Multiplex PCR for the detection of sea, seb, sec, sed and see genes of Staphylococcus aureus: Characterization of isolates from food
The presence of Staphylococcus aureus in food represents a potential risk to public health, being its enterotoxins the major virulence factor. Enterotoxin detection can be determined by ELISA, but only for the pool of enterotoxins SEA, SEB, SEC, SED and SEE. The main aims of this study were to optimize two PCR techniques for detection of S. aureus sea, seb, sec, sed and see, and to characterize Staphylococcus spp. isolates associated with food intoxication. Two PCR techniques were optimized and 115 Staphylococcus spp. isolates from Ciudad Autónoma de Buenos Aires, and Buenos Aires, Córdoba, and Neuquén provinces were characterized. The characterization was performed by biochemical tests, ELISA and PCR. Sixty-eight isolates (59.1%) were positive by ELISA, while 61 (53%) were positive by PCR. Out of the positive PCR isolates, 34 (55.7%) carried the sea gene, 9 (14.8%) the seb gene, 5 (8.1%) the see gene, 4 (6.5%) the sec gene, 6 (9.9%) were positive for sea and seb genes, 2 (3.3%) for sea and sec genes, and 1 (1.7%) for sea and sed genes. This is the first study of genotypic characterization of S. aureus isolates associated with food intoxication from different provinces of Argentina.Fil: Manfredi, E.A.. From Component-Based Architectures to Microservices: A 25-years-long Journey in Designing and Realizing Service-Based Systems
Distributed information systems and applications are generally described in terms of components and interfaces among them. How these component-based architectures have been designed and implemented evolved over the years, giving rise to the so-called paradigm of Service-Oriented Computing (SOC). In this chapter, we will follow a 25-years-long journey on how design methodologies and supporting technologies influenced one each other, and we discuss how already back in the late 90s the ancestors of the SOC paradigm were there, already paving the way for the technological evolution recently leading to microservice architectures and serverless computing
On the Deployment of IoT Systems: An Industrial Survey
Internet of Things (IoT) systems are complex and multifaceted, and the design of their architectures needs to consider many aspects at a time. Design decisions concern, for instance, the modeling of software components and their interconnections, as well as where to deploy the components within the available hardware infrastructure in the Edge-Cloud continuum. A relevant and challenging task, in this context, is to identify optimal deployment models due to all the different aspects involved, such as extra-functional requirements of the system, heterogeneity of the hardware resources concerning their processing and storage capabilities, and constraints like legal issues and operational cost limits. To gain insights about the deployment decisions concerning IoT systems in practice, and the factors that influence those decisions, we report about an industrial survey we conducted with 66 IoT architects from 18 countries across the world. Each participant filled in a questionnaire that comprises 15 questions. By analyzing the collected data, we have two main findings: (i) architects rely on the Cloud more than the Edge for deploying the software components of IoT systems, in the majority of the IoT application domains; and (ii) the main factors driving deployment decisions are four: reliability, performance, security, and cost
Enhancing Web Applications Observability through Instrumented Automated Browsers
In software engineering, observability is the ability to determine the current state of a software system based on its external outputs or signals such as metrics, logs, or traces. Web engineers rely on the web browser console as the primary tool to monitor the client-side of web applications during end-to-end tests. However, this is a manual and time-consuming task due to the different browsers available. This paper presents BrowserWatcher, an open-source browser extension providing cross-browser capabilities to observe web applications and automatically gather browser console logs in different browsers (e.g., Chrome, Firefox, or Edge). We have leveraged this extension to conduct an empirical study analyzing the browser console of the top-50 public websites manually and automatically. The results show that BrowserWatcher gathers all the well-known log categories such as console or error traces. It also reveals that each web browser additionally includes other types of logs, which differ among browsers, thus providing distinct pieces of information for the same website. This work was partially supported in part by the Ministerio de Ciencia e Innovación-Agencia Estatal de Investigación, Spain (10.13039/501100011033) through the H2O Learn project under Grant PID2020-112584RB-C31, in part by the Madrid Regional Government through the e-Madrid-CM Project, Spain under Grant S2018/TCS-4307, and in part supported by the Comunidad de Madrid and Universidad Politécnica de Madrid, Spain through the V-PRICIT Research Programme Apoyo a la realización de Proyectos de I+D para jóvenes investigadores UPM-CAM, under Grant APOYOJOVENES-QINIM8-72-PKGQ0J. Resistin, visfatin, leptin and omentin are differently related to hormonal and metabolic parameters in growth hormone-deficient children
PURPOSE:
The effect of growth hormone (GH) on adipose tissue and the role of adipokines in modulating metabolism are documented, but with discordant data. Our aim was to evaluate the impact of GH treatment on a series of selected adipokines known to have a metabolic role and poorly investigated in this setting.
METHODS:
This is a prospective study. Thirty-one prepubertal children (25 M, 6 F; aged 8.5 ± 1.6 years) with isolated GH deficiency treated with GH for at least 12 months and 30 matched controls were evaluated. Auxological and metabolic parameters, insulin sensitivity indexes, leptin, soluble leptin receptor, adiponectin, visfatin, resistin, omentin, adipocyte fatty acid-binding protein and retinol-binding protein-4 were evaluated before and after 12 months of treatment.
RESULTS:
At baseline, no significant difference in metabolic parameters was found between GHD children and controls, except for higher LDL cholesterol (p = 0.004) in the first group. At multivariate analysis, LDL cholesterol was independently associated with resistin (B 0.531; p = 0.002), while IGF-I was the only variable independently associated with visfatin (B 0.688; p < 0.001). After 12 months, a significant increase in fasting insulin (p = 0.008), Homa-IR (p = 0.007) and visfatin (p < 0.001) was found, with a concomitant decrease in LDL cholesterol (p = 0.015), QUICKI (p = 0.001), ISI Matsuda (p = 0.006), leptin (p = 0.015) and omentin (p = 0.003)]. At multivariate analysis, BMI was the only variable independently associated with leptin (B 0.485; p = 0.040).
CONCLUSIONS:
GH treatment modifies adipokine secretion and the perturbation of some adipokine levels could contribute to the clinical and metabolic changes observed during the follow-up
Genotypic Characterization of Non-O157 Shiga Toxin–Producing Escherichia coli in Beef Abattoirs of Argentina
The non-O157 Shiga toxin-producing Escherichia coli (STEC) contamination in carcasses and feces of 811 bovines in nine beef abattoirs from Argentina was analyzed during a period of 17 months. The feces of 181 (22.3%) bovines were positive for non-O157 STEC, while 73 (9.0%) of the carcasses showed non-O157 STEC contamination. Non-O157 STEC strains isolated from feces (227) and carcasses (80) were characterized. The main serotypes identified were O178:H19, O8:H19, O130:H11, and O113:H21, all of which have produced sporadic cases of hemolytic-uremic syndrome in Argentina and worldwide. Twenty-two (7.2%) strains carried a fully virulent stx/eae/ehxA genotype. Among them, strains of serotypes O103:[H2], O145:NM, and O111:NM represented 4.8% of the isolates. XbaI pulsed-field gel electrophoresis pattern analysis showed 234 different patterns, with 76 strains grouped in 30 clusters. Nine of the clusters grouped strains isolated from feces and from carcasses of the same or different bovines in a lot, while three clusters were comprised of strains distributed in more than one abattoir. Patterns AREXSX01.0157, AREXBX01.0015, and AREXPX01.0013 were identified as 100% compatible with the patterns of one strain isolated from a hemolytic-uremic syndrome case and two strains previously isolated from beef medallions, included in the Argentine PulseNet Database. In this survey, 4.8% (39 of 811) of the bovine carcasses appeared to be contaminated with non- O157 STEC strains potentially capable of producing sporadic human disease, and a lower proportion (0.25%) with strains able to produce outbreaks of severe disease.Fil: Masana, Marcelo. A Survey on the Application of Process Mining to Smart Spaces Data
During the last years, a number of studies have experimented with applying process mining (PM) techniques to smart spaces data. The general goal has been to automatically model human routines as if they were business processes. However, applying process-oriented techniques to smart spaces data comes with its own set of challenges. This paper surveys existing approaches that apply PM to smart spaces and analyses how they deal with the following challenges identified in the literature: choosing a modelling formalism for human behaviour; bridging the abstraction gap between sensor and event logs; and segmenting logs in traces. The added value of this article lies in providing the research community with a common ground for some important challenges that exist in this field and their respective solutions, and to assist further research efforts by outlining opportunities for future work
