Semisynthetic Aurones Inhibit Tubulin Polymerization at the Colchicine-Binding Site and Repress PC-3 Tumor Xenografts in Nude Mice and Myc-Induced T-ALL in Zebrafish
Structure-activity relationships (SAR) in the aurone pharmacophore identified heterocyclic variants of the (Z)-2-benzylidene-6-hydroxybenzofuran-3(2H)-one scaffold that possessed low nanomolar in vitro potency in cell proliferation assays using various cancer cell lines, in vivo potency in prostate cancer PC-3 xenograft and zebrafish models, selectivity for the colchicine-binding site on tubulin, and absence of appreciable toxicity. Among the leading, biologically active analogs were (Z)-2-((2-((1-ethyl-5-methoxy-1H-indol-3-yl)methylene)-3-oxo-2,3-dihydrobenzofuran-6-yl)oxy)acetonitrile (5a) and (Z)-6-((2,6-dichlorobenzyl)oxy)-2-(pyridin-4-ylmethylene)benzofuran-3(2H)-one (5b) that inhibited in vitro PC-3 prostate cancer cell proliferation with IC50 values below 100 nM. A xenograft study in nude mice using 10 mg/kg of 5a had no effect on mice weight, and aurone 5a did not inhibit, as desired, the human ether-à-go-go-related (hERG) potassium channel. Cell cycle arrest data, comparisons of the inhibition of cancer cell proliferation by aurones and known antineoplastic agents, and in vitro inhibition of tubulin polymerization indicated that aurone 5a disrupted tubulin dynamics. Based on molecular docking and confirmed by liquid chromatography-electrospray ionization-tandem mass spectrometry studies, aurone 5a targets the colchicine-binding site on tubulin. In addition to solid tumors, aurones 5a and 5b strongly inhibited in vitro a panel of human leukemia cancer cell lines and the in vivo myc-induced T cell acute lymphoblastic leukemia (T-ALL) in a zebrafish model
Discovery and Development of Small-Molecule Inhibitors of Glycogen Synthase
The overaccumulation of glycogen appears as a hallmark in various glycogen storage diseases (GSDs), including Pompe, Cori, Andersen, and Lafora disease. Accumulating evidence suggests that suppression of glycogen accumulation represents a potential therapeutic approach for treating these GSDs. Using a fluorescence polarization assay designed to screen for inhibitors of the key glycogen synthetic enzyme, glycogen synthase (GS), we identified a substituted imidazole, (rac)-2-methoxy-4-(1-(2-(1-methylpyrrolidin-2-yl)ethyl)-4-phenyl-1H-imidazol-5-yl)phenol (H23), as a first-in-class inhibitor for yeast GS 2 (yGsy2p). Data from X-ray crystallography at 2.85 Å, as well as kinetic data, revealed that H23 bound within the uridine diphosphate glucose binding pocket of yGsy2p. The high conservation of residues between human and yeast GS in direct contact with H23 informed the development of around 500 H23 analogs. These analogs produced a structure–activity relationship profile that led to the identification of a substituted pyrazole, 4-(4-(4-hydroxyphenyl)-3-(trifluoromethyl)-1H-pyrazol-5-yl)pyrogallol, with a 300-fold improved potency against human GS. These substituted pyrazoles possess a promising scaffold for drug development efforts targeting GS activity in GSDs associated with excess glycogen accumulation
Rock fishladder as element of ecological corridor restoration for strongly degradeted rivers
W ostatnich dziesięcioleciach w wyniku regulacji rzek i przegradzania dolin
rzecznych nastąpiły niekorzystne zmiany w hydromorfologii koryt rzecznych
[Bartnik, 2006]. Zwiększenie retencji wodnej i wykorzystanie hydroenergetyczne
rzek, które pociąga za sobą budowę nowych obiektów hydrotechnicznych wymaga
nowego spojrzenia na ciągłość korytarza rzecznego.
Zgodnie z RDW „silnie zmieniona część wód oznacza część wód powierzchniowych,
których charakter został znacznie zmieniony na skutek fizycznego
oddziaływania człowieka […]” [Dyrektywa 2000/60/WE]. W silnie zmienionych
częściach wód należy dążyć do osiągnięcia dobrego potencjału
ekologicznego wód. Nastąpić to może poprzez budowę lub przebudowę budowli
wodnych w sposób umożliwiający odpowiednie warunki migracji i życia dla ryb
i innych organizmów wodnych.
Celem pracy jest wyznaczenie dopuszczalnych wymiarów konstrukcji
przepławki ryglowej spełniające kryteria biologiczne ryb w zlewni rzeki Skawy.
W tym celu zostały przeprowadzone pomiary laboratoryjne na fizycznym modelu
przepławki ryglowej. Na podstawie badań modelowych przepławki ryglowej określono
warunki stabilności biologicznej: przepływu, prędkości wody, napełnienia
i dyssypacji objętościowej.In the recent decades, due to improper management of river ecosystems adverse
effects occurred in riverbeds hydromorphology. Increased water retention
and use of hydroelectric rivers, which entails the construction of new hydro power
plants requires a new look at the continuity of the river corridor.
According to the RDW "heavily modified water body means a body of surface
water, the nature of which has been significantly altered as a result of physical
alterations by human [...]"[Dyrektywa 2000/60/WE]. The heavily modified
water bodies should aim to achieve good ecological potential of waters. It is to be
achieved through the construction or reconstruction of water structures to ensure
efficient migration and living conditions for fish and other aquatic organisms.
The aim of this study is to determine the construction limits of rock fishladders
that meet the biological criteria of fish in the Skawa river. Laboratory measurements
were carried out on the physical model of rock fishladder. The results of
laboratory measurement were used to calculate hydraulic condition within fishladder
to determine biological stability of fishes: discharge, velocity of water,
water depth and energy dissipation
Hydraulic conditions of alluring stream occurrence for transom ladders
Opracowane przez Komisję Europejską w 2007 roku szczegółowe rekomendacje
wprowadzają nowe uwarunkowania związane z zapewnieniem ciągłości
korytarzy rzecznych. Istniejące budowle piętrzące powinny być wyposażone
w przepławki dla zachowania warunków restytucji ryb, a nowe budowle hydrotechniczne
powinny uwzględniać takie rozwiązania techniczne, które nie powodują
zmian stanu hydromorfologicznego. Celem zachowania warunków stabilności
biologicznej dla ryb dwuśrodowiskowych wg Instytutu Rybactwa muszą być spełnione
następujące kryteria stabilności biologicznej przy projektowaniu i eksploatacji
przepławek:
– kryterium prądu wabiącego,
– kryterium prędkości granicznych,
– kryterium dopuszczalnego napełnienia w przepławce,
– kryterium dopuszczalnej wielkości strat Δh przy przepływie przez rygiel
dla przepławek ryglowych.
W artykule przedstawiono wyniki modelowania numerycznego 400-
metrowego odcinka rzeki obejmującego jaz powłokowy, małą elektrownię wodną
oraz przepławkę dla ryb typu ryglowego. Podjęte działania skupiały się na takim
ukształtowaniu przepływu w przepławce i modyfikacji konfiguracji dna na wylocie
z elektrowni, aby wytworzyć prąd wabiący. Wyniki modelowania numerycznego
zostały porównane z wartościami otrzymanymi ze wzorów empirycznych.Detailed recommendations formulated by European Commission in 2007
introduce new regulations regarding the continuity of river channels. The existing
hydraulic structures should be equipped with fish ladders in order to keep the fish
restitution conditions and the new hydro-technical structures should assure technical
solutions that do not cause any change in hydro-morphological state. According
to the Fisheries Institute the following conditions of biological stability
should be followed to keep the biological stability conditions for diadromic fishes:
– an alluring stream criterion,
– a terminal velocity criterion,
– a criterion of admissible fish ladder filling,
– a criterion of admissible loss Δh for the transom ladder discharge.
The paper presents results of numerical modeling of 400-meter sector of
a river that includes coated weir, small hydrostation and a fish transom ladder.
The action taken concentrated on such formation of the discharge in the ladder
and modification of the bed configuration in the hydro mouth that creates the alluring
stream. Numerical modeling results were compared to results of calculations
by empirical formula
Monitoring survey of nitrate content in beetroot, radish and cabbage in Poland
Background. Nitrates, compounds commonly occurring in nature, are present for example in vegetables, where they
accumulate and become their contaminants. It is estimated that approximately 70-90% of nitrates intake comes from
vegetables, which are therefore the main source of human exposure to these compounds through dietary intake. The
European Union legislation limits nitrates as contaminants to three leafy vegetables, i.e. lettuce, spinach and rucola.
The EU Member States are obliged to monitor nitrate levels in vegetables which may contain significant levels of these
compounds.
Objective. To present the results of monitoring surveys on nitrate levels in radish and beetroot as well as in cabbage
carried out in Poland between 2012 and 2019.
Material and methods. A total of 966 vegetable samples were tested. Chemical analyses were carried out in accredited
laboratories of the State Sanitary Inspection. Analyses were performed by spectrophotometric methods using nitrate
reduction on cadmium columns or by HPLC.
Results. The median nitrate content in beetroot was 1,260.0 mg NO−
3 /kg, whilst at the 95th percentile level - 3,222.2 mg
NO−
3 /kg. The levels of nitrates in beetroot preserves were lower: 1,030.3 mg NO−
3 /kg (median) and 2337,2 mg NO−
3 /kg
(95th percentile). The median content of nitrates in radish and cabbage was 1,337.0 mg NO−
3 /kg and 369,0 mg NO−
3 /kg
respectively, while at the 95th percentile the content of these compounds was found to be 3,381.5 mg NO−
3 /kg and 1545,8 mg
NO−
3 /kg, respectively.
Conclusions. The nitrate content in radish and cabbage does not pose a risk to the health of consumers, whilst the
consumption of beetroot containing significant amounts of the above mentioned compounds may result in exceeding the
ADI especially for young children
Application of 1d models (HEC-RAS, MIKE 11) to designate flood hazard areas on the Lubcza river, the Wislok basin
Do obliczenia układu zwierciadła wody dla przepływów prawdopodobnych
w przypadku wyznaczenia stref zagrożenia powodziowego stosuje się modele jednowymiarowe,
np. HEC-RAS, MIKE 11. Programy hydroinformatyczne HECRAS
i MIKE 11 służą do modelowania przepływu wody w korycie i dolinie
rzecznej wraz z obiektami inżynierskimi o złożonych konstrukcjach (mosty, przepusty,
jazy i inne). Etapami powstawania modelu są: schematyzacja sieci rzecznej,
pomiary geodezyjne, obejmujące koryto rzeczne i budowle inżynierskie, identyfikacja
współczynników szorstkości, obliczenia hydrologiczne przepływów w wybranych
przekrojach modelowanego cieku, stanowiące warunki brzegowe, obliczenia
numeryczne wraz z kalibracją i weryfikacją modelu oraz wizualizacja
wyników obliczeń.
W artykule przedstawiono wyniki modelowania odcinka rzeki Lubcza
w zlewni Wisłoka o długości ok. 8,7 km (zlewnia niekontrolowana), na którym
zlokalizowanych jest 17 mostów oraz 12 stopni i progów wodnych. Podstawą analiz
i porównania uzyskanych wyników modelowania są rzędne zwierciadła wody
dla przepływów o określonym prawdopodobieństwie przewyższenia oraz utworzony
na ich podstawie Numeryczny Model Powierzchni Wody (NMPW). Strefy
zagrożenia powodziowego powstają z przecięcia numerycznego modelu terenu
(NMT) i NMPW. Znajomość układu zwierciadła wody umożliwia określenie stref
zagrożenia powodziowego.1-dimensional models, e.g. HEC-RAS, MIKE 11 are used to calculate water
surface elevation concerning probable discharges for flood hazard areas designation.
Hydro-informatic software programs HEC-RAS and MIKE 11 are used for
modeling of water discharge in the river bed and in the river valley, with engineering
objects of complicated structure (bridges, culverts, weirs etc.). The following
stages of modeling may be distinguished: schematisation of water network, geodetic
measurements of the river bed and engineering buildings, identification of
roughness coefficients, hydrological calculations of discharges in selected profiles
of the modelled watercourse considering the boundary conditions, numeric calculations
with calibration and verification of the model as well as visualisation of the
calculations’ results.
The paper presents the results of modeling of a sector of The Lubcza River
in the Wislok basin having length equal 8,7 km (uncontrolled basin), in which
17 bridges and 12 weirs are located. The elevations of the water surface for discharges
of the defined probability of exceedances and the Digital Model of the
Water Surface (DMWS) are the basis for the analyses and comparison of the obtained
results. Flood hazard areas occur on the intersection of the Digital Terrain
Model (DTM) and DMWS. The knowledge of water surface permit the destination
of the flood hazard areas
