Molecular Analysis of N6-Methyladenine Patterns in \u3cem\u3eTetrahymena thermophila\u3c/em\u3e Nuclear DNA
We have cloned two DNA fragments containing 5\u27-GATC-3\u27 sites at which the adenine is methylated in the macronucleus of the ciliate Tetrahymena thermophila. Using these cloned fragments as molecular probes, we analyzed the maintenance of methylation patterns at two partially and two uniformly methylated sites. Our results suggest that a semiconservative copying model for maintenance of methylation is not sufficient to account for the methylation patterns we found during somatic growth of Tetrahymena. Although we detected hemimethylated molecules in macronuclear DNA, they were present in both replicating and nonreplicating DNA. In addition, we observed that a complex methylation pattern including partially methylated sites was maintained during vegetative growth. This required the activity of a methylase capable of recognizing and modifying sites specified by something other than hemimethylation. We suggest that a eucaryotic maintenance methylase may be capable of discriminating between potential methylation sites to ensure the inheritance of methylation patterns
Understanding the role of promoters in catalysis: operando XAFS/DRIFTS study of CeO<sub>x</sub>/Pt/Al<sub>2</sub>O<sub>3</sub> during CO oxidation
A combined operando XAFS/DRIFTS study on CeOx/Pt/Al2O3 catalysts has been performed during CO oxidation and provides insights into the changes in nanoparticle structure and adsorbed species during the reaction profile. The onset of CO2 formation is shown to be concurrent with a rapid re-oxidation of the Pt nanoparticles, evidenced by XAFS spectroscopy, and the loss of bridge bonded CO adsorbed on Pt, as shown by simultaneous DRIFTS acquisition. The continued appearance of linear bound CO on the catalyst surface is shown to remain long after catalytic light off. The interaction of Pt and CeOx is evidenced by the improved performance towards CO oxidation, compared to the non-CeOx modified Pt/Al2O3, and changes in the CO adsorption properties on Pt previously linked to Pt-CeO2 interfaces
Application of a Bayesian Method to Absorption Spectral-Line Finding in Simulated ASKAP Data
The large spectral bandwidth and wide field of view of the Australian SKA
Pathfinder radio telescope will open up a completely new parameter space for
large extragalactic HI surveys. Here we focus on identifying and parametrising
HI absorption lines which occur in the line of sight towards strong radio
continuum sources. We have developed a method for simultaneously finding and
fitting HI absorption lines in radio data by using multi-nested sampling, a
Bayesian Monte Carlo algorithm. The method is tested on a simulated ASKAP data
cube, and is shown to be reliable at detecting absorption lines in low
signal-to-noise data without the need to smooth or alter the data. Estimation
of the local Bayesian evidence statistic provides a quantitative criterion for
assigning significance to a detection and selecting between competing

The Contribution of the Negro to the Drama
The Negro has had some part in American drama practically from its inception. 1 Whatever else the Negro may contribute as a gift to this composite civilization, there is already the general recognition that his folk-music born of the pangs and sorrows of slavery, has made America and the world his eternal debtor. The same racial characteristics that are responsible for this music are destined to express themselves with similar excellences in the kindred art of drama.
The drama of Negro life is developing primarily because a native drama is in process of evolution. This, although it heralds the awakening of the dormant dramatic gifts of the Negro and has meant the phenomenal rise within a decade\u27s span of a Negro drama and a possible Negro Theatre. For pioneering genii in the development of the native American drama such as Eugene O\u27Neill, Ridgely Torrence, and Paul Green, now see and recognize the dramatically undeveloped potentialities of Negro life and folk ways as a promising province of native idioms and source material in which a developing national drama can find distinctive new themes, characteristics, and typical situations, authentic atmosphere. The growing number of successful and representative plays of this type form a valuable and significant contribution to the theatre of today, and open intriguing and fascinating possibilities for the theatre of tomorrow. 2

Rigidity analysis of HIV-1 protease
We present a rigidity analysis on a large number of X-ray crystal structures
of the enzyme HIV-1 protease using the 'pebble game' algorithm of the software
FIRST. We find that although the rigidity profile remains similar across a
comprehensive set of high resolution structures, the profile changes
significantly in the presence of an inhibitor. Our study shows that the action
of the inhibitors is to restrict the flexibility of the beta-hairpin flaps
which allow access to the active site. The results are discussed in the context

Next Generation Higgs Bosons: Theory, Constraints and Discovery Prospects at the Large Hadron Collider
Particle physics model building within the context of string theory suggests
that further copies of the Higgs boson sector may be expected. Concerns
regarding tree-level flavor changing neutral currents are easiest to allay if
little or no couplings of next generation Higgs bosons are allowed to Standard
Model fermions. We detail the resulting general Higgs potential and mass
spectroscopy in both a Standard Model extension and a supersymmetric extension.
We present the important experimental constraints from meson-meson mixing,
loop-induced decays and LEP2 direct production limits. We
investigate the energy range of valid perturbation theory of these ideas. In
the supersymmetric context we present a class of examples that marginally aids
the fine-tuning problem for parameter space where the lightest Higgs boson mass
is greater than the Standard Model limit of 114 GeV. Finally, we study collider
physics signatures generic to next generation Higgs bosons, with special
emphasis on signal events, and describe the capability of
