    Relationship between polymorphism of growth hormone and precocity traits in Nellore heifers

    Avaliaram-se as relações entre o polimorfismo do gene do hormônio do crescimento (GH) e as características de precocidade, em novilhas da raça Nelore. Amostras de sangue periférico foram obtidas de 181 animais de três rebanhos distintos do estado da Bahia, nas quais foi realizada a extração de DNA e a amplificação por PCR, seguidas por digestão com enzima de restrição AluI. Os fragmentos resultantes da digestão enzimática foram analisados em gel de agarose 2% para determinação dos respectivos genótipos. A frequência do alelo Leu nas amostras analisadas foi estimada em 100%. Em decorrência da alta incidência de homozigose para o alelo Leu, sugere-se que o restriction fragment lenght polymorphism AluI do gene GH não possa ser considerado como marcador molecular para precocidade sexual em novilhas Nelore nesses rebanhos.The relationships between polymorphism of growth hormone gene (GH) and precocity traits in Nellore heifers were evaluated. A total of 181 animals from three different farms of Bahia state, Brazil, were blood sampled. The DNA of each animal was extracted, amplified by PCR, and digested by "AluI" restriction enzyme, and the resultant fragments were analyzed in 2% agarose gel for genotype identification. The frequency of Leu allele in the analyzed samples was estimated in 100%.     Molecular mechanisms of zinc toxicity in the potworm Enchytraeus crypticus, analysed by high-throughput gene expression profiling

    Zinc (Zn) is known to be relatively toxic to some soil-living invertebrates including the ecologically important enchytraeid worms. To reveal the molecular mechanisms of zinc toxicity we assessed the gene expression profile of Enchytraeus crypticus (Enchytraeidae), exposed to the reproduction effect concentrations EC10 and EC50, over 4 consecutive days, using a high-throughput microarray (species customized). Three main mechanisms of toxicity to Zn were observed: 1) Zn trafficking (upregulation of zinc transporters, a defence response to regulate the cellular zinc level), 2) oxidative stress (variety of defence mechanisms, triggered by Reactive Oxygen Species (ROS)), and 3) effects on the nervous system (possibly the primary lesion explaining the avoidance behaviour and also why enchytraeids are relatively susceptible to Zn). The adverse outcome at the organism level (reproduction EC50) could be predicted based on gene expression (male gonad development, oocyte maturation), with Zn at the EC50 affecting processes related to higher stress levels. The gene expression response was time-dependent and reflected the cascade of events taking place over-time. The 1 to 4 days of exposure design was a good strategy as it captured the time for sequence of events towards zinc adverse outcomes in E. crypticus

    Toxic effects multi-walled carbon nanotubes on bivalves: comparison between of functionalized and non-functionalized nanoparticles

    Despite of the large array of available carbon nanotube (CNT) configurations that allow different industrial and scientific applications of these nanoparticles, their impacts on aquatic organisms, especially on invertebrate species, are still limited. To our knowledge, no information is available on how surface chemistry alteration (functionalization) of CNTs may impact the toxicity of these NPs to bivalve species after a chronic exposure. For this reason, the impacts induced by chronic exposure (28 days) to unfunctionalized MWCNTs (Nf-MWCNTs) in comparison with functionalized MWCNTs (f-MWCNTs), were evaluated in R. philippinarum, by measuring alterations induced in clams' oxidative status, neurotoxicity and metabolic capacity. The results obtained revealed that exposure to both MWCNT materials altered energy-related responses, with higher metabolic capacity and lower glycogen, protein and lipid concentrations in clams exposed to these CNTs. Moreover, R. philippinarum exposed to Nf-MWCNTs and f-MWCNTs showed oxidative stress expressed in higher lipid peroxidation and lower ratio between reduced and oxidized glutathione, despite the activation of defense mechanisms (superoxide-dismutase, glutathione peroxidase and glutathione S-transferases) in exposed clams.     Simbióticos e monensina sódica no desempenho e na qualidade da carne de novilhas mestiças Angus confinadas

    Resumo:O objetivo deste trabalho foi avaliar o efeito da inclusão de simbióticos e de monensina sódica em dietas sobre o desempenho, as características de carcaça e a qualidade da carne de novilhas mestiças Angus em confinamento. Utilizou-se um delineamento inteiramente casualizado, com 64 animais distribuídos em quatro tratamentos: controle, dieta sem suplementação; dieta suplementada com simbióticos; dieta com monensina sódica; e dieta com simbióticos e monensina sódica. A dieta com monensina sódica e a dieta com simbióticos e monensina sódica proporcionaram maior ganho de peso médio diário e ganho de peso vivo total, além de melhorar a eficiência alimentar e biológica dos animais em comparação ao controle. A dieta de simbióticos, a de monensina e a de simbióticos com monensina sódica ocasionaram a redução da ingestão de matéria seca, elevaram o peso do filé da costela e os teores de lipídeos totais na carne. A dieta com simbióticos proporcionou a redução da força de cisalhamento da carne em relação ao controle. A inclusão de simbióticos, com ou sem monensina sódica, na dieta de novilhas mestiças Angus confinadas, beneficia o desempenho animal e algumas características físico-químicas da carne
