    Verbal Component of Advertisement and the Problem of its Perception

    The article touches upon the problem of impact the proper use of the language of advertising can have on people. Since today, advertisement has a significant impact on consumer choice of a customer. In the first part of this article we analyze such theoretical issues as stylistics of advertising text, its phonetic, syntactic and lexical aspects, means of language manipulation and readability index. Comparative analysis of linguistic peculiarities of the advertisement on the example of advertising texts of famous sport brands is shown in the second part. The main conclusion is that in spite of the fact that readability index of the text can be rather high, the level of its attractiveness to the public also depends on other factors that include non-verbal component of the message, personal characteristics of addressee, etc. The main idea of this article can be valuable for the world of psycholinguistics, lexicology and modern communication because it describes all the important linguistic peculiarities that can be used in the modern language of advertising

    El estudio tiene como objetivo desarrollar medidas para reducir la salida (migración) de jóvenes y mejorar el capital humano ante el complejo panorama socioeconómico global. El documento determina las prioridades estratégicas para la regulación estatal de los procesos migratorios de jóvenes. Se concluye que los instrumentos para regular la migración de jóvenes y preservar el potencial intelectual y de recursos humanos deben dirigirse a reducir el nivel de aspiraciones migratorias positivas entre los jóvenes estudiantes y aprendices, asegurando el empleo y la remuneración efectiva del trabajo. El sistema de instrumentos de política pública preventiva para gestionar la migración de jóvenes incluye esfuerzos para aumentar el empleo, desarrollar el sector de las micro y pequeñas empresas, elevar el nivel de preparación de los jóvenes migrantes para reemigrar, mejorar la competitividad del sector educativo y crear proyectos internacionales para desarrollar el capital humano. Además, el estudio aboga por mejorar la competitividad del sector educativo y fomentar proyectos internacionales que contribuyan al desarrollo del capital humano

    The Black Sea Region Energy Cooperation: Current Trends and Prospects

    The Black Sea region is one of the most complex regions in terms of energy development. It hosts several major powers and some developing countries that need to cut energy costs. In general, the region is controversial. It is influenced by external actors, and therefore regional stability is very difficult to achieve. In addition, institutional players such as the EU, the Belt and Road Initiative, the Black Sea Trade and Development Bank, etc., have their own vision of the future of the Black Sea region. The article is aimed at assessing the regional balance of power and estimating the interests of the countries of the region. Based on this assessment, the authors have classified the countries in the region, predicted potential alliances, and provided recommendations on how the countries should behave in the region. The key findings comprise the rejection of the two hypotheses: the countries of the region cooperate mainly through similar institutions; and the countries of the region can efficiently cooperate within the framework of a single strategy.     Formate overflow drives toxic folate trapping in MTHFD1 inhibited cancer cells

    Cancer cells fuel their increased need for nucleotide supply by upregulating one-carbon (1C) metabolism, including the enzymes methylenetetrahydrofolate dehydrogenase-cyclohydrolase 1 and 2 (MTHFD1 and MTHFD2). TH9619 is a potent inhibitor of dehydrogenase and cyclohydrolase activities in both MTHFD1 and MTHFD2, and selectively kills cancer cells. Here, we reveal that, in cells, TH9619 targets nuclear MTHFD2 but does not inhibit mitochondrial MTHFD2. Hence, overflow of formate from mitochondria continues in the presence of TH9619. TH9619 inhibits the activity of MTHFD1 occurring downstream of mitochondrial formate release, leading to the accumulation of 10-formyl-tetrahydrofolate, which we term a 'folate trap'. This results in thymidylate depletion and death of MTHFD2-expressing cancer cells. This previously uncharacterized folate trapping mechanism is exacerbated by physiological hypoxanthine levels that block the de novo purine synthesis pathway, and additionally prevent 10-formyl-tetrahydrofolate consumption for purine synthesis. The folate trapping mechanism described here for TH9619 differs from other MTHFD1/2 inhibitors and antifolates. Thus, our findings uncover an approach to attack cancer and reveal a regulatory mechanism in 1C metabolism.In this study, Green, Marttila, Kiweler et al. characterize one-carbon metabolism rewiring in response to a dual MTHFD1 and MTHFD2 inhibitor.     Small molecule inhibitor of OGG1 blocks oxidative DNA damage repair at telomeres and potentiates methotrexate anticancer effects

    The most common oxidative DNA lesion is 8-oxoguanine which is mainly recognized and excised by the 8-oxoG DNA glycosylase 1 (OGG1), initiating the base excision repair (BER) pathway. Telomeres are particularly sensitive to oxidative stress (OS) which disrupts telomere homeostasis triggering genome instability. In the present study, we have investigated the effects of inactivating BER in OS conditions, by using a specific inhibitor of OGG1 (TH5487). We have found that in OS conditions, TH5487 blocks BER initiation at telomeres causing an accumulation of oxidized bases, that is correlated with telomere losses, micronuclei formation and mild proliferation defects. Moreover, the antimetabolite methotrexate synergizes with TH5487 through induction of intracellular reactive oxygen species (ROS) formation, which potentiates TH5487-mediated telomere and genome instability. Our findings demonstrate that OGG1 is required to protect telomeres from OS and present OGG1 inhibitors as a tool to induce oxidative DNA damage at telomeres, with the potential for developing new combination therapies for cancer treatment

    Pathogen Entrapment by Transglutaminase—A Conserved Early Innate Immune Mechanism

    Clotting systems are required in almost all animals to prevent loss of body fluids after injury. Here, we show that despite the risks associated with its systemic activation, clotting is a hitherto little appreciated branch of the immune system. We compared clotting of human blood and insect hemolymph to study the best-conserved component of clotting systems, namely the Drosophila enzyme transglutaminase and its vertebrate homologue Factor XIIIa. Using labelled artificial substrates we observe that transglutaminase activity from both Drosophila hemolymph and human blood accumulates on microbial surfaces, leading to their sequestration into the clot. Using both a human and a natural insect pathogen we provide functional proof for an immune function for transglutaminase (TG). Drosophila larvae with reduced TG levels show increased mortality after septic injury. The same larvae are also more susceptible to a natural infection involving entomopathogenic nematodes and their symbiotic bacteria while neither phagocytosis, phenoloxidase or—as previously shown—the Toll or imd pathway contribute to immunity. These results firmly establish the hemolymph/blood clot as an important effector of early innate immunity, which helps to prevent septic infections. These findings will help to guide further strategies to reduce the damaging effects of clotting and enhance its beneficial contribution to immune reactions

    Targeting OGG1 arrests cancer cell proliferation by inducing replication stress

    Altered oncogene expression in cancer cells causes loss of redox homeostasis resulting in oxidative DNA damage, e.g. 8-oxoguanine (8-oxoG), repaired by base excision repair (BER). PARP1 coordinates BER and relies on the upstream 8-oxoguanine-DNA glycosylase (OGG1) to recognise and excise 8-oxoG. Here we hypothesize that OGG1 may represent an attractive target to exploit reactive oxygen species (ROS) elevation in cancer. Although OGG1 depletion is well tolerated in non-transformed cells, we report here that OGG1 depletion obstructs A3 T-cell lymphoblastic acute leukemia growth in vitro and in vivo, validating OGG1 as a potential anti-cancer target. In line with this hypothesis, we show that OGG1 inhibitors (OGG1i) target a wide range of cancer cells, with a favourable therapeutic index compared to non-transformed cells. Mechanistically, OGG1i and shRNA depletion cause S-phase DNA damage, replication stress and proliferation arrest or cell death, representing a novel mechanistic approach to target cancer. This study adds OGG1 to the list of BER factors, e.g. PARP1, as potential targets for cancer treatment
