    Impact of endostatin gene therapy on myeloid-derived suppressor cells from a metastatic renal cell carcinoma

    Aim: To evaluate the role of endostatin (ES) gene therapy on myeloid-derived suppressor cells (MDSC) in a metastatic model of renal cell carcinoma (RCC). Materials and Methods: Balb/C mice bearing orthotopic Renca tumors were treated with NIH/3T3LendSN or, as a control, with NIH/3T3-LXSN cells. At the end of in vivo experiment, plasma and tissue lung samples were collected. Plasma ES and granulocyte colony stimulating factor (G-CSF) levels were measured by ELISA and Milliplex, respectively. Quantification of CD11b⁺Gr⁻1⁺ cells and their subsets was performed by flow cytometry. Reactive oxygen species (ROS) production was measured in CD11b⁺Gr⁻1⁺ MDSC using the DCFDA marker by flow cytometry. Results: Metastatic RCC (mRCC) induced expansions of CD11b⁺Gr⁻1⁺ MDSC and promoted accumulation of these cells and their subtypes in lymphoid organ and metastases. ES treatment promoted low G-CSF plasmatic levels which were produced by the tumor microenvironment, reflecting the reduced metastatic accumulation of CD11b⁺Gr⁻1⁺ MDSC in the lungs. However, the therapy was selective for granulocytic cells, thus reducing the production of ROS. Conclusion: These findings confirm the expansion of MDSC during metastatic progression of RCC and indicate the important role of ES in reducing MDSC and possible use of ES therapy in combined anticancer treatment

    Collagen XVIII/endostatin expression in experimental endotoxemic acute renal failure

    Acute renal failure (ARF) is a frequent complication of Gram-negative sepsis, with a high risk of mortality. Lipopolysaccharide (LPS)-induced ARF is associated with hemodynamic changes that are strongly influenced by the overproduction of nitric oxide (NO) through the cytokine-mediated up-regulation of inducible NO synthase. LPS-induced reductions in systemic vascular resistance paradoxically culminate in renal vasoconstriction. Collagen XVIII is an important component of the extracellular matrix expressed in basement membranes. Its degradation by matrix metalloproteases, cathepsins and elastases results in the formation of endostatin, claimed to have antiangiogenic activity and to be a prominent vasorelaxing agent. We evaluated the expression of endostatin/collagen XVIII in an endotoxemic ARF model. ARF was induced in C57BL/6 mice by intraperitoneal injection of LPS (10 mg/kg) followed by sacrifice 4 and 12 h later. Kidney tissue was the source of RNA and protein and the subject of histological analysis. As early as 4 h after LPS administration, blood urea, creatinine and NO levels were significantly increased compared to control. Endostatin/collagen XVIII mRNA levels were 0.71 times lower than sham-inoculated mice 4 h after LPS inoculation, returning to normal levels 12 h after LPS inoculation. Immunohistological examination revealed that acute injury caused by LPS leads to an increase of endostatin basement membrane staining in association with the decrease of CD31 endothelial basement membrane staining. These results indicate that in the early phase of endotoxemic ARF the endostatin levels were not regulated by gene expression, but by the metabolism of collagen XVIII

    Spectroscopy by frequency entangled photon pairs

    Quantum spectroscopy was performed using the frequency-entangled broadband photon pairs generated by spontaneous parametric down-conversion. An absorptive sample was placed in front of the idler photon detector, and the frequency of signal photons was resolved by a diffraction grating. The absorption spectrum of the sample was measured by counting the coincidences, and the result is in agreement with the one measured by a conventional spectrophotometer with a classical light source.Comment: 11 pages, 5 figures, to be published in Phys.     Neutrino masses from beta decays after KamLAND and WMAP (Updated including the NC enhanced SNO data)

    The first data released by the KamLAND collaboration have confirmed the strong evidence in favour of the LMA solution of the solar neutrino problem. Taking into account the ranges for the oscillation parameters allowed by the global analysis of the solar, CHOOZ and KamLAND data, we update the limits on the neutrinoless double beta decay effective neutrino mass parameter and analyze the impact of all the available data from neutrinoless double beta decay experiments on the neutrino mass bounds, in view of the latest WMAP results. For the normal neutrino mass spectrum the range (0.05-0.23) eV is obtained for the lightest neutrino mass if one takes into account the Heidelberg-Moscow evidence for neutrinoless double beta decay and the cosmological bound. It is also shown that under the same conditions the mass of the lightest neutrino may not be bounded from below if the spectrum is of the inverted type. Finnaly, we discuss how future experiments can improve the present bounds on the lightest neutrino mass set by the Troitsk, Mainz and WMAP results. In the addendum we update the allowed ranges for the effective Majorana neutrino mass parameter in view of the latest NC enhanced SNO data.Comment: Updated including the recent NC enhanced SNO data.     Measurement of the CP-Violating Asymmetry Amplitude sin2β\beta

    We present results on time-dependent CP-violating asymmetries in neutral B decays to several CP eigenstates. The measurements use a data sample of about 88 million Y(4S) --> B Bbar decays collected between 1999 and 2002 with the BABAR detector at the PEP-II asymmetric-energy B Factory at SLAC. We study events in which one neutral B meson is fully reconstructed in a final state containing a charmonium meson and the other B meson is determined to be either a B0 or B0bar from its decay products. The amplitude of the CP-violating asymmetry, which in the Standard Model is proportional to sin2beta, is derived from the decay-time distributions in such events. We measure sin2beta = 0.741 +/- 0.067 (stat) +/- 0.033 (syst) and |lambda| = 0.948 +/- 0.051 (stat) +/- 0.017 (syst). The magnitude of lambda is consistent with unity, in agreement with the Standard Model expectation of no direct CP violation in these modes

    Determination of the Form Factors for the Decay B0 --> D*-l+nu_l and of the CKM Matrix Element |Vcb|

    We present a combined measurement of the Cabibbo-Kobayashi-Maskawa matrix element Vcb|V_{cb}| and of the parameters ρ2\rho^2, R1R_1, and R2R_2, which fully characterize the form factors of the B0D+νB^0 \to D^{*-}\ell^{+}\nu_\ell decay in the framework of HQET, based on a sample of about 52,800 B0D+νB^0 \to D^{*-}\ell^{+}\nu_\ell decays recorded by the BABAR detector. The kinematical information of the fully reconstructed decay is used to extract the following values for the parameters (where the first errors are statistical and the second systematic): ρ2=1.156±0.094±0.028\rho^2 = 1.156 \pm 0.094 \pm 0.028, R1=1.329±0.131±0.044R_1 = 1.329 \pm 0.131 \pm 0.044, R2=0.859±0.077±0.022R_2 = 0.859 \pm 0.077 \pm 0.022, F(1)Vcb=(35.03±0.39±1.15)×103\mathcal{F}(1)|V_{cb}| = (35.03 \pm 0.39 \pm 1.15) \times 10^{-3}. By combining these measurements with the previous BABAR measurements of the form factors which employs a different technique on a partial sample of the data, we improve the statistical accuracy of the measurement, obtaining: ρ2=1.179±0.048±0.028,R1=1.417±0.061±0.044,R2=0.836±0.037±0.022,\rho^2 = 1.179 \pm 0.048 \pm 0.028, R_1 = 1.417 \pm 0.061 \pm 0.044, R_2 = 0.836 \pm 0.037 \pm 0.022, and F(1)Vcb=(34.68±0.32±1.15)×103. \mathcal{F}(1)|V_{cb}| = (34.68 \pm 0.32 \pm 1.15) \times 10^{-3}. Using the lattice calculations for the axial form factor F(1)\mathcal{F}(1), we extract Vcb=(37.74±0.35±1.25±1.441.23)×103|V_{cb}| =(37.74 \pm 0.35 \pm 1.25 \pm ^{1.23}_{1.44}) \times 10^{-3}, where the third error is due to the uncertainty in F(1)\mathcal{F}(1)

    A search for the decay B+K+ννˉB^+ \to K^+ \nu \bar{\nu}

    We search for the rare flavor-changing neutral-current decay B+K+ννˉB^+ \to K^+ \nu \bar{\nu} in a data sample of 82 fb1^{-1} collected with the {\sl BABAR} detector at the PEP-II B-factory. Signal events are selected by examining the properties of the system recoiling against either a reconstructed hadronic or semileptonic charged-B decay. Using these two independent samples we obtain a combined limit of B(B+K+ννˉ)<5.2×105{\mathcal B}(B^+ \to K^+ \nu \bar{\nu})<5.2 \times 10^{-5} at the 90% confidence level. In addition, by selecting for pions rather than kaons, we obtain a limit of B(B+π+ννˉ)<1.0×104{\mathcal B}(B^+ \to \pi^+ \nu \bar{\nu})<1.0 \times 10^{-4} using only the hadronic B reconstruction method.Comment: 7 pages, 8 postscript figures, submitted to Phys.     High-reflectivity broadband distributed Bragg reflector lattice matched to ZnTe

    We report on the realization of a high quality distributed Bragg reflector with both high and low refractive index layers lattice matched to ZnTe. Our structure is grown by molecular beam epitaxy and is based on binary compounds only. The high refractive index layer is made of ZnTe, while the low index material is made of a short period triple superlattice containing MgSe, MgTe, and ZnTe. The high refractive index step of Delta_n=0.5 in the structure results in a broad stopband and the reflectivity coefficient exceeding 99% for only 15 Bragg pairs.Comment: 4 pages, 3 figure

    Study of the Exclusive Initial-State Radiation Production of the DDˉD \bar D System

    A study of exclusive production of the DDˉD \bar D system through initial-state r adiation is performed in a search for charmonium states, where D=D0D=D^0 or D+D^+. The D0D^0 mesons are reconstructed in the D0Kπ+D^0 \to K^- \pi^+, D0Kπ+π0D^0 \to K^- \pi^+ \pi^0, and D0Kπ+π+πD^0 \to K^- \pi^+ \pi^+ \pi^- decay modes. The D+D^+ is reconstructed through the D+Kπ+π+D^+ \to K^- \pi^+ \pi^+ decay mode. The analysis makes use of an integrated luminosity of 288.5 fb1^{-1} collected by the BaBar experiment. The DDˉD \bar D mass spectrum shows a clear ψ(3770)\psi(3770) signal. Further structures appear in the 3.9 and 4.1 GeV/c2c^2 regions. No evidence is found for Y(4260) decays to DDˉD \bar D, implying an up per limit \frac{\BR(Y(4260)\to D \bar D)}{\BR(Y(4260)\to J/\psi \pi^+ \pi^-)} < 7.6 (95 % confidence level)

    EuFe2_2As2_2 under high pressure: an antiferromagnetic bulk superconductor

    We report the ac magnetic susceptibility χac\chi_{ac} and resistivity ρ\rho measurements of EuFe2_2As2_2 under high pressure PP. By observing nearly 100% superconducting shielding and zero resistivity at PP = 28 kbar, we establish that PP-induced superconductivity occurs at TcT_c \sim~30 K in EuFe2_2As2_2. ρ\rho shows an anomalous nearly linear temperature dependence from room temperature down to TcT_c at the same PP. χac\chi_{ac} indicates that an antiferromagnetic order of Eu2+^{2+} moments with TNT_N \sim~20 K persists in the superconducting phase.
