    lHuman cytotoxic T lymphocytes with reduced sensitivity to Fas-induced apoptosis

    Effector-memory T cells expressing Fas (Apo-1/CD95) are switched to an apoptotic program by cross-linking with Fas-ligand (FasL). Consequently, tumors that express FasL can induce apoptosis of infiltrating Fas-positive T lymphocytes and subdue any antitumor host immune response. Since Epstein-Barr virus (EBV)-associated tumors such as Hodgkin lymphoma (HL) and nasopharyngeal carcinoma (NPC) express FasL, we determined whether EBV-specific cytotoxic T lymphocytes (EBV-CTLs) could be modified to resist this evasion strategy. We show that long-term down-modulation of Fas can be achieved in EBV-CTLs by transduction with small interfering RNA (siRNA) encoded in a retrovirus. Modified T cells resisted Fas/FasL-mediated apoptosis compared with control cells and showed minimal cleavage of the caspase3 substrate poly(ADP-ribose) polymerase (PARP) protein after Fas engagement. Prolonged Fas stimulation selected a uniformly Fas(low) and FasL resistant population. Removal of responsiveness to this single death signal had no other discernible effects on EBV-CTLs. In particular, it did not lead to their autonomous growth since the modified EBV-CTLs remained polyclonal, and their survival and proliferation retained dependence on antigen-specific stimulation and on the presence of other physiologic growth signals. EBV-CTLs with knocked down Fas should have a selective functional and survival advantage over unmodified EBV-CTLs in the presence of tumors expressing FasL and may be of value for adoptive cellular therapy.     International audienceTouch screens rapidly and significantly replace physical keyboards on mobile devices. Hence, text entry is now dependent on software (or virtual) keyboards that are widely used by sighted people, but raise accessibility issues for visually impaired users. These users rely on tactile exploration with vocal feedback of the whole screen for entering text, which is time consuming. We designed a software keyboard that aims reducing tactile exploration and speeding up text entry for VI users. It relies on the selection of the first letter of a word and rapid and inaccurate typing of the remaining letters. It then proposes a list of words having the same first letter and a similar total distance between letters. The evaluation with twelve VI users showed that this keyboard is very efficient for words larger than five characters. It also helps preventing certain typing errors.L'utilisation des Ă©crans tactiles et en particulier les claviers logiciels est extrĂȘmement compliquĂ©e pour les non-voyants qui manquent de repĂšres physiques sur ce type d'appareil.     fMRI visualization of transient activations in the rat olfactory bulb using short odor stimulations

    International audienceOdor-evoked activity in the olfactory bulb displays both spatial and temporal organization. The difficulty when assessing spatio-temporal dynamics of olfactory representation is to find a method that reconciles the appropriate resolution for both dimensions. Imaging methods based on optical recordings can reach high temporal and spatial resolution but are limited to the observation of the accessible dorsal surface. Functional magnetic resonance imaging (fMRI) may be useful to overcome this limitation as it allows recording from the whole brain. In this study, we combined ultra fast imaging sequence and short stimulus duration to improve temporal resolution of odor-evoked BOLD responses. Short odor stimulations evoked high amplitude BOLD responses and patterns of activation were similar to those obtained in previous studies using longer stimulations. Moreover, short odor exposures prevented habituation processes. Analysis of the BOLD signal time course in the different areas of activation revealed that odorant response maps are not static entities but rather are temporally dynamic as reported by recent studies using optical imaging. These data demonstrated that fMRI is a non-invasive method which could represent a powerful tool to study not only the spatial dimension of odor representation but also the temporal dimension of information processing

    L23VV Auger transition has been studied in Si, SiO2, Al, AlN, Al2O3 by electron spectroscopy excited either by electron beam or X Rays. A strong difference is observed in intensity between pure solid and oxide or nitride under electron bombardment. Auger intensity is very sensitive to changes in the backscattering coefficient or inelastic mean free path. However transient local trapping of electrons seems to be responsible for the large change observed

    The set up of an ion gun, producing a focused beam in the analysis chamber of a Scanning Auger Microscope permits ion scattering experiments: surface studies performed by electron spectroscopies can then be enlarged by Ion Scattering Spectroscopy (I.S.S.) to get additional information. I.S.S. appears to be very sensitive to the cleanliness of the surface: comparison between Electron Energy Loss Spectroscopy (E.E.L.S.) and I.S.S. studies on clean samples show that I.S.S. can still detect oxygen even when it is not detectable by E.E.L.S. Preliminary results on oxidation of Al (111) and Al (100) give oxidation curves in good agreement with those obtained by Auger Electron Spectroscopy (A.E.S.) and X Ray Photoemission Spectroscopy (X.P.S.)

    Trend correlations for coastal eutrophication and its main local and whole-sea drivers – Application to the Baltic Sea

    Coastal eutrophication is a major environmental issue worldwide. In the Baltic Sea, eutrophication affects both the coastal waters and the open sea. Various policy frameworks aim to hinder its progress but eutrophicationrelevant water quality variables, such as chlorophyll-a concentrations, still exhibit opposite temporal trends in various Baltic Sea marine and coastal waters. In this study, we investigate the temporal-trend linkages of measured water quality variables and their various anthropogenic, climatic and hydrospheric drivers over the period 1990-2020 with focus on the Swedish coastal waters and related marine basins in the Baltic Sea. We find that it is necessary to distinguish more and less isolated coastal waters, based on their water exchanges with the open sea, to capture different coastal eutrophication dynamics. In less isolated coastal waters, eutrophication is primarily related to nitrogen concentrations, while it is more related to phosphorus concentrations in more isolated coastal waters. In the open sea, trends in eutrophication conditions correlate best with trends in climatic and hydrospheric drivers, like wind speed and water salinity, respectively. In the coastal waters, driver signals are more mixed, with considerable influences from anthropogenic land-based nutrient loads and sea ice cover duration. Summer chlorophyll-a concentration in the open sea stands out as a main change driver of summer chlorophyll-a concentration in less isolated coastal waters. Overall, coastal waters are a melting pot of driver influences over various scales, from local land-based drivers to large-scale total catchment and open sea conditions. The latter in turn depend on long-term integration of pathway-dependent influences from the various coastal parts of the Baltic Sea and their land-based nutrient load drivers, combined with overarching climate conditions and internal feedback loops. As such, our results challenge any unidirectional local source-to-sea paradigm and emphasize a need for concerted local land-catchment and whole-sea measures for robust coastal eutrophication management.     AbstractThe coronavirus spike glycoprotein is a class I membrane fusion protein with two characteristic heptad repeat regions (HR1 and HR2) in its ectodomain. Here, we report the X-ray structure of a previously characterized HR1/HR2 complex of the severe acute respiratory syndrome coronavirus spike protein. As expected, the HR1 and HR2 segments are organized in antiparallel orientations within a rod-like molecule. The HR1 helices form an exceptionally long (120 Å) internal coiled coil stabilized by hydrophobic and polar interactions. A striking arrangement of conserved asparagine and glutamine residues of HR1 propagates from two central chloride ions, providing hydrogen-bonding “zippers” that strongly constrain the path of the HR2 main chain, forcing it to adopt an extended conformation at either end of a short HR2 α-helix

    Differential expression of synaptophysin and synaptoporin during pre- and postnatal development of the hippocampal network

    The closely related synaptic vesicle membrane proteins synaptophysin and synaptoporin are abundant in the hippocampal formation of the adult rat. But the prenatal hippocampal formation contains only synaptophysin, which is first detected at embryonic day 17 (E17) in perikarya and axons of the pyramidal neurons. At E21 synaptophysin immunoreactivity extends into the apical dendrites of these cells and in newly formed terminals contacting these dendrites. The transient presence of synaptophysin in axons and dendrites suggests a functional involvement of synaptophysin in fibre outgrowth of developing pyramidal neurons. Synaptoporin expression parallels the formation of dentate granule cell synaptic contacts with pyramidal neurons: the amount of hippocampal synaptoporin, determined in immunoblots and by synaptoporin immunostaining of developing mossy fibre terminals, increases during the first postnatal week. Moreover, in the adult, synaptoporin is found exclusively in the mossy fibre terminals present in the hilar region of the dentate gyrus and the regio inferior of the cornu ammonis. In contrast, synaptophysin is present in all synaptic fields of the hippocampal formation, including the mossy fibre terminals, where it colocalizes with synaptoporin in the same boutons. Our data indicate that granule neuron terminals differ from all other terminals of the hippocampal formation by the presence of both synaptoporin and synaptophysin. This difference, observed in the earliest synaptic contacts in the postnatal hippocampus and persisting into adult life, suggests distinct functions of synaptoporin in these nerve terminals
