    Linking dietary intake, circadian biomarkers, and clock genes on obesity: A study protocol

    BackgroundThe prevalence of obesity continues to rise, and although this is a complex disease, the screening is made simply with the value of the Body Mass Index. This index only considers weight and height, being limited in portraying the multiple existing obesity phenotypes. The characterization of the chronotype and circadian system as an innovative phenotype of a patient's form of obesity is gaining increasing importance for the development of novel and pinpointed nutritional interventions. ObjectiveThe present study is a prospective observational controlled study conducted in Portugal, aiming to characterize the chronotype and determine its relation to the phenotype and dietary patterns of patients with obesity and healthy participants. MethodsAdults with obesity (study group) and healthy adults (control group), aged between 18 and 75, will be enrolled in this study. Data will be collected to characterize the chronotype, dietary intake, and sleep quality through validated questionnaires. Body composition will also be assessed, and blood samples will be collected to quantify circadian and metabolic biomarkers. DiscussionThis study is expected to contribute to a better understanding of the impact of obesity and dietary intake on circadian biomarkers and, therefore, increase scientific evidence to help future therapeutic interventions based on chronobiology, with a particular focus on nutritional interventions

    Development and validation of predictive equations for spirometry in Portuguese children

    Background There are no data on lung function reference values for Portuguese children, and the contribution from the Portuguese data set in the Global Lung Function Initiative (GLI) is scant. Objectives We aimed to estimate new up-to-date reference values for Portuguese children by fitting a multivariable regression model to a general population sample. Further, we intended to assess the external validity of the obtained reference values and to compare them to the GLI reference values. Methods A random sample of 858 children from 20 primary schools were screened by health questionnaire, physical examination, and spirometry. Spirometric parameters recorded were FVC, FEV1, and FEF25-75. Multiple regression models were used to derive reference equations. Results Overall, 481 children, aged between 7 and 12 years, 267 boys (55.5%), were included. Boys had higher values for FVC and FEV1 than girls (P < .05). The strongest correlation was found for FVC with height (r = 0.71 for boys and 0.70 for girls), while the lowest correlation was observed in both sexes for FEF25-75 with age (r = 0.23). Height was the most significant predictor of FVC, FEV1, and FEF25-75 in our models. Weight and body mass index were not significant predictors for boys but had a significant effect on girls’ equations for all spirometry parameters. Compared to obtained reference equations with GLI, they performed better for FVC in boys, FEV1 in girls, and FEF25-75 in both boys and girls.     Quantitative studies on the enzymatic hydrolysis of milk proteins brought about by cardosins precipitated by ammonium sulfate

    Hydrolysis of whey proteins may produce peptide mixtures with better functional properties than the original protein mixture, viz. higher solubilites and lower allergenic effects. Cynara cardunculus is a wild plant that possesses (aspartic) proteases in its flower cells; those enzymes exhibit general proteolytic and specific milk clotting activities, which are rather useful in traditional cheesemaking. This study was thus aimed at characterizing the enzymatic action of crude extracts of said plant after preliminary purification by salting out with ammonium sulfate at two different concentration levels, viz. 30% and 70% saturation. The coagulant activity on milk, and the proteolytic activity using casein and azocasein as substrates, of the crude extract and of each precipitated fraction were measured at 37°C and pH 5.2. The profile of hydrolysis of the major whey proteins, i.e. α-lactalbumin (α-La), β-lactoglobulin (β-Lg) and bovine serum albumin (BSA) was characterized by gel permeation chromatography and polyacrylamide gel electrophoresis (PAGE) in the presence of sodium dodecyl sulfate. The 30% and 70% saturation fractions exhibited lower coagulant and proteolytic specific activities than the crude extract. However, the relative ratio of coagulant to proteolytic activity, which is a useful indicator of appropriateness for cheesemaking, was higher for the partially purified fractions. The extents of hydrolysis of whey proteins brought about by the partially purified extracts were above those by their crude counterpart, but qualitative hydrolysis patterns were essentially identical to each other; by 24 h, α-La was substantially depleted, whereas β-Lg was very poorly hydrolyzed and BSA was only slightly hydrolyzed.     Chondrosia reniformis marine-sponge collagen membranes for skin re-epithelialization

    Chondrosia reniformis collagen has been identified as mainly of type IV. Being collagen IV the main component of the epidermal basal layer [1], C. reniformis represents a valuable source to be explored in the skin regeneration field. This work envisaged the production of C. reniformis collagen membranes for the selection of rapidly adherent epidermal cells, like the commercial collagen coatings, and for their subsequent culture. This approach would permit a single system for culturing and carrying basal epidermal cells aimed at re-epithelialize skin wounds. Materials and Methods The collagen of C. reniformis marine-sponge was extracted with 100mM Tris-HCl, 10mM EDTA, 8M urea and 100mM 2-mercaptoethanol. To define the best re-solubilization conditions, the obtained precipitate was dissolved in five different solutions: Solution A: 100mM Tris-HCl+8M Urea+10mM EDTA (pH 9.5); Solution B: 50mM Tris-HCl+1M NaCl (pH 7.4); Solution C: 100mM Tris-HCl (pH 7.4); Solution D: 0.5% H2O2 (v/v) (pH 11) and Solution E: 100mM Tris-HCl (pH 9.5). Solutions of 1% collagen were prepared and cross-linking was performed with HMDI, genipin and EDC/NHS at different concentrations. The membranes were obtained by solvent-casting and/or freeze-drying, and their stability was tested both in PBS and culture medium, for at least 7 days. Morphological characterization of the membranes was carried out by scanning electron microscopy (SEM). Cytotoxicity, based on metabolic activity (MTS assay) and cell proliferation (DNA quantification) analysis of the 100mM Tris-HCl (pH 9.5) and 8mM EDC/NHS cross-linked collagen membranes, was assessed with L929 cells.     Taekwondo (TKD) is a combat sport organized by two main federations: International TKD (ITF) and the World TKD (WTF). The present study analyzed the kinematic differences in roundhouse kick performed by ITF and WTF athletes. Seventeen athletes participated in the study (8 ITF; 9 WTF). DVideo system was used to reconstruct the 3D coordinates of sixteen markers. Pelvis and hips angles and angular velocities were used to compare the groups. The results highlight two kick strategies to hit the target. WTF athletes performed the kick with increased pelvis left rotation while ITF used increased pelvis anteversion, hip flexion and abduction on supporting leg. These differences should be considered for adaptation of rules and training
