

    Growth response of explants of Irvingia gabonensis to in vitro treatment was investigated using full, half and one quarter strength mineral components based on Murashige and Skoog medium. Plant growth regulator (kinetin-Kin) with concentration levels of 0, 1, 2, 3, 4 and 5mg/l were used for shoots initiation, while axillary formed shoots were rooted in various concentration levels of 0, 1, 2, 3, 4 and 5mg/l of indole butyric acid (IBA). One quarter strength growth medium containing 3mg/l kinetin, which supported the highest growth performance was used to study the effect of explant source and number of leaf primordia on shoot initiation of the species. Shoot length of axillary bud explant significantly (P<0.05) increased in one quarter strength medium than those of half and full strength media. Axillary bud explants with 1-2 leaf primordia gave optimum response in terms of regeneration frequency and coefficient of velocity of bud burst. High regeneration frequency and coefficient of velocity of bud burst with low percentage contamination were recorded by seedling and stump sprout explants. This study suggests that axillary bud explants of Irvingia gabonensis can respond to in vitro treatment under optimum culture conditions

    Multilevel Safety Climate in The UK Rail Industry: A Cross Validation of the Zohar and Luria MSC Scale

    Despite a downward trend in injury rates in UK workplaces, accident occurrence remains an on-going issue for the rail workforce. Results from the RSSB annual survey reveal that there were 164 major injuries in 2016/17. Safety climate is defined as “shared perceptions with regard to safety policies, procedures and practices.” Many studies have examined the positive effects of safety climate on safety performances by individuals, teams, organizations. Despite widespread attempts to measure safety climate, the validity of measurement tools has not been systematically tested in the rail industry. The primary goal of our research was to validate Zohar and Luria’s (2005) Multilevel Safety Climate Scale in a sample of rail infrastructure workers (N = 528). A cross-validation strategy was adopted. Half of the data were used to conduct exploratory factor analysis (EFA), with the remaining data submitted to confirmative factor analysis (CFA). The statistical results reveal a three-factor structure with organizational safety climate (OSC), supervisor safety communication (SSC), supervisor safety monitoring (SSM). A nomological analysis showed that SSC and SSM presented distinct correlation patterns with other measures of relevance for safety, risk and health management. SSM was found more strongly related with variables such as: safety priorities; safety systems; reporting attitudes; safety compliance. On the other hand, SSC was mainly related with measures refereed to distinct forms of organizational support: supervisor support; peer support; support to change. Overall, our findings showed the validity of a multidimensional approach on the study of safety climate and safety supervision in the rail industry

    Enhancement of toxin- and virus-neutralizing capacity of single-domain antibody fragments by N-glycosylation

    Single-domain antibody fragments (VHHs) have several beneficial properties as compared to conventional antibody fragments. However, their small size complicates their toxin- and virus-neutralizing capacity. We isolated 27 VHHs binding Escherichia coli heat-labile toxin and expressed these in Saccharomyces cerevisiae. The most potent neutralizing VHH (LT109) was N-glycosylated, resulting in a large increase in molecular mass. This suggests that N-glycosylation of LT109 improves its neutralizing capacity. Indeed, deglycosylation of LT109 decreased its neutralizing capacity three- to fivefold. We also studied the effect of glycosylation of two previously isolated VHHs on their ability to neutralize foot-and-mouth disease virus. For this purpose, these VHHs that lacked potential N-glycosylation sites were genetically fused to another VHH that was known to be glycosylated. The resulting fusion proteins were also N-glycosylated. They neutralized the virus at at least fourfold-lower VHH concentrations as compared to the single, non-glycosylated VHHs and at at least 50-fold-lower VHH concentrations as compared to their deglycosylated counterparts. Thus, we have shown that N-glycosylation of VHHs contributes to toxin- and virus-neutralizing capacity

    ELECTRONIC-STRUCTURE OF AMORPHOUS SI-N COMPOUNDS

    We have measured valence-band photoemission spectra and dark conductivity of a-SiN(x):H compounds for compositions between x = 0 and x = 1.35. The photoemission spectra have been measured with Zr Mzeta and Al Kalpha radiation of 151.4 and 1486.6 eV, respectively. At hnu = 151.4 eV the spectra resemble directly the total density-of-states (DOS) of the system; the most important change with x is the shift of spectral weight from near the valence-band maximum (VBM) toward the center of the band, indicating the change from a band of Si-Si bonding states to a band of Si-N bonding states. At hnu = 1486.6 eV the spectra are dominated by the contribution of the Si-3s partial DOS; this contribution is located at the bottom of the band and shifts toward higher binding energies with increasing x. We compare our results at x = 0.36 and x = 1.35 with those of two recent calculations. Combining results of the dark-conductivity measurements and the photoemission spectra with a previous determination of the optical gaps we make a plot of the VBM, Fermi-level position, and conduction-band minimum (CBM) versus x. It is shown that the sudden opening of the gap at x approximately 1 is due mainly to the recession of the CBM.4919134461345

    Construction and Application of an Electronic Spatiotemporal Expression Profile and Gene Ontology Analysis Platform Based on the EST Database of the Silkworm, Bombyx mori

    An Expressed Sequence Tag (EST) is a short sub-sequence of a transcribed cDNA sequence. ESTs represent gene expression and give good clues for gene expression analysis. Based on EST data obtained from NCBI, an EST analysis package was developed (apEST). This tool was programmed for electronic expression, protein annotation and Gene Ontology (GO) category analysis in Bombyx mori (L.) (Lepidoptera: Bombycidae). A total of 245,761 ESTs (as of 01 July 2009) were searched and downloaded in FASTA format, from which information for tissue type, development stage, sex and strain were extracted, classified and summed by running apEST. Then, corresponding distribution profiles were formed after redundant parts had been removed. Gene expression profiles for one tissue of different developmental stages and from one development stage of the different tissues were attained. A housekeeping gene and tissue-and-stage-specific genes were selected by running apEST, contrasting with two other online analysis approaches, microarray-based gene expression profile on SilkDB (BmMDB) and EST profile on NCBI. A spatio-temporal expression profile of catalase run by apEST was then presented as a three-dimensional graph for the intuitive visualization of patterns. A total of 37 query genes confirmed from microarray data and RT—PCR experiments were selected as queries to test apEST. The results had great conformity among three approaches. Nevertheless, there were minor differences between apEST and BmMDB because of the unique items investigated. Therefore, complementary analysis was proposed. Application of apEST also led to the acquisition of corresponding protein annotations for EST datasets and eventually for their functions. The results were presented according to statistical information on protein annotation and Gene Ontology (GO) category. These all verified the reliability of apEST and the operability of this platform. The apEST can also be applied in other species by modifying some parameters and serves as a model for gene expression study for Lepidoptera

    An observational study of patient characteristics associated with the mode of admission to acute stroke services in North East, England

    Objective Effective provision of urgent stroke care relies upon admission to hospital by emergency ambulance and may involve pre-hospital redirection. The proportion and characteristics of patients who do not arrive by emergency ambulance and their impact on service efficiency is unclear. To assist in the planning of regional stroke services we examined the volume, characteristics and prognosis of patients according to the mode of presentation to local services. Study design and setting A prospective regional database of consecutive acute stroke admissions was conducted in North East, England between 01/09/10-30/09/11. Case ascertainment and transport mode were checked against hospital coding and ambulance dispatch databases. Results Twelve acute stroke units contributed data for a mean of 10.7 months. 2792/3131 (89%) patients received a diagnosis of stroke within 24 hours of admission: 2002 arrivals by emergency ambulance; 538 by private transport or non-emergency ambulance; 252 unknown mode. Emergency ambulance patients were older (76 vs 69 years), more likely to be from institutional care (10% vs 1%) and experiencing total anterior circulation symptoms (27% vs 6%). Thrombolysis treatment was commoner following emergency admission (11% vs 4%). However patients attending without emergency ambulance had lower inpatient mortality (2% vs 18%), a lower rate of institutionalisation (1% vs 6%) and less need for daily carers (7% vs 16%). 149/155 (96%) of highly dependent patients were admitted by emergency ambulance, but none received thrombolysis. Conclusion Presentations of new stroke without emergency ambulance involvement were not unusual but were associated with a better outcome due to younger age, milder neurological impairment and lower levels of pre-stroke dependency. Most patients with a high level of pre-stroke dependency arrived by emergency ambulance but did not receive thrombolysis. It is important to be aware of easily identifiable demographic groups that differ in their potential to gain from different service configurations
