Mesenteric ischemia/reperfusion (I/R) is associated with high rates of morbidity and mortality. We studied the effect of mesenteric I/R on structural and mechanical properties of rat mesenteric resistance artery (MRA) that, once disrupted, might impact the outcome of this devastating clinical condition. Superior mesenteric artery from Wistar–Kyoto rats was occluded (90 min) and reperfused (24 h). The effect of tezosentan, a dual endothelin (ET)-receptor antagonist, was studied in ischemic (IO) and sham-operated (SO) animals. MRA structure and mechanics were assessed by pressure myography. Nuclei distribution, elastin content and organization, collagen I/III and ET-1 expression, ET-1 plasma levels, superoxide anion (O2⋅−) production, and mRNA levels of NAD(P)H-oxidase subunits were measured. To assess ET-1 effects on O2⋅− production, MRA from non-operated rats were incubated in culture medium with ET-1. Mesenteric I/R increased MRA wall thickness (P < 0.05) and cross-sectional area (P < 0.05) but decreased wall stiffness (P < 0.05). Arterial remodeling was paralleled by enhancement of: (i) collagen I/III expression (P < 0.01), ET-1 expression (P < 0.05), and O2⋅− formation (P < 0.01) in the vessel wall; (ii) number of internal elastic lamina (IEL) fenestrae (P < 0.05); and (iii) plasma levels of ET-1 (P < 0.05). Moreover, ET-1 increased O2⋅− (P < 0.05) production in cultured MRA. Tezosentan prevented hypertrophic remodeling and collagen I/III deposition, and enhanced O2⋅− production, but it did not affect the decreased wall stiffness after mesenteric I/R. These results indicate that 90 min occlusion/24 h reperfusion induces hypertrophic remodeling of MRA linked to ET-1-mediated increase of collagen and O2⋅−. Decreased stiffness may be associated with increased number of IEL fenestrae. The resulting MRA remodeling, initially adaptive, might become maladaptive contributing to the pathology and poor outcome of mesenteric I/R, and might be a valuable treatment target for mesenteric I/R

Numerical and experimental evaluation of dielectric properties of thermally aged insulating paper used in power transformers

Due to the relevant role of the cellulosic insulation in the dielectric system of a power transformer, it is necessary to know its dielectric properties and its evolution as insulating material. The dependence of the dielectric properties of solid insulation on the moisture content, temperature, or pressure is well-defined. However, it is not clear the effect of the paper degradation on its insulating capacity. To study the impact of ageing on the dielectric properties, samples of Kraft paper were subjected to an accelerated hygrothermal ageing. Degree of polymerization (DP) value was reduced from 1000 to 200, with intermediate study points. All samples were conditioned to the same moisture content in order to only evaluate the effect of the paper degradation. Different dielectric properties, such as loss factor (tanδ) and complex permittivity (ε) were measured using the dielectric spectroscopy technique. These experimental results were used to develop a numerical model with the finite element-based tool Comsol Multiphysics. This model provided some extra information about the material properties, such as the electric field distribution. Results showed that the degradation of Kraft paper modifies its dielectric response. We acknowledge the support of the Spanish Ministry of Science and Innovation by means of the National Research Project Asset management of biodegradable-fluid-based transformers (PID2019-107126RB-C22/AEI/ 10.13039/501100011033). ObjectiveThis study aimed to monitoring the prevalence of previously identified thyroid disorders and hypothyroidism monitoring before pregnancy.Material and methodsA retrospective cross-sectional study of women whose pregnancies occurred between 2014 and 2016 was conducted, including 120,763 pregnancies in Catalonia (Spain). The presence of thyroid disorders in women was based on disease diagnostic codes and/or prescription of levothyroxine or antithyroid drugs. To evaluate the thyroid disorder diagnosis and monitoring, thyrotropin (TSH), free T4 (FT4), antiperoxidase antibody (TPOAb), and anti-TSH receptor antibody (TRAb) records were gathered and categorised according to the reference values of each laboratory.ResultsThe prevalence of recorded thyroid disorders before the last menstrual period was 5.09% for hypothyroidism and 0.64% for hyperthyroidism, showing a significant increase with age. A thyroid monitoring test was not performed in the year before the last menstrual period in approximately 40% of women with a known thyroid disorder. Amongst the women with hypothyroidism who underwent a TSH test, 31.75% showed an above-normal result. Amongst women previously unknown to have thyroid disorders, 3.12% had elevated TSH levels and 0.73% had low TSH levels.ConclusionA high percentage of Catalan women with a known thyroid disorder were not properly monitored during the year before pregnancy. Amongst those monitored, more than one-third had TSH values outside the reference range. Therefore, it is important to evaluate women with thyroid disorders during pre-pregnancy visits

Seven strains of equine herpesvirus 1 (EHV-1) isolated in Argentina from 1979 to 1996 and reference Japanese strain, were compared by restriction fragments analysis (RFA), polyacrylamide gel electrophoresis (SDS-PAGE) and inoculation of baby mice. The RFA revealed that all EHV-1 strains belonged to genome type 1P. Only the first isolate (SP strain) showed differences. This strain and the LP strain isolated in 1985 presented a small differences for SDS-PAGE analysis. Baby mice inoculation revealed that three biotypes could be distinguished on the basis of pathogenicity. However, no strict correlation with pathogenicity in the natural host was seen

Combating virulence of Gramnegative bacilli by OmpA inhibition

Preventing the adhesion of pathogens to host cells provides an innovative approach to tackling multidrug-resistant bacteria. In this regard, the identifcation of outer membrane protein A (OmpA) as a key bacterial virulence factor has been a major breakthrough. The use of virtual screening helped us to identify a cyclic hexapeptide AOA-2 that inhibits the adhesion of Acinetobacter baumannii, Pseudomonas aeruginosa and Escherichia coli to host cells and the formation of bioflm, thereby preventing the development of infection in vitro and in a murine sepsis peritoneal model. Inhibition of OmpA ofers a strategy as monotherapy to address the urgent need for treatments for infections caused by Gram-negative bacilli.Instituto de Salud Carlos III, Ministerio de Economía y Competitividad REIPI RD12/0015/0001Unión Europea ES P201431400Ministerio de Economía y Competitividad CP15/01358Junta de Andalucía CTS-6173/12 y 2014LLAV-00064Junta de Andalucía PI12–006

Multidimensional performance of periurban horticulture:Assessing agroecological transition and soil health

Get PDF
This study aims to produce evidence on the performance of horticultural systems in the peri-urban area of Buenos Aires (Argentina), based on the 10 Elements of Agroecology with an emphasis on soil health. To this end, we performed a survey with the Tool for Agroecology Performance Evaluation (TAPE) Step 1, and we measured soil physical, chemical, and biological properties in family farms self-defined as agroecological or conventional. Results reflected overall low transition-to-agroecology indices and poor soil quality in the studied area. However, self-defined agroecological systems have made significant strides toward agroecological transition with higher scores mainly affecting elements describing management and innovation aspects of the agroecological transition but also those describing the social aspects and the enabling environment. Finally, self-defined agroecological farms exhibit better soil health scores for the indicators aggregates stability, soil organic carbon, and soil basal respiration, than conventional farms. Nevertheless, there is still potential to be exploited and constraints to be overcome to improve the transition to agroecology of horticultural systems in this area.</p
