    HELLP babies link a novel lincRNA to the trophoblast cell cycle

    The HELLP syndrome is a pregnancy-associated disease inducing hemolysis, elevated liver enzymes, and low platelets in the mother. Although the HELLP symptoms occur in the third trimester in the mother, the origin of the disease can be found in the first trimester fetal placenta. A locus for the HELLP syndrome is present on chromosome 12q23 near PAH. Here, by multipoint nonparametric linkage, pedigree structure allele sharing, and haplotype association analysis of affected sisters and cousins, we demonstrate that the HELLP locus is in an intergenic region on 12q23.2 between PMCH and IGF1. We identified a novel long intergenic noncoding RNA (lincRNA) transcript of 205,012 bases with (peri)nuclear expression in the extravillous trophoblast using strand-specific RT-PCR complemented with RACE and FISH. siRNA-mediated knockdown followed by RNA-sequencing, revealed that the HELLP lincRNA activated a large set of genes that are involved in the cell cycle. Furthermore, blocking potential mutation sites identified in HELLP families decreased the invasion capacity of extravillous trophoblasts. This is the first large noncoding gene to be linked to a Mendelian disorder with autosomal-recessive inheritance

    Constraint methods for determining pathways and free energy of activated processes

    Activated processes from chemical reactions up to conformational transitions of large biomolecules are hampered by barriers which are overcome only by the input of some free energy of activation. Hence, the characteristic and rate-determining barrier regions are not sufficiently sampled by usual simulation techniques. Constraints on a reaction coordinate r have turned out to be a suitable means to explore difficult pathways without changing potential function, energy or temperature. For a dense sequence of values of r, the corresponding sequence of simulations provides a pathway for the process. As only one coordinate among thousands is fixed during each simulation, the pathway essentially reflects the system's internal dynamics. From mean forces the free energy profile can be calculated to obtain reaction rates and insight in the reaction mechanism. In the last decade, theoretical tools and computing capacity have been developed to a degree where simulations give impressive qualitative insight in the processes at quantitative agreement with experiments. Here, we give an introduction to reaction pathways and coordinates, and develop the theory of free energy as the potential of mean force. We clarify the connection between mean force and constraint force which is the central quantity evaluated, and discuss the mass metric tensor correction. Well-behaved coordinates without tensor correction are considered. We discuss the theoretical background and practical implementation on the example of the reaction coordinate of targeted molecular dynamics simulation. Finally, we compare applications of constraint methods and other techniques developed for the same purpose, and discuss the limits of the approach

    Onset of Superfluidity in 4He Films Adsorbed on Disordered Substrates

    We have studied 4He films adsorbed in two porous glasses, aerogel and Vycor, using high precision torsional oscillator and DC calorimetry techniques. Our investigation focused on the onset of superfluidity at low temperatures as the 4He coverage is increased. Torsional oscillator measurements of the 4He-aerogel system were used to determine the superfluid density of films with transition temperatures as low as 20 mK. Heat capacity measurements of the 4He-Vycor system probed the excitation spectrum of both non-superfluid and superfluid films for temperatures down to 10 mK. Both sets of measurements suggest that the critical coverage for the onset of superfluidity corresponds to a mobility edge in the chemical potential, so that the onset transition is the bosonic analog of a superconductor-insulator transition. The superfluid density measurements, however, are not in agreement with the scaling theory of an onset transition from a gapless, Bose glass phase to a superfluid.     Transverse spin effects in hadron-pair production from semi-inclusive deep inelastic scattering

    First measurements of azimuthal asymmetries in hadron-pair production in deep-inelastic scattering of muons on transversely polarised ^6LiD (deuteron) and NH_3 (proton) targets are presented. The data were taken in the years 2002-2004 and 2007 with the COMPASS spectrometer using a muon beam of 160 GeV/c at the CERN SPS. The asymmetries provide access to the transversity distribution functions, without involving the Collins effect as in single hadron production. The sizeable asymmetries measured on the NH_ target indicate non-vanishing u-quark transversity and two-hadron interference fragmentation functions. The small asymmetries measured on the ^6LiD target can be interpreted as indication for a cancellation of u- and d-quark transversities.Comment: 13 pages, 4 figures, updated to the published versio

    Measurement of the Collins and Sivers asymmetries on transversely polarised protons

    The Collins and Sivers asymmetries for charged hadrons produced in deeply inelastic scattering on transversely polarised protons have been extracted from the data collected in 2007 with the CERN SPS muon beam tuned at 160 GeV/c. At large values of the Bjorken x variable non-zero Collins asymmetries are observed both for positive and negative hadrons while the Sivers asymmetry for positive hadrons is slightly positive over almost all the measured x range. These results nicely support the present theoretical interpretation of these asymmetries, in terms of leading-twist quark distribution and fragmentation functions.Comment: 9 Pages, 5 figure

    A new measurement of the Collins and Sivers asymmetries on a transversely polarised deuteron target

    New high precision measurements of the Collins and Sivers asymmetries of charged hadrons produced in deep-inelastic scattering of muons on a transversely polarised 6LiD target are presented. The data were taken in 2003 and 2004 with the COMPASS spectrometer using the muon beam of the CERN SPS at 160 GeV/c. Both the Collins and Sivers asymmetries turn out to be compatible with zero, within the present statistical errors, which are more than a factor of 2 smaller than those of the published COMPASS results from the 2002 data. The final results from the 2002, 2003 and 2004 runs are compared with naive expectations and with existing model calculations.Comment: 40 pages, 28 figure

    Evidence for an Excess of Soft Photons in Hadronic Decays of Z^0

    Soft photons inside hadronic jets converted in front of the DELPHI main tracker (TPC) in events of qqbar disintegrations of the Z^0 were studied in the kinematic range 0.2 < E_gamma < 1 GeV and transverse momentum with respect to the closest jet direction p_T < 80 MeV/c. A clear excess of photons in the experimental data as compared to the Monte Carlo predictions is observed. This excess (uncorrected for the photon detection efficiency) is (1.17 +/- 0.06 +/- 0.27) x 10^{-3} gamma/jet in the specified kinematic region, while the expected level of the inner hadronic bremsstrahlung (which is not included in the Monte Carlo) is (0.340 +/- 0.001 +/- 0.038) x 10^{-3} gamma/jet. The ratio of the excess to the predicted bremsstrahlung rate is then (3.4 +/- 0.2 +/- 0.8), which is similar in strength to the anomalous soft photon signal observed in fixed target experiments with hadronic beams.Comment: 37 pages, 9 figures, Accepted by Eur.     Energy dependence of Cronin momentum in saturation model for p+Ap+A and A+AA+A collisions

    We calculate s\sqrt{s} dependence of Cronin momentum for p+Ap+A and A+AA+A collisions in saturation model. We show that this dependence is consistent with expectation from formula which was obtained using simple dimentional consideration. This can be used to test validity of saturation model (and distinguish among its variants) and measure xx dependence of saturation momentum from experimental data.Comment: LaTeX2e, 12 pages, 8 figure
